    Notes: Abstract The KASMMER (Kakioka Automatic Standard Magnetometer) has been constructed in 1972 for the standard magnetic observation at Kakioka (Yanagiharaet al., 1973). Then KASMMER system has been improved successively for the high level standard magnetic observation. The purpose of the present paper is to introduce an outline of the improved KASMMER system. The KASMMER system consists of four parts which are four optical pumping magnetometers, a fluxgate magnetometer (Supporting system to the optical pumping magnetometers), a calibration system and a data acquisition system (computer system). The calibration system consists of a proton magnetometer and a magnetic theodolite (Di-72). Details for each part are described and the ability of the improved system is shown to be higher than that of classical magnetic observatory systems.

    Notes: Summary The organization of noradrenergic fibers in the lumbosacral anterior column of rats and dogs was examined in detail using a modification of a highly sensitive glyoxylic acid fluorescence histochemical method. In both rat and dog, there were greater concentrations of fluorescing noradrenergic fibers around the motoneurons innervating the perineal striated muscles (Onuf's nucleus) than around other motoneuronal groups. The preferential accumulation of noradrenergic fibers in Onuf's nucleus may indicate that the noradrenergic neuron system in the spinal cord of rodents and carnivores is closely related to the functional peculiarities of the perineal striated muscles, including the external anal and urethral sphincter muscles.

    Notes: Summary The distribution and morphological aspects of the serotonin-containing neurons in the paraventricular organ of the carp, frog, turtle and chicken were studied by means of an immunoperoxidase technique using serotonin antiserum. In all species the serotonin-containing neurons were seen to have the appearance of the CSF-contacting neurons and to be distributed in the pars ependymalis and the pars hypendymalis of the organ. Particularly, in the frog, the serotonin-containing CSF-contacting neurons, mostly bipolar in shape, were also observed in the pars distalis. Their proximal processes protruded into the ventricular lumen through the ependymal layer with a globular- and triangular-shape. The distal processes projected ependymofugally to the pars distalis and formed a fine plexus in the neuropil of this part. The density of the serotonin fibers in the pars distalis was greater in the carp than in the other species.

    Notes: Summary Selective immunohistochemical identification of enterochromaffin (EC) cells in the human duodenum and colon has been carried out, using a highly specific rabbit antiserotonin serum and the unlabeled peroxidase-antiperoxidase complex (PAP) technique. The sensitivity of the reaction, and morphology of the cells, were compared with those of the argentaffin or argyrophil reactions on the same sections at the light microscopic level. All argentaffin cells were argyrophil and PAP positive, but a small number of PAP-positive cells were neither argentaffin nor argyrophil. In addition, a few EC cells stained by the PAP method were shown to be argyrophil but not argentaffin. The PAP method permitted the clear morphological identification of the entire cell, particularly its cytoplasmic processes, whereas both the argentaffin and argyrophil reactions were basically seen in the basal regions of the cells and often failed to demonstrate the cytoplasmic processes. These findings indicate that the immunohistochemical method is more sensitive and advantageous than silver impregnations for the selective demonstration of EC cells.

    Notes: Summary Radioautographic studies of rat duodenal mucosa were carried out after single injections of 3H-thymidine, and the labelling indices of enterochromaffin (EC) cells identified by serotonin immunohistochemistry and by the diazo reaction were compared. The labelling index of EC cells by serotonin immunohistochemistry was 2.3% (mean percentage of 2 rats), while no labelled cells were found in diazo-positive EC cells. This finding indicates that serotonin immunohistochemistry reveals a less mature population of EC cells than conventional techniques such as the diazo reaction.

    Notes: Summary An intense thiaminepyrophosphatase (TPPase) activity was demonstrated in glial cells and blood vessels in the central nervous system (CNS), when incubation was carried out with thiaminepyrophosphate (TPP, cocarboxylase), using the method of Novikoff and Goldfischer (1961). Glial cells with TPPase activity were identified as microglia because they were morphologically similar to microglial cells in the sections stained with silver impregnation. TPPase activity was localized in the microglial perikaryon and in the processes, as viewed under a light microscope. Electron microscopically, enzyme activity was localized in the plasma membrane of microglia. We consider this activity to be a true TPPase activity hydrolyzing TPP, and we then went on to examine the substrate specificity, optimum pH, effect of chemical inhibitors and activators, and the effect of glutaraldehyde fixation. Our data are reported herein.

    Notes: Summary Distribution of the serotonin nerve fibers in the neurohypophysis of adult cats was demonstrated using the peroxidase-antiperoxidase method. The serotonin nerve fibers were distributed in the internal and external layers of the infundibulum, terminating on the wall of the capillary loops and the outer surface of the external layer. A small number of the fibers scattered about in the posterior lobe penetrated the intercellular spaces in the intermediate lobe.

    Notes: Summary The direct innervation of the pial blood vessels by serotonin neurons has been demonstrated with a modified peroxidase-antiperoxidase technique in the mammalian central nervous system. The pia mater covering the ventrolateral surface of the medulla oblongata is innervated by numerous varicose serotonin fibers originating from the serotonin neurons of the lower brainstem. Scattered serotonin fibers were observed in the pia mater in every part of the brain and spinal cord.

    Notes: Summary The distribution of serotonin (5-HT) positive fibers in the olfactory bulb of the rat, cat and monkey was studied using the peroxidase-anti-peroxidase (PAP) immunohistochemical method and highly specific antibodies to 5-HT. In general, 5-HT fibers were present throughout all layers in the olfactory bulb of these species except for the olfactory nerve layer and different as well as restricted laminar patterns of 5-HT distribution were observed. There were also species-related differences in the pattern of 5-HT distribution, in each layer. The most notable species difference was apparent in the glomerular layer of the main olfactory bulb. In case of the rat and cat, a very dense plexus of 5-HT fibers was observed to be diffuse in the glomerulus, while in the monkey, the distribution of 5-HT fibers was scanty and partial, as was seen in the accessory olfactory bulb of the rat.

    Notes: Summary By means of the peroxidase-antiperoxidase technique, in conjunction with a specific anti-CRF serum, it was shown that a large number of immunoreactive nerve fibers were demonstrable on the capillary loops of the hypophysial portal vessels in the external layer of the median eminence of the rat and monkey, particularly in its medial part. This result confirms the radioimmunological determination of CRF immunoreactivity in the median eminence.
