    Notes: Summary The course of regeneration of aminergic nerves in rat cerebral arteries was studied by means of histochemical methods, after uni- or bilateral cervical sympathectomy. Degeneration of aminergic nerves started on day 1 and was complete between days 3 and 7 after surgery. Between weeks 4 and 6, regenerating nerves started to appear from the proximal internal carotid artery. Regenerated aminergic nerve fibres were generally unbeaded and intensity of fluorescence was weak. The circular nerves appeared earlier than the longitudinal ones. The number of regenerating nerves reached the maximum, between months 9 and 12, at about half the normal level. AChE activity of the cerebral arteries showed no significant changes at any stage.

    Notes: Summary The localization of plasma retinol-binding protein (RBP) in the human kidney was determined by two immunocytochemical techniques, the PAP method and the protein A-gold technique. By using the affinity purified antibody against RBP obtained from the urine of the patients with cadmium poisoning (Itai-Itai disease), the immunoreactive substances were located by light microscopy in the proximal tubules of the human kidney. By immuno-electron microscopy, the stained organelles were identified as lysosomes in both S1 and S2 segments of the tubules. These data suggested that the reabsorption of low molecular weight plasma proteins like RBP can occur in the two segments. We inferred a similarity between the S1 and S2 segments concerning the reabsorption of RBP.

    Notes: Summary Postnatal development of autonomic nerves in the mouse iris was studied histochemically from one day to five months of age. For the demonstration of aminergic nerves the glyoxylic acid method was used, while for cholinergic nerves, Karnovsky and Roots' method was utilized on the whole-mount preparations of irises. The results obtained were as follows. In one-day-old mice, many aminergic nerves could already be seen, while cholinergic nerves were scarcely observable. Both types of nerve increased rapidly in the first 2 weeks. Both completed development between 3 and 4 weeks, although aminergic nerves were observed to develop earlier than the cholinergic.

    Notes: Summary 3H-befunolol was administered intravenously to pigmented rabbits. Thirty minutes after the administration, the iris, ciliary body and retina were fixed, embedded and processed for light microscopic radioautography. Radioautographical silver grains were observed over the pigment granules of the iris, ciliary body, choroid and retina. From these results it is concluded that 3H-befunolol is incorporated into the pigment granules of these cells. The mechanism of incorporation is discussed.

    Notes: Summary In order to clarify the underlying mechanism of malignant transformation from cirrhosis to hepatoma the cell kinetics of hepatocytes were studied in these two conditions. The content and synthesis of DNA in hepatocyte nuclei were investigated, by means of Feulgen-microspectrophotometry and tritiated thymidine radioautography, in cirrhotic and noncancerous parts of hepatoma with concomitant cirrhosis. The distribution of ploidy patterns was widely spread, from hypodiploid to hyperpolyploid, in the noncancerous parts of a cirrhotic liver containing hepatoma. In normal liver, each paired nuclear DNA content of a binucleate cell recorded almost the same amount, whereas in the noncancerous as well as in hepatoma cells much difference of DNA content was observed between the paired nuclei of the binucleate cells. The ploidy pattern of hepatocytes in patients with liver cirrhosis, who had developed hepatoma during follow-up periods of several months to several years, appeared to resemble that in noncancerous parts of hepatoma cases. On the other hand, the incorporation of tritiated thymidine into hepatocytes was found to be markedly increased in noncancerous parts as well as in cirrhotic liver developing hepatoma during follow-up periods. These results suggest the possibility that the hepatocytes in noncancerous parts of hepatoma have deranged cell-kinetics which might be a driving factor for the development of malignancy.

    Notes: Summary A consecutive demonstration of both aminergic and cholinergic nerves of rat cerebral arteries was carried out on whole mount preparations. For demonstration of aminergic nerves the glyoxylic acid method was used, while for cholinergic nerves Karnovsky's technique was utilized consecutively. This procedure provided a highly sensitive and reproducible demonstration of the two systems on the same specimen. The results obtained were as follows: 1) Aminergic and cholinergic nerves were distributed densely in the proximal portions of the major cerebral arteries examined. They were dense in the arteries of the anterior circulation and sparse in the posterior circulation. Each nerve had two different patterns, i.e. circular and longitudinal. Both circular and longitudinal patterns were observed in the proximal portions of the arteries, while in the distal portions, longitudinal fibres were predominantly present. 2) Superior cervical ganglionectomy produced no change in cholinergic nerve distribution, while it caused decreased density of aminergic nerves on the ipsilateral side on the arteries of the anterior circulation. The contralateral anterior cerebral arteries were partially affected. Bilateral sympathectomies abolished aminergic nerves in all arteries except the vertebral artery.

    Notes: Summary Localization of instilled 14C-bupranolol was studied in the rabbit eye by means of light microscopic radioautography. After the administration of 50 μl of 14C-bupranolol into the eyes of two rabbits, both eyes were enucleated at intervals of 15 and 30 min respectively, and radioautographic procedures were carried out on cryostat sections by means of both drymounting and wet-mounting methods. Although with the former it was possible to demonstrate many radioautographic silver grains in the specimens, with the latter it was not possible to detect their localization. On the dry-mounting procedure, radioautographic grains were observed mainly in the epithelium of the ciliary body 15 and 30 min after the administration of 14C-bupranolol. The numbers of grains were counted on the 15 and 30 min specimens. They were decreased in the conjunctiva, and cornea 30 min after the administration. On the contrary, they were remarkably increased in the epithelium of the ciliary body. From these results it is concluded that the 14C-bupranolol first penetrates into the epithelium of the conjunctiva and cornea, then moves rapidly to the ciliary body, and that only very small amount of 14C-bupranolol is changed into the insoluble form. Also, these results suggest that bupranolol plays a role in the control of aqueous production in the ciliary processes.

    Notes: Summary A method has been developed for electron microscopic histochemical demonstration of phospholipase B, (lecithinase B, E C 3.1.1.5, lysolecithin acyl hydrolase), which hydrolyzes α- and β-positions of phospholipids in mouse liver, kidney and adrenal tissues. Tissues either fixed in cold 1% paraformaldehyde or unfixed were cut into 40 μm frozen sections and were incubated at 37° C in a medium at pH 6.6 or 4.5 containing 2 μM lysolecithin and 0.25 mM CaCl2 for 20 min. The fatty acids liberated by enzymatic hydrolysis were trapped as calcium precipitate and were converted to lead precipitate by treatment with lead nitrate. The reaction products were observed by electron microscopy to be localized on the end of the smooth endoplasmic reticulum at pH 6.6 and in lysosomes and lipid droplets at pH 4.5. It is concluded that the products showed the localization of phospholipase B activity.
