Notes:
Summary A method for the determination of isosorbide dinitrate and diazepam in pharmaceutical preparations was developed. These substances are separated by HPLC on nucleosil C18 (mobile phase: methanol-ethylacetate-acetate buffer) and detected amperometrically at a SMDE at −1.15 V (pulse mode, basis potential: −0.4 V, pulse amplitude: −0.75 V). The detection limit is 8 ng of isosorbide dinitrate resp. 20 ng of diazepam (S/N=3). Pharmaceutical preparations, which additionally contain lecithines or triglycerides, are cleaned up. Lecithines are removed by adsorption on a silicagel cartridge, triglycerides are extracted by petrol ether. The amperometric detection is compared with UV detection. Isosorbide dinitrate is detected amperometrically more sensitively than by UV. On the contrary, diazepam is better detected by UV absorption.
