    Notes: The influence upon differnt cellular and humoral parameters of hemopoiesis of three structurally unrelated, highly purified bacterial cell-wall components (BCWC) was investigated. The spleens of C57BL/6 mice assayed 6 days after the injection of either lipid A or outer-membrane lipoportein, but not murein, showed a marked increase in granulocyte-macrophage, eosinophil, and megakaryocyte progenitor cell levels. The number of pluripotent hemopoietic stem cells (CFU-S) also increased in the spleens of mice treated with either lipid A or lipoprotein. Similar results were obtained following the injection of lipoprotein or lipid A into CBA or C57BL/6.nu mice. Genetically anemic Wf/Wf mice were found to have spontaneously elevated numbers of splenic progenitor cells, which increased further after the injection of lipid A. The proportions of the different splenic progenitor cell types were similar in both untreated and lipid A treated Wf/Wf mice, and in normal littermate controls.When tested in vitro, unfractionated or partially purified post-lipid A serum was found to stimulate the growth of granulocyte-macrophage progenitor cells (GM-CFC), but no detectable stimulation of eosinohphil, megakryocyte, or erythroid progenitor cells was observed. The data suggest that the rise in splenic levels of the different progenitor cells is not mediated by the corresponding types of CSF, but more likely by proliferation and differentiation of CFU-S.

    Notes: Molecular changes occur at the surface of hemopoietic cells during differentiation from progenitor cells to mature granulocytes and macrophages. The differential expression of surface carbohydrate residues has been probed using lectins and the results used to purify normal mouse granulocyte-macrophage progenitor cells. Ten different lectins were screened for selective interaction with mouse hemopoietic colony-forming cells (CFCs), using agglutination or a quantitative analysis of the number of fluoresceinated lectin molecules bound per cell using a fluorescence activated cell sorter (FACS). Pokeweed mitogen (PWM), Helix pomatia agglutinin (HPA), soybean agglutinin (SBA), and peanut agglutinin (PNA) preferentially bound to CFCs so that it was possible to enrich 4 to 10-fold for these progenitor cells by sorting for the highly fluorescent cells. Further analysis of the low and high angle light scattering characteristics of the CFCs indicated that these cells were polydisperse, but could be enriched ten-fold by selecting for cells with high intensity low angle (90°) scatter and low intensity high angle (90°) scatter. PWM gave the best enrichment (10 to 15-fold) for adult bone marrow CFCs, for CFCs from fetal sources (fetal liver, fetal blood), and for CFCs from the spleens of mice injected previously with outer membrane lipoprotein from E. coli. Three parameter sorting for CFC using the FACS (low angle scatter, high angle scatter, and PWM-fluorescence) resulted in large enrichment factors (16 to 50-fold) for CFCs from all the above sources. Over 7% of the cells sorted from bone marrow, 10% of the cells sorted from post-lipoprotein spleen, and 28% of the cells sorted from fetal peripheral blood were hemopoietic CFCs. Ninety percent of the cells in these fractions had the morphology of blast cells or myelocytes. Thus, it was possible to identify the morphological characteristics of the hemopoietic progenitor cells. Screening of other developmental systems using quantitation of fluorescence with lectins should prove of general value for the purification of selected differentiation states.

    Notes: Merocyanine 540 (MC 540) is an impermeant fluorescent dye that binds preferentially to fluidlike domains of the cell membrane. Photoexcitation of membrane-bound dye causes a breakdown of the normal permeability properties of the membrane and, eventually, cell death. We have used in vitro and in vivo clonal assays to determine the relative sensitivities of different classes of normal murine hematopoietic progenitor cells to MC 540-mediated photo-sensitization. Late erythroid progenitors (CFU-E) were the most sensitive cells, followed in order of decreasing sensitivity by early erythroid progenitors (BFU-E), megakaryocyte progenitors (CFU-Meg), day 7-spleen colony forming cells (day 7-CFU-S), granulocyte/macrophage progenitors (CFU-GM), and day 11-spleen colony forming cells (day 11-CFU-S). Bipotent progenitors of the granulocyte/macrophage lineage were more sensitive than unipotent macrophage progenitors but less sensitive than unipotent granulocyte progenitors. Progenitors giving rise to large granulocyte/macrophage colonies were more sensitive than progenitors giving rise to small colonies (“clusters”). We conclude that sensitivity to MC 540-mediated photosensitization is developmentally regulated and that differences occur even between the most closely related classes of progenitor cells. Our findings indicate the usefulness of MC 540 as a plasma membrane probe. They also support the contention that early and late-appearing spleen colonies are the progeny of two distinct classes of progenitor cells.

    Notes: In culture, vascular smooth muscle cells proliferate until they form a confluent sheet of cells. At that time the morphology of the culture becomes altered and the cells form multilayered regions that eventually develop into nodular aggregations. We now demonstrate that the transition from monolayer culture to nodular culture is influenced by the presence of components in conditioned media. The development of nodules is enhanced by conditioned medium made from nodular cultures but is either inhibited or unaffected by monolayer culture-conditioned medium. Examination of the two types of conditioned media using NaDodSO4-polyacrylamide gels reveals many similarities and one major difference. Nodular-conditioned medium contains a prominent 42 kilodalton polypeptide which is not present in monolayer-conditioned medium. Further, we demonstrate that although both nodular and monolayer cultures produce fibronectin the transition to nodular culture does not occur in the presence of exogeneously added plasma fibronectin.

    Notes: Before antigen ever enters the immune system we believe that lymphocytes with antigen-specific receptors preexist. For B lymphocytes we know that these antigen binding stuctures are immunoglobulin (Ig) molecules, antibodies. One B cell makes one of the many Ig molecules and displays them in of antibody molecules we are tempted to think that any antigen can find its specific binding receptors, i.e., its preexisting antibody molecules. Essential problems of how to produce large quantities of such antigen-specific antibodies are how to stimulate the preexisting lymphocytes from their resting state and how to increase the production of that specific antibody. Proliferation and maturation to high rate-antibody secreting cells follows stimulation and therefore provides ways by which the synthesis of the preexisting antibodies is expanded. The motivation to understand the induction of specific andtibody production and secretion in the corresponding B lymphocytes in vitro may come either from the desire to understand the rules that govern the reactions of B cells leading to such antibody production, or from the need to do it outside of the body due to the potentially harmful effects of some antigens in vivo.
