    Notes: Summary MDCK cells form monolayers which have many of the properties usually found in transporting epithelia. The present article is devoted to the study of the structural and functional polarization of MDCK cells, which is one of the central features of transporting epithelia. The results show: (i) that MDCK monolayers transport 2.6 μmol hr−1 cm−2 of sodium in the apical to basolateral direction; (ii) the passive flux of this ion is relatively large (20.3 mole hr−1 cm−2), which is a characteristic of leaky epithelia; (iii) a large fraction of the penetration of sodium into the cells proceeds through an amiloride-sensitive channel, and the exit is operated mainly by a ouabain-sensitive pump; (iv) the net transport of sodium from the apical to the basolateral side agrees with the asymmetric labeling of the pumps with3H-ouabain; (v) this asymmetric labeling agrees, in turn, with a higher concentration of intramembrane particles (IMPs) in freeze-fracture replicas of the basolateral side of the plasma membrane; (vi) the structural polarization of confluent MDCK cells is also revealed by the location of microvilli, occluding junctions, and pinocytotic vesicles; and (vii) the presence of a continuous ring formed by actin microfilaments visualized by immunofluorescence under the lateral aspect of the plasma membrane that may be related to the distribution of the occluding junctions, which act as barriers separating apical from basolateral membrane components.

    Notes: Summary MDCK cells (epithelioid of renal origin) form monolayers which are structurally and functionally similar to transporting epithelia. One of these similarities is the ability to form occluding junctions and act as permeability barriers. This article studies the junctions of MDCK monolayers formed on a permeable and transparent support (a disk of nylon cloth coated with collagen) by combining two different approaches: (i)Scanning of the electric field: the disk is mounted as a flat sheet between two Lucite chambers and pulses of 20–50 μA cm−2 are passed across. The apical surface of the monolayer is then scanned with a microelectrode to detect those points where the current is flowing. This shows that the occluding junctions of this preparation are not homogeneous, but contain long segments of high resistance, intercalated with sites of high conductance. (ii)Freeze fracture electron microscopy: the junctions are composed of regions of eight to ten strands intercalated with others where the strands are reduced to one or two ridges. The sites of high conductance may correspond to those segments where the number of junctional strands is reduced to 1 or 2. It is concluded that the occluding junctions of MDCK monolayers are functionally and morphologically heterogeneous, with “tight” regions intermixed with “leaky” ones.

    Notes: Summary Clostridium thermocellum ATCC 27405 (and its improved cellulase-producing mutant, AS-39) is an anaerobic thermophile that produces endo-β-glucanase and exo-β-glucanase when grown on cellobiose or cellulose as major carbon source (Shinmyo et al. 1979). The site of cellulase accumulation was at least 95% extracellular. Optimum conditions for endo-β-glucanase production in flasks included 1% (w/v) cellobiose, 0.2% (w/v) urea as a nitrogen source, 0.1 M morpholinopropane-sulfonic acid buffer, an initial pH of 7.4, and a yeast extract concentration of 0.6% (w/v). An improved medium (GS medium) was devised for future studies. Xylan was degraded by an extracellular enzyme (s) produced during cultivation on cellobiose, although C. thermocellum does not grow on xylan.

Si presentano i parametri del reticolo, la banda proibita, la suscettività magnetica e il punto di fusione di tutti i materiali.
    Abstract: Резюме Приготовляются монокристаллы AgInS2 и AgInSe2, исполъзуя метод транспорта иода и метод Бридгмана. Также проводится синтез четярехкомпонентнях сплавов AgInSe2(1−x)S2x. Из спектра фотопроводимости определяются расцепления верхних валентных зон в орторомбическом AgInS2. Приводятся параметры рещетки, щирина запрещенной зоны, магнитная воспримчивостъ и точка плавления для всех соединений.
    Notes: Summary AgInS2 and AgInSe2 single crystals have been prepared by chemical transport with iodine and by the Bridgman method. Syntheses of the quaternary alloys AgInSe2(1−x)S2x have also been carried out. The splittings of the uppermost valence bands in orthorhombic AgInS2 have been determined from the photoconductivity spectrum. The lattice parameters, the energy gap, the magnetic suceptibility and the melting point of all the materials are presented.

    Notes: Abstract The stress-strain diagrams of a series of melt-crystallized polyethylene samples with a varying number of chain defects have been investigated. The elastic modulus,E, and the surface hardness,MH, markedly decrease with increasing number of defects. The mechanical behaviour of the lamellar structure of PE modulated by a major exclusion of chain defects from the crystals is discussed in the light of Takayanagi's two-phase model. The data suggest thatE is very sensitive to the fraction of tight crystalline bridges between lamellae. The correlation found betweenE andMH emphasizes, in addition, the different and complementary role played by the amorphous layer in each mechanical test. In the former case one measures the elastic deformation of the layer reinforced by tie molecules. In the latter test the plastic deformation under compression of the lamellae sandwiched between noncrystalline layers is contemplated. In both cases the influence of the number of defects drastically affects the nature of the crystalline lamellae and surface layer and consequently substantially modifies both types of properties.

    Notes: Summary The presence of the microtubule-associated protein (MAP2) in the brain of several species has been investigated by SDS-gel electrophoresis and by radioimmunoassay. This assay had a sensitivity of approx. 10 ng and it was capable of measuring the protein either in purified microtubules or in crude brain extracts. As determined with this radioimmunoassay, MAP2 accounted for about 10% of the porcine brain microtubule protein and 1% of the protein from a brain extract. Taking porcine MAP2 as a reference, we have detected polypeptides with the same electrophoretic mobility in brain microtubules from cow, sheep, rat and chicken. Nevertheless, the MAP2 from these species showed a variable degree of immunoreactivity. Bovine MAP2 appeared closely related to the porcine protein whereas the rat antigen showed low cross-reaction and chicken MAP2 appeared immunologically unrelated to porcine MAP2. Our results suggest a higher variability of the MAP2 sequences as compared to that reported by other authors for the brain microtubule protein, tubulin.

    Notes: Summary Depolymerization kinetics of microtubules assembled to steady-state by pod ophyllotoxin treatment show a dose-dependent effect of this mitotic poison on the net rate of microtubule disassembly. Pulse-chase experiments with microtubules at steady-state indicate that the depolymerization effect induced by superstoichiometric concentrations of podophyllotoxin relative to tubulin is polar and time-dependent, i.e. the rate of tubulin loss decreases along with the time of treatment in the presence of the drug. Under these conditions the rate of microtubule disassembly is much faster than one could expect from a unique effect of drug-tubulin complex on the microtubule assembly end. Podophyllotoxin-tubulin complex is not able to induce active depolymerization of microtubules, while free podophyllotoxin is. These results are consistent with the hypothesis that this drug acts on the microtubule assembly-disassembly process by two different mechanisms: 1) as a free drug, it actively promotes polar depolymerization of microtubules, and 2) as a drug-tubulin complex, it retards the addition of subunits into the microtubule ends.
