    Notes: Summary Single and sequential double immunocytochemical techniques were applied to localize gamma-aminobutyric acid (GABA)-and choline acetyltransferase (ChAT)-like immunoreactivity (-LI) in the hypoglossal nucleus of the rat. After subsequential double staining a relatively high number of hypoglossal motor neurons showed the coexistence of both ChAT-and GABA-LI. Coexistence of both substances was also revealed in the axons of the hypoglossal nerve situated within the medulla oblongata. Cells showing only ChAT-or GABA-LI were also observed. Differences in immunostaining between the different cell groups of the hypoglossal nucleus were established. Following axotomy of the right hypoglossal nerve, a decrease or loss of the immunoreactivity for both ChAT and GABA in the motor neurons was established until the 3rd week after the operation. The results obtained do not give evidence on the origin of the GABA-like immunoreactive material and its functional significance in the cholinergie neurons. It can be only speculated that the GABA-like material is either taken up from the intercellular space or is synthesized by the ChAT-LI nerve cells. Functionally, the importance of GABA for the synthesis of gamma-hydroxybutyrate (a novel neurotransmitter candidate) and its postsynaptic transmitter action or presynaptic regulatory action (through autoreceptors in the membrane of the nerve endings) on the release of acetylcholine (ACh) should be taken into consideration.

    Notes: Summary Ultrastructural localization of adenylate cyclase (AC) activity was investigated in suspensions of unfixed isolated rat thymocytes using a medium containing 0.6 mM 5′-adenylylimidodiphosphate (AMP-PNP) as a substrate, 10 mM MgSO4 as an activator, 5 mM theophylline as an inhibitor of 3′,5′-AMP-phosphodiesterase and 2 mM lead nitrate as a capturing agent. AC activity was demonstrated in plasma membrane, perinuclear space, endoplasmic reticulum, Golgi complex, centriole microtubules and mitochondria. AC was activated with 10−4 M adrenalin in the presence of 5′-guanylylimido-diphosphate (GMP-PNP) as well as with 10−2 M NaF. In the cells incubated in a medium devoid of theophylline and containing 5′-AMP instead of AMP-PNP, 5′-nucleotidase activity was observed in the same cell structures as AC activity. Hydrolysis of 5′-AMP in the nucleus was much stronger than that of AMP-PNP. 10 mM NaF markedly inhibited hydrolysis of 5′-AMP in all cell structures. No staining was observed with 2 mM β-glycerophosphate as a substrate. Incubation of unfixed thymocytes in media containing AMP-PNP, 5′-AMP or p-nitrophenyl phosphate, but not β-glycerophosphate, induced both in the nucleus and in the cytoplasm in some cells an appearance of a transitory reticular formation consisting of about 30 nm thick strands which could penetrate the nuclear envelope and plasma membrane and form connections with adjacent cells. The transitory reticular formation seems to belong to the cytoskeleton and to be involved in cell aggregation.

    Notes: Summary The effects of different preparative and cytochemical procedures on adenylate cyclase (AC) activity in heart muscle homogenates were studied by quantitative analysis. We were mainly concerned with perfusion prefixation, using glutaraldehyde (GA) with and without DMSO, and with the influence of cytochemical incubation with lead ions as the capture reagent. Furthermore, we measured the direct effect on the AC activity in lead, cobalt, and strontium ions in prefixed heart homogenates. We also studied the influence of phosphatidylinositol and 2′,5′-dideoxyadenosine. The following results were obtained: 1. Perfusion fixation using 2% GA buffered with cacodylate reduced the AC activity by about 20%. After the entire cytochemical procedure was finished, 20% of the original AC activity was still present. Stimulation by epinephrine, histamine and fluoride, which increased the activity of AC two or three times in our experiments, was only slightly reduced by the cytochemical treatments. 2. Lead ions (2 mM), added to the biochemical assay without chelating compounds, reduced the AC activity about 90%. 5 μM phosphatidylinositol stabilized the fluoride- and hormone-sensitive AC activity. 3. Co2+ also reduced the AC activity, though less than Pb2+. Sr2+ had no effect on the basic activity of the AC but had a slightly inhibitory effect on the hormone and fluoride stimulation. 4. 5% DMSO added to the fixative had no influence on the basic activity of the AC. However, this solvent definitely reduced the level of stimulation by fluoride and guanine nucleotide plus hormones. 5. A potential inhibitor of enzyme activity and of the hormone- and fluoride-sensitive AC was the adenosine derivative, 2′,5′-dideoxyadenosine. This compound, at a concentration of 10−3 M, inhibited all AC activity in the heart homogenates.

    Notes: Summary The localization of guanylate cyclase activity was cytochemically studied in heart tissue from guinea pig and pigeon. The method, based on a lead precipitation technique with GPPNHP as the substrate, was tested by quantitative biochemical analysis. The data obtained showed that in heart homogenates GPPNHP is an acceptable substrate for guanylate cyclase. The guanylate cyclase activity of glutaraldehyde prefixed heart tissue was also measured in the presence of 2 mM lead nitrate, in 30% of the untreated control hearts. The residual guanylate cyclase responded to the addition of sodium nitroprusside with a 7-fold increase in its activity. Furthermore, the guanylate cyclase requirement for Mn2+ ions was so changed by this activator that Mg2+ was as active as Mn2+. In heart muscle cells of guinea pigs and pigeons the plasma membrane of the sarcolemma and the junctional sarcoplasmic reticulum are the precipitation sites of the reaction product. In guinea pig hearts the T-tubule membranes were likewise covered with precipitates. Sodium nitroprusside stimulation of guanylate cyclase activity was indicated by increased precipitation and by shortening of the incubation time.

    Notes: Abstract The dielectric and piezoelectric properties (d-p properties) of four kinds of doped lead titanate zirconate piezoelectric ceramics (PZTs) have been measured from 4.2 to 300 K. The d-p properties of the materials converge with decreasing temperature down to liquid helium temperature, even though the properties have large differences at room temperatue. The values of mechanical and electrical quality factors,Q m,Q e, and of the frequency constant,N, of the materials increased at low temperature. It is evident from the freeze out inK′ and the associate temperature: frequency-dependent maxima in tan 5 that the relaxation processes including ferroelectric domain wall motion and thermal defect motion contribute to the d-p properties. The Navy type-III composition has a minimum temperature coefficient of d-p parameters and it is evident that PZT ceramics modified with Fe2O3 can provide good stability and also give the strongest piezoelectric response at liquid helium temperature.

    Notes: Abstract It has been previously established that the surface plasmon of small spherical silver particles, which are embedded in a noble gas matrix, shifts to higher energies (blue shift) as the mean diameterD of the particles decreases (100 Å〉D〉20 Å). This blue shift has also been found for supported silver particles, and quite recently we observed it by elastic light scattering in the gas phase. This latter experiment proves unambiguously that the blue shift in small silver particles is not induced by interactions with the environment, the presence of which is clearly recognized in less inert matrices such as O2 or CO. From self-consistent calculations of the surface response of planar jellium surfaces one would expect a red shift, which is also directly confirmed by a few calculations for selected jellium spheres. The contradiction between the observed blue shift for small particles and the predicted red shift for jellium spheres disappears, if one accounts for thed-electrons of silver in a very simple approximation.

    Notes: Abstract The structures of Ag, Fe and Ge microclusters were determined using EXAFS. The measurements were performed over a wide range of clusters sizes. The clusters were prepared using the gas aggregation technique and isolated in solid argon at 4.2 K. The measurements were performed at the National Synchrotron Light Source (NSLS) at beam line X-18B. A strong contraction of the interatomic distances was observed for Ag dimers and multimers. Silver clusters larger than 12 A mean diameter show a small contraction of thenn distance and a structure consistent with an fcc lattice. By contrast clusters smaller than 12 A show the presence of a small expansion and a strong reduction or absence ofnnn in the EXAFS signal. This points towards a different crystallographic structure for Ag microclusters with diameter less than 12 A. In iron clusters we observe a gradual reduction of thenn distance as the cluster size decreases. The interatomic distance for iron dimers was determined to be 1.94 A, in good agreement with earlier measurements. The iron microclusters show a bcc structure down to a mean diameter of 9 A. Iron clusters with 9 A mean diameter show a structure inconsistent with a bcc lattice. The new structure is consistent with an fcc or hcp lattice. The measurements on Ge clusters show the presence of only nearest neighbors. There was clear evidence of temporal annealing as determined by variations in the near edge structure of the K-absorption edge. Absorption edge measurements were also performed for free Ge clusters travelling perpendicular to the direction of the synchrotron radiation beam. The measurements performed on the free clusters were consistent with those obtained for matrix isolated clusters.
