    Notes: Previous studies suggest that, besides the maldigestion of lactose in the small intestine, the colonic processing of lactose might play a role in lactose intolerance. β-Galactosidase is the bacterial enzyme which catalyzes the first step of lactose fermentation in the colon. We propose a practical method to differentiate and identify bacteria with β-galactosidase activity in faeces which combines a colony-lift filter assay with X-gal (5-bromo-4-chloro-3-indolyl-β-d-galactopyranoside) as substrate for differentiation and the fluorescent in situ hybridization technique for identification. The method was applied to faeces from lactase non-persistent subjects. After 28 subjects had undergone one glucose and two lactose challenges, consistent intolerant (n= 5) and tolerant (n= 7) groups were defined according to their symptom scores. Of the 28 faecal samples, 80.6% (mean, SD: 12.1, range: 47.8–100%) of the total cultured bacteria were found to possess β-galactosidase activity, which indicates that the bacterial β-galactosidase is abundant in the colon. The tolerant and intolerant groups did not differ in the percentage or composition of the bacteria with β-galactosidase activity or β-galactosidase activity in faeces. Results suggest that the percentage or composition of the bacteria with β-galactosidase activity in faeces do not play a role in lactose intolerance.

    Notes: Transport and metabolism of 8-14 C-zeatin, applied to an attached de-tipped one-year-old mature leaf of a Yucca plant bearing a bleeding inflorescent stalk, has been studied. Radioactive zeatin ribotide was found in the exudate of the bleeding inflorescence, which was collected over a period of 5 days. Radioactive zeatin ribotide was mainly extracted from the fed leaf. Minor conversion products in this leaf were zeatin ribotide, zeatin-o-β-glucoside and zeatinriboside o-β-glucoside.In not zeatin fed plants, zeatin-o-β-glucoside was tentatively identified as the main endogenous cytokinin in one-year-old mature leaves. In the bleeding sap of not treated plants no free bases of zeatin or zeatin ribosides were found. After alkaline phosphatase treatment zeatin-riboside was detected by combined gas chromatography-mass spectrometry, indicating the presence of zeatin ribotide in the bleeding sap. High β-glucosidase activity was found in the stern.Results suggest that stared cytokinin glucosides from Yucca leaves are, converted by β-glucosidase in leaves and stem, transported through the inflorescent stalk as zeatin nucleotides.

    Notes: Various sources of random deviations occurring in the determination of X-ray intensities with the aid of film methods are discussed and corresponding mathematical expressions are derived. From a comparison with experimental values of the standard deviations obtained from measurements on homogeneously exposed films, it follows that the deviations are primarily caused by the variations of the X-ray photon flux due to the stochastic nature of the X-ray emission process. The accuracy attainable with most X-ray films is found not to be limited by saturation of the number of excited AgBr particles occurring at increasing exposures, but at low exposures film methods are at a disadvantage because of the presence of fog. If, however, small signals superimposed on a high sample background have to be measured, relatively high exposures are required; in this case film registration is found to be much faster than methods involving step-scanning counters, and in general little slower than registration with the aid of a position-sensitive counter. It is also shown that the dynamic range of the film method may be extended without sacrificing accuracy by resorting to slower films.

    Notes: The method of Ruland [Acta Cryst. (1961). 14, 1180–1185] for the determination of crystallinity has been modified in order to take account of a difference in chemical composition of the crystallized and non-crystallized fractions, such as occurs in glass-ceramic materials. Application to two samples leads to higher crystallinities than found by other X-ray methods, and shows a large increase of disorder with decreasing crystallinity.

    Notes: Abstract Flower-bud blasting in Iris occurs in the winter when low light intensities and short days prevail. After introduction of 14CO2 to one leaf the transport of assimilates was studied under controlled culture conditions in a control light treatment and in a treatment of 7 days darkness followed by standard light conditions. Little assimilate transport was found in the direction of the bud in dark-treated plants. However, zeatin injection into the flower buds of the plants subjected to the dark treatment clearly promoted assimilate transport ot these buds. Abscisic acid levels, determined by gas chromatography, were found to increase in the buds of dark-treated plants. Zeatin injection into the flower bud resulted in a suppression of the abscisic acid level. The latter treatment also resulted in higher percentage of flowering. Removal of flower parts was found to inhibit peduncle elongation. The peduncle elongation of complete flowers started in a well defined period, and the fresh weight of buds was found to increase mainly in the last part of that period. Assimilate transport under low light intensities in relation to abscisic acid and supposed gibberellin is discussed.

    Notes: Abstract The soil ecosystem is composed of various groups of organisms which have complex relations. The physical structure and chemical characteristics of the soil provide the boundary conditions. In view of various deteriorating human activities, it is important to find soil quality characteristics with respect to its most important function: the ecological function. An enumeration has been given of chemical, physical and biological soil parameters which are more or less important for soil quality. Several of these parameters are discussed. For use as indicators of deterioration, for a given site, the optimum values of the soil parameters have to be established, as well as acceptable deviations from the optimum, taking into account natural fluctuation. It is concluded that, due to lack of data, such an approach is not possible at this moment. However, it might be possible to identify those soil parameters which should be taken into consideration when evaluating human activities.
