    Notes: Abstract Heart sarcolemma has been shown to possess three catalytic sites (I, II and III) for methyl transferase activity (Panagia V, Ganguly PK and Dhalla NS. Biochim Biophys Acta 792: 245–253, 1984). In this study we examined the effect of phosphatidylethanolamine N-methylation on ATP-independent Ca2+ binding and ATPase activities in isolated rat heart sarcolemma. Both low affinity (1.25 mM Ca2+) and high affinity (50 µM Ca2+) Ca2+ binding activities were decreased following incubation of sarcolemmal membranes with AdoMet under optimal conditions for site II and III. Similarly, Ca2+ ATPase activities measured at 1.25 mM and 4 mM Ca2+ were depressed by phospholipid N-methylation. S-adenosyl homocysteine, a specific inhibitor of phospholipid N-methylation, prevented the depression of low affinity Ca2+ binding and Ca2+ ATPase activities, whereas the methylation-induced effect on the high affinity Ca2+ binding was not influenced by this agent. Pretreatment of sarcolemma with methyl acetimidate hydrochloride, an amino group blocking agent, also prevented the methylation-induced inhibition of both Ca2+ binding and Ca2+ ATPase. A further decrease in Ca2+ binding and Ca2+ ATPase activities together with a marked increase in the intramembranal level of PC was seen when membranes were methylated under the site III conditions in the presence of phosphatidyldimethylethanolamine as exogenous substrate. There was no effect of phospholipid methylation on sarcolemmal Na+-K+ ATPase and Mg2+ ATPase activities. These results indicate a role of phospholipid N-methylation in the regulation of sarcolemmal Ca2+ ATPase and low affinity ATP-independent Ca2+ binding.

    Notes: Abstract Premethylation of purified porcine cardiac sarcolemma (SL) in the presence of 0.15, 10 and 150 µM S-adenosyl-L-methionine (AdoMet) did not change the phosphorylation of SL proteins catalyzed either by intrinsic cyclic AMP-dependent protein kinase (cAK) or by added catalytic (C) subunit of this enzyme. On the other hand, membrane exhibited increased lipid methyltransferase activity after preincubation with MgATP and C subunit. Prephosphorylation of membranes stimulated the total [3H]-methyl incorporation into SL lipids assayed at 0.15 µM [3H]AdoMet due to an enhancement of Vmax and without changes in the Km value for AdoMet. Analysis of the methylated lipid products revealed an increased methyl group incorporation into a nonpolar lipid fraction whereas phosphatidylethanolamine-N-methylation was not affected by phosphorylation. The results suggest that the cyclic AMP-mediated signal transduction at the level of cardiac SL is not affected by methylation-induced modifications of the membrane lipid microdomains. On the other hand, an intrinsic SL lipid methyltransferase activity is apparently not related to the N-methylation of phospholipids, is modulated by cyclic AMP-dependent protein phosphorylation.

    Description / Table of Contents: Riassunto Sono state rilevate durante un monitoraggio aerobiologico effettuato a Pietra Ligure (Savona) nel corso del 1987 le concentrazioni polliniche di 50 taxa ed è stata valutata l'influenza relativa dei fattori meteorologici. Le osservazioni palinologiche sono state rapportate alle concentrazioni sieriche delle IgE specifiche, alla reattività bronchiale specifica ed aspecifica valutate in 101 pazienti allergici (rinitici ed asmatici), sensibilizzati a Graminaceae ed Urticaceae (Parietaria) al fine di riconoscere correlazioni tra le caratteristiche aerobiologiche di questi allergeni edi meccanismi patogenetici che sostengono la reattività bronchiale.
    Notes: Summary Airborne pollen grains in Pietra Ligure (Savona, Italy) related to immunological and bronchoreactive characters in atopic subjects. The pollen concentration of 50 taxa and the influence of the meteorological parameters was observed by means of an aerobiological monitoring carried out in Pietra Ligure (Savona, Italy) from January to December 1987. The study includes a survey of atmospheric pollen types of main clinical significance, i.e. Oleaceae, Urticaceae (Pellitory), Graminaceae and Compositae. For these families the variations of pollen counts are presented taking into account both daily means and five-day running means. The palynological observations have been related with the serum specific IgE concentrations, with specific and non specific bronchial reactivity measured in 101 rhinitic and asthmatic patients sensitized to Graminaceae and Urticaceae (Pellitory) in order to recognize the correlation between the aerobiological features of these allergens and the pathogenetic mechanism producing the bronchial hyperresponsiveness.

    Notes: Abstract Both phosphatidylethanolamine(PE)-N-methylation and phosphatidyl-inositol bisphosphate(PI-bisphosphate) breakdown potentially modify the microdomains in the sarcolemmal lipid bilayer. In this study the possibility of a mutual interaction between the enzymes responsible for these phospholipid reactions is examined. In sarcolemma purified from rat heart, prior hydrolysis of PI lipids by exogenous specific phospholipase C inhibited (to 75, 59 and 78% of control for sites 1, 11 and 11, respectively) the PE-N-methyltransferase system. In cultured rat cardiomyocytes the addition of L-methionine, a precursor for the methyl donor S-adenosylmethionine, stimulated PE-N-methylation in a concentration (0.2–300 µM)-dependent manner. Methionine (50 µM) decreased the basal rate of PI-bisphosphate hydrolysis (to 72% of control), but had no effect on the phenylephrine-stimulated PI-bisphosphate hydrolysis. Maximal activation of the PI-bisphosphate breakdown by 30 µM phenylephrine did not affect the rate of PE-N-methylation in the presence of exogenous methionine (50 µM). These findings support the existence of interactions, although discrete, between the enzymes involved in the PE-N-methylation and PI turnover.

    Notes: Abstract In order to identify defects in Na+-Ca2+ exchange and Ca2+-pump systems in cardiomyopathic hearts, the activities of sarcolemmal Na+-dependent Ca2+ uptake, Na+-induced Ca2+ release, ATP-dependent Ca2+ uptake and Ca2+-stimulated ATPase were examined by employing cardiomyopathic hamsters (UM-X7.1) and catecholamine-induced cardiomyopathy produced by injecting isoproterenol into rats. The rates of Na+-dependent Ca2+ uptake, ATP-dependent Ca2+ uptake and Ca2+-stimulated ATPase activities of sarcolemmal vesicles from genetically-linked cardiomyopathic as well as catecholamine-induced cardiomyopathic hearts were decreased without any changes in Na+-induced Ca2+-release. Similar results were obtained in Ca2+-paradox when isolated rat hearts were perfused for 5 min with a medium containing 1.25 mM Ca2+ following a 5 min perfusion with Ca2+-free medium. Although a 2 min reperfusion of the Ca2+-free perfused hearts depressed sarcolemmal Ca2+-pump activities without any changes in Na+-induced Ca2+-release, Na+-dependent Ca2+ uptake was increased. These results indicate that alterations in the sarcolemmal Ca2+-efflux mechanisms may play an important role in cardiomyopathies associated with the development of intracellular Ca2+ overload.

    Description: Some exploding stars (supernovae) are powerful emitters of centimeter radio radiation. Detailed observations have shown that these supernovae quickly become detectable in the radio range, first at shorter wavelengths (higher frequencies) and later at progressively longer and longer wavelengths (lower frequencies). This part of the phenomenon appears to be well explained by a monotonic decrease in the amount of ionized material surrounding the radio-emitting regions as the shock from the explosion travels outward. The radio emission itself is of a nonthermal, synchrotron origin, as is the case in most bright cosmic radio sources. Once the absorption effects become negligible, the radio intensity declines with time until reaching the detection limit of the telescope. Models suggest that the absorbing material originates in a dense wind of matter lost by the supernova progenitor star, or by its companion if it is in a binary system, in the last stages of evolution before the explosion. The synchrotron radio emission can be generated either externally by the shock wave from the explosion propagating through this same high density stellar wind or internally by a rapidly rotating neutron star, which is the collapsed core of the exploded star.
