    Description / Table of Contents: Whole cells of Corynebacter. simplex were entrapped by crosslinking water-soluble poly[acrolein-co-(1-vinyl-2-pyrrolidone)]. As crosslinking agents, several alkylenedioxydiamines or oxa-alkylenedioxydiamines were synthesized. The polymer-cell-conjugates showed steroid-Δ1-dehydrogenase activity and the production of prednisolon from cortisol was investigated. The activity was dependent on the hydrophobicity of the entrapping gel and could be increased about tenfold by enzyme induction and incubation in peptone containing nutrient media. The steroid transformation was advantageously carried out with an excess of microcrystalline cortison. In this case, the transforming capacity was 0.57 g prednisolone per day and per g biocatalyst.
    Notes: Ganze, lebensfähige Zellen von Corynebacter. simplex wurden in ein wasserlösliches Poly[acrolein-co-(1-vinyl-2-pyrrolidon)] durch Vernetzung eingeschlossen. Als Vernetzer wurden verschiedene Alkylendioxydiamine und Oxaalkylendioxydiamine synthetisiert. Die Polymer-Zell-Konjugate zeigten eine Steroid-Δ1-dehydrogenase-Aktivität, und die Bildung von Prednisolon aus Hydrocortison wurde untersucht. Die Aktivität war abhängig von der Hydrophobie des einschließenden Gels und konnte durch eine Enzyminduktion und eine Behandlung mit einer Pepton-enthaltenden Nährlösung auf ungefähr das Zehnfache gesteigert werden. Vorteilhafterweise konnte die Steroidumwandlung mit einem Überschuß an mikrokristallinem Hydrocortison durchgeführt werden. In diesem Fall wurden 0.57 g Prednisolon pro Tag und pro g Biokatalysator erhalten.

    Notes: Several amide derivatives of phenylalanin and prolin were prepared as spacer/ligand compounds by reaction of unprotected or monoprotected aliphatic and aromatic diamines and monoamines with the succinimido esters of the corresponding N-protected amino acids. The formation of diacylated aliphatic diamines was fast under normal conditions. In the case of aromatic diamines the presence of a bases is necessary. For the preparation of monoacylated diamines best results were obtained with monoprotected diamines. The acylated diamines were purified by column chromatography or by extraction.

    Notes: Starting from dibenzo-18-crown-6, 1,10-diaza-18-crown-6 and 1,7-diaza-15-crown-5, several crown ether surfactants were synthesized. They all form stable monolayers on water. The methacryloyloxy derivative of 1,10-bis(3-dodecyloxy-2-hydroxypropyl)-1,10-diaza-18-crown-6, 2c, could be polymerized in the monolayer by UV irradiation. This polymerizable crown ether surfactant also forms synthetic vesicles by sonication in water.
