    Notes: The sensitivity and selectivity of double modulation Fourier transform infrared reflection absorption spectroscopy for absorbing species on a reflecting surface has been employed for the in situ analysis of low-temperature plasma enhanced chemical vapor deposition formed SiO2 films deposited on HgCdTe, silicon, and aluminum substrates. An oblique angle of incidence of ∼55° was chosen to yield maximum sensitivity for the longitudinal optical (LO) phonon mode of SiO2 on Si. The peak frequency and shape of the LO mode absorption band varied with the quality of the SiO2 films thus providing a means of in situ assessment of reaction conditions at any stage of film growth. This diagnostic technique can be readily applied to the in situ analysis of dielectric thin films formed under a variety of reaction conditions.

    Notes: Abstract Different proteases from the culture fluids of recombinant Streptomyces lividans strains were isolated. Several individual proteases were separated and characterized. A chymotrypsin-chylike activity (CLA) was identified that specifically degrades a fusion protein between the α-amylase inhibitor from S. tendae (Tendamistat, HOE467) and proinsulin from the monkey Macaca fascicularis. The effective chemical inhibition of the degrading enzyme is demonstrated.

    Description / Table of Contents: Zusammenfassung Die vorliegende Arbeit behandelt den elastischen Stoß von Kugeln auf Sandwichplatten mit weichem Kern. Eine Integro-Differentialgleichung für den Stoßprozeß wird analytisch hergeleitet und numerisch integriert. Die Auslenkung der Platte unter der Aufprallstelle wird als Funktion der Zeit berechnet; es ergibt sich eine befriedigende Übereinstimmung mit experimentellen Werten. -Im Vordergrund steht das Resultat, daß dünne isotrope Platten und Sandwichplatten grundsätzlich verschieden auf einen Stoß reagieren.
    Notes: Abstract An integro-differential equation for the elastic impact of spheres on soft core sandwich plates is analytically derived. The impact equation is numerically integrated and the displacement of the sandwich plate underneath the point of impact is calculated as a function of time. Fairly good agreement with experimental results obtained by the author was found. It is emphasized that, physically, the impact response of a sandwich plate is fundamentally different from the response of an isotropic plate.

    Description / Table of Contents: Zusammenfassung Die vorliegende Arbeit diskutiert das physikalische Verhalten von “Biegewellen” in Sandwichplatten mit im Vergleich zum Kern dünnen, steifen und schweren Druckplatten. Mit Hilfe asymptotischer Entwicklungen der Grundgleichungen des linearelastischen Kontinuums wird gezeigt, daß in verschiedenen Frequenzbereichen ganz verschiedene physikalische Verhaltensweisen vorherrschen. Innerhalb beschränkter Frequenzbereiche können daher verschiedene, dafür verhältnismäßig einfache Bewegungsgleichungen Verwendung finden. Die Voraussagen einer solchen Gleichung, gültig für mäßige Frequenzen, werden verglichen mit Messungen, die an Glas-Polyurethan-Sandwichplatten ausgeführt worden sind. Die theoretischen und die experimentellen Dispersionskurven für rotationssymmetrische und somit auch für ebene Wellen stimmen gut überein.
    Notes: Summary This paper discusses the physical behaviour of “bending” waves in sandwich plates in which the facings are thin, stiff and heavy as compared with the core. By means of asymptotic expansions of the basic equations of linear elasticity, it is shown that different “physical mechanims” predominate in different frequency ranges. The consequence is that different, but relatively simple equations of motion may be used within limited frequency ranges. The predictions of such an equation, valid for moderate frequencies, are compared with measurements performed on glass-polyurethane sandwich plates. The agreement of theoretical and experimental dispersion curves for axisymmetric waves, and hence for plane waves as well, was found to be very satisfactory.

    Notes: Abstract In this study we addressed the question of whether scaffold structures produced from purified mitotic chromosomes are an artefact of dehistonization, and whether the integrity of the chromatin fibres is necessary for the maintenance of the well-known shape of mitotic chromosomes. Purified mitotic chromosomes from Friend erythroleukemia cells were treated either with increasing NaCl concentrations up to 500 mM, or with 6 M urea in the presence or absence of 10 mM 2-mercaptoethanol. The main criterion for the intactness of the overall chromosome shape as seen by electron microscopy was the characteristic X-or U-like appearance with clearly discernable chromatid axes. Histone H1 is known to be essential for the integrity of chromatin fibres. Its removal in sucrose gradients containing 500 mM NaCl did not lead to loss of the overall chromosome shape. However, treatment of chromosomes in sucrose gradients containing 10 mM 2-mercaptoethanol and 6 M urea led to loss of the structure probably due to dissociation (or denaturation) of shape-determining (scaffolding) components. Under these conditions most of the histones remained bound to the chromosomes, and the fibres in this chromatin material, after removal of excess urea and 2-mercaptoethanol, still showed condensation of the nucleosome filaments into the characteristic fibre structures upon increasing ionic strength. Our observations are compatible with the model that specific non-histone components, independently of histone-DNA interactions, organize or stabilize the structure of metaphase chromosomes.

    Notes: Abstract The ability of trichloroacetic acid (TCA) to act as an initiator or promoter of carcinogenesis was studied in the γ-glutamyl transpeptidase (GGT) enzyme altered foci (EAF) assay, using the rat partial hepatectomy model and diethylnitrosamine (DEN) phenobarbital (PB) positive controls. The capacity of TCA to induce peroxisomal proliferation was examined by measuring the induction of hepatic peroxisomal-dependent14C-palmitoyl-CoA. Induction of hepatic ornithine decarboxylase (ODC) was also measured. No evidence of initiating activity by TCA was evident in male Sprague-Dawley rats by the EAF assay. However, a significant increase in numbers and size of GGT-positive foci compared to controls were observed in rats exposed to 50, 500 or 5,000 mg/L TCA as a promoter for 3–12 months in the drinking water following initiation with DEN. A significant, though weak (10–20% over controls), stimulation of hepatic peroxisomal-dependent14C-palmitoyl-CoA was seen only in groups of rats promoted with TCA at 5,000 mg/L. A single large dose (1,500 mg/kg) of TCA produced a significant (26–70 fold) increase in hepatic ornithine decarboxylase activity 5 and 18 hr after administration. No changes in organ/body weights or significant histopathological lesions could be attributed to TCA exposure. These data suggest TCA possesses promoting activity in the rat hepatic model system. However, there was no dose-related correlation between the very weak peroxisomal proliferating effect and promoting activity.
