    Notes: An investigation was carried out to probe into the mating-type structure of a local population of the marine ciliate, Euplotes minuta. From this population, nine different mating types belonging to a unique set were isolated. The nine type-representative wild stocks analyzed were found to be heterozygous at the mating-type (mat) locus and provided, together with their sexual progeny, a total of 15 pure mating types. In E. minuta, the high-multiple nature of the basic mating system controlled by a series of peck-order alleles at a single locus should be considered a virtual certainty. The relationships among the genetic economies of the similar bottom-dwelling marine ciliates of the genus Euplotes, the E. vannus-crassus-minuta group, are discussed.

    Notes: A mating-type analysis was performed on 78 stocks of the marine hypotrich ciliate, Aspidisca sp., from a sufficient number of diverse geographic locations, some widely separated. Evidence is provided for the existence of a binary mating system in this “morphospecies.” The collected stocks have been challenged by the most rigorous criterion, namely breeding affinity in the laboratory, and have yielded at least four reproductively, not necessarily geographically, isolated groups that are in fact “biological species,” here referred to as “syngens.” Different syngens contain different pairs of mating types. Syngens are morphologically indistinguishable; hence Aspidisca sp. can be considered a conservative taxon comprising a number of “cryptic” or “sibling species.” Information is also presented about the mating behavior and the pattern of nuclear events at conjugation in Aspidisca sp. Search for soluble pheromones of the mating types gave only negative results. Hence, direct contact with potential partners is postulated to play a critical role in preparing individuals to mate. Mating reaction and mating which actually involves cross-fertilization (conjugation, sensu stricto) are completely inhibited by 10 μg/ml cycloheximide, suggesting the necessity of protein synthesis for recognition and union in conjugation of potential partners.

    Notes: Abstract This report deals with the modulation of activity and expression of the hepatic asialoglycoprotein receptor, in pregnant or diethylstilbestrol-treated rats. The results show a two-fold increase in the total cell associated binding activity, both in pregnant and in estrogen-treated animals, with respect to normal values. On the contrary the surface expression was shown to be strongly enhanced only in the liver of pregnant rat. Therefore the modulation shown by this receptor system in pregnancy seems to be only partially estrogen-dependent.

    Notes: Abstract The timing and magnitude of diel migration in two daphnid assemblages were determined from a series of vertical profiles of daphnid size distribution. Animals were collected concurrently for gut fullness determination. Only large daphnids (〉 1.4 mm) migrated, but these animals could account for substantial vertical and diel differences in phosphorus excretion rate. Gut fullness measurements and time courses of diel vertical migration suggested that large Daphnia can cause a net downward flux of phosphorus during summer in thermally stratified lakes.

    Notes: Summary Using a direct conjugate of urokinase and ferritin, the binding has been followed at the plasma membrane and the internalization of urokinase into BALB/C-3T3 fibroblasts, cultured in plasminogen-free conditions. At 0° C, the conjugate was observed bound on both coated and uncoated cell surface regions as singlets, and small and large clusters. No binding was observed in the presence of excess native urokinase. The binding was impaired by preincubation of the conjugate with a competitive inhibitor of the catalytic site, suggesting an interaction between the receptor and the catalytic site of the enzyme. Within 1 min at 37° C, urokinase clustered on coated regions of the plasma membrane. At 5 min after warming, ferritin was found on deeply indented coated pits and in both coated and uncoated vesicles close to the cell surface. By 10 min at 37° C, ferritin particles were present in uncoated endosomes and in multivesicular bodies in the Golgi area. Within 10 min, the receptors on the surface strongly decreased. New receptors were observed on the membrane after 20 min at 37° C. At this time, ferritin was observed both in endosomes or multivesicular bodies and in vesicles close to the plasma membrane.

    Notes: The effect of glucagon and insulin on the incorporation of 1-14C-acetate into cholesterol and fatty acids and on the enzymes involved in the first steps of cholesterol synthesis (3-hydroxy-3-methyl-glutaryl-coenzyme A reductase, 3-hydroxy-3-methyl-glutaryl-coenzyme A synthase, and acetoacetyl-coenzyme A thiolase) was investigated. Isolated rat hepatocytes at different stages of fetal and postnatal development were employed. Data obtained show the appearance of hormonal control on the 18th day of fetal life, indicating the same pattern, as regards acetate incorporation and HMGCoA reductase prepared and assayed in the presence of NaF. On the contrary, HMGCoA reductase, prepared without NaF, HMGCoA synthase, and acetoacetyl CoA thiolase, does not appear to respond to hormonal stimulation. In the perinatal period, the hormonal effect is no longer detectable, probably because of a hormone resistance of this metabolic pathway.

    Notes: The effect of amino acid depletion or supplementation and the effect of glucagon and insulin on the amino acid transport mediated by system A were investigated by determining the uptake of either 2-amino [1-14C]isobutyric acid (AIB) or N-methyl 2-amino [1-14C]isobutyric acid (MeAIB) in rat hepatocytes, freshly isolated at different stages of pre- and postnatal development. The data obtained show that the Na+-dependent uptake was higher at the earliest developmental stages, and steadily decreased until the adult level. The hormones increased AIB and MeAIB uptake enhancing the Vmax, while the Km was unchanged. This effect was evident in cells from adult and 18-20-day-old fetuses, while no response was present before the 18th day of fetal life and in the perinatal period. Actinomycin D or cycloheximide abolished this hormone-dependent increase. A decrease in AIB and MeAIB transport after incubation in an amino acid-rich medium was demonstrated at all ages tested, but was particularly evident in the prenatal life. The increase in the activity of the system following amino acid starvation was shown to be mostly dependent from de novo protein synthesis in the fetal life; on the contrary in the adult the increase appeared to be more linked to the releas from transinhibition of the transport.
