Notizen: Abstract Several small bodies of metabasite (maximum dimensions of 1000 m x 500 m) are included in the metamorphic rocks of the Nevado-Filabride Complex in the Betic Cordilleras (Almeria Region). The body of 400 m x 100 m, located 200 m due west of the Lubrin village, contains troctolitic gabbro with well-preserved igneous textures and mineral compositions, wholly amphibolitized gabbro, garnet-bearing metagabbro eclogite. Along with the textural and mineral changes, sensible and regular geochemical variations can be observed, where the content of MgO decreases from 24% to 11%, while that of CaO and Na2O increases from 7% to 11% and from 2% to 3%, respectively. In addition, the content of some minor elements such as Sr, Y, Nb, Zr and Sc increases while that of Ni and Cr decreases from troctolitic gabbro to the eclogite. The amphibolitized gabbro shows values scattered around those of the troctolitic gabbro. These geochemical variations are ascribed to inherited differences in the pre-metamorphic protolith, i.e. a fractionated gabbro which varies from olivine-rich to clinopyroxene-rich gabbro. Nevertheless, some metasomatism affected the Lubrin body without changing the main chemical trends, as documented by the significantly different 87Sr/86Sr ratios of each rock-type. This points to a metasomatism which involved the introduction of crustal radiogenic strontium. The petrographical and mineral chemical features are interpreted to be the result of syn-metamorphic fluid circulation possibly combined with deformation by shearing. The igneous texture and mineral chemistry have been retained wherever both fluid circulation and shearing were ineffective. On the contrary, where both events were effective, the formation of eclogite occurred. Later, the entire body underwent a retrogressive amphi-bolitic stage under greenschist facies conditions, which was probably responsible for the formation of the amphibolitized gabbro portion and for the retrogression of the eclogite.

Notizen: Abstract. A device for self-separating newly metamorphosed post-larvae prawns from larval culture tanks was designed based on the rheotactic activity exhibited by freshwater prawns after metamorphosis into post-larvae. The performance and effectiveness of the post-larvae selector were evaluated under (a) 24-h periods, (b) four water flows and (c) at two tank depths. A high migratory response (90% confidence level) during the dark period with a water flow range of approximately 11 pm was found. Thus the device has shown to be highly efficient in separating post-larvae in mixed population tanks. The avoidance of excessive handling stress, and the continuous automatic removal of post-larvae immediately following metamorphosis into juveniles are two of the most important advantages of this design as compared to other separation devices.

Notizen: Naegleria fowleri cells, grown axenically, contain high levels of β-D-glucosidase which catalyzes the hydrolysis of 4-methylumbelliferyl-β-D-glucopyranoside (4MUGlc) (Km, 0.9 mM), octyl-β-D-glucoside (Km, 0.17 mM), and p-nitrophenyl-β-D-glucopyranoside at relative rates of 1.00, 2.88, and 1.16, respectively (substrate concentration, 3.0 mM). When the amebae are subjected to freeze-thawing, sonication, and centrifugation (100,000 g, 1 h), 85% of the β-glucosidase activity appears in the supernatant fraction. The β-glucosidase was purified 40-fold (34% yield) using a combination of chromatographic steps involving DE-52 cellulose, concanavalin A-Sepharose, and hydroxylapatite followed by isoelectric focusing. The predominant soluble β-D-galactosidase activity in the Naegleria extract copurifies with the β-D-glucosidase; the two activities have the same isoelectric point (pI, 6.9), similar heat stabilities, are both inhibited by lactobionic acid (Ki, 0.40 mM), and exhibit optima at pH 4.5, indicating that they are probably the same enzyme. The Naegleriaβ-D-glucosidase has an apparent molecular weight of 66,000, a Stokes radius of 25 Å, and a sedimentation coefficient of 4.2S. The β-glucosidase is not inhibited by conduritol β-epoxide or galactosylsphingosine but is completely inhibited by 1.25 mM bromo conduritol β-epoxide. The latter compound, when present in the growth medium, inhibits the growth of the organism and profoundly alters its ultrastructure, the main effect being the apparent inhibition of cytokinesis and the generation of multinucleate cells. The issue of the role of the β-glucosidase in the metabolism of the ameba and its possible role in pathogenic mechanisms are discussed.

Notizen: . Extracts of the pathogenic ameba Naegleria fowleri, prepared by freeze-thawing and sonication, were analyzed for their content of various hydrolytic enzymes that have acid pH optima. The organism is rich in acid phosphatase activity as well as a variety of glycosidases which include β-glucosidase, β-galactosidase, β-fucosidase, α-mannosidase, hexosaminidase, arylsulfatase A, and β-glucuronidase. The crude extract contained only negligible levels of sphingomyelinase, neuraminidase, or arylsulfatase B. All of the hydrolases exhibited higher activity at pH 5.5 than at 7.0, indicating that they are truly “acid” hydrolases. In general, after centrifugation (100,000 g, 1 h), except for arylsulfatase B, more than half of the activity of each of the various hydrolases was recovered in the supernatant fraction. The acid phosphatase in the high-speed supernatant was purified 45-fold (32% yield) by chromatography on QAE-Sephadex and Sephadex G-200 and shown to have the following properties: 1) pH optima, 5.5; 2) Km (4-methylumbelliferyl phosphate), 0.60 mM; 3) molecular weight (estimated by gel filtration chromatography), 92,000; 4) inhibited by heteropolymolybdate complexes but not by L(+) sodium tartrate (0.5 mM) or sodium fluoride (0.5 mM). In addition, unlike the tartrate-resistant acid phosphatase of Leishmania donovani, the major acid phosphatase of N. fowleri is less than 5% as effective in inhibiting superoxide anion production by f-Met-Leu-Phe-stimulated human neutrophils. The finding of high levels of a number of acid hydrolases in Naegleria fowleri raises several questions that merit further study: Do the hydrolases perform a housekeeping function in this single cell eukaryote or do they play some role in the pathogenic process that ensues when the organism infects a suitable host?

Notizen: The cellular bases of the powerful cytolytic activity of the human protozoan parasite Entamoeba histolytica were explored by studying the effect of the virulent strain HM1:IMSS on epithelial monolayers of MDCK cells using a combination of time-lapse microcinematography and transmission and scanning electron microscopy. Early alterations of the epithelial cell membranes were detected by measuring changes in the transepithelial electrical resistance of MOCK monolayers mounted in Ussing chambers. The aggressive mechanism of E. histolytica trophozoites was found to be a complex, multifactorial phenomenon that included hit-and-run damage to the plasma membrane of effector cells mediated through contact, phagocytosis of lysed or apparently intact, but detached, MDCK cells, and inlracellular degradation of ingested cells. Following contact with amebas, the epithelial monolayers showed a pronounced lowering of transepithelial resistance, opening of tight junctions, distortion of microvilli, surface blebbing, and the presence of minute focal discontinuities in the plasma membrane. There was no evidence of amebic exocytosis, membrane fusion, or junction formation between the parasite and host plasma membranes. Although modifications in the epithelial cell membranes usually preceded lysis, the cytolytic activity of the parasite did not exclusively involve damage to the plasma membrane of the cultured host cells but also was mediated by avid phagocytosis, the displacement and separation of neighboring cells by means of pseudopodial activity, and the “pinching-off” of the peripheral cytoplasm of epithelial cells.

Notizen: The author has investigated the role played by project selection methods in defining a firm's technology strategy, as exemplified by a sample of innovative companies in Spain. The information was collected by a combination of questionnaire and interview with key personnel. The author was able to classify strategies into four groups: (1) a planning strategy, essentially a negotiation comprising top-down and bottom-up elements; (2) an economic strategy in which a large number of economic criteria set in advance by top management are used to evaluate projects; (3) a market strategy in which R&D is seen more or less as an adjunct to the Marketing function, which defines the products needed and negotiates the programme with top management; (4) a technical strategy, used in circumstances in which technological innovation is essential and in which, therefore, economic factors take a subsidiary place. The author's data show that in companies operating a planning strategy project selection methods of various, perhaps ad hoc kinds play a key role in reaching a company consensus. For those using an economic strategy the selection criteria are predetermined and selection methods inflexible. In the case of the market strategy, evaluation methods are used solely to help to rank projects prior to selection. Firms employing the technical strategy clearly base their decisions purely on the technical merit of the various projects put up for implementation. The paper includes information on the types of selection method used, broken down by company size and other similar criteria, and comparisons with practice in the USA, France and some other countries.
