    Notes: We have isolated a hybrid gene, composed of the first 455 nucleotides of hisP and nucieotides 275–1107 of malK, the genes coding for the nucleotide-binding components of the high-affinity transport systems for histidine and maltose in Salmonella typhimurium, respectively. The fusion had occurred by recombination within 11 homologous base pairs located between the two DNA fragments. In the chimeric protein peptidic motifs A and B, proposed to be part of the nucleotide-binding fold, originate from HisP and MalK, respectively. Plasmid pES42-39, harbouring the hybrid gene, was shown to complement only a malK mutation but failed to complement a hisP deletion mutation. The chimeric protein was identified by immunoblotting as a protein with an apparent molecular mass of 49kDa. Removal of the C-terminal 77 amino acid residues from the chimeric protein resulted in the loss of function in transport. In contrast, 51 amino acid residues could be removed from the C-terminus of wild-type MalK without any effect. Upon overproduction the chimeric protein, as wild-type MalK, inhibited expression of the malB regulon. However, both truncated proteins, when overproduced, did not exhibit this activity. Based on these results, a tentative model of the functional domains of MalK is presented.

    Notes: In the framework of the European project aimed at the sequencing of the Bacillus subtilis genome the DNA region located between gerB (314°) and sacXV (333°) was assigned to the Institut Pasteur. In this paper we describe the cloning and sequencing of a segment of 97 kb of contiguous DNA. Ninety-two open reading frames were predicted to encode putative proteins among which only forty-two were found to display significant similarities to known proteins present in databanks, e.g. amino acid permeases, proteins involved in cell wall or antibiotic biosynthesis, various regulatory proteins, proteins of several dehydrogenase families and enzymes II of the phosphotransferase system involved in sugar transport. Additional experiments led to the identification of the products of new B. subtilis genes, e.g. galactokinase and an operon involved in thiamine biosynthesis.

    Description / Table of Contents: Zusammenfassung In Wahl- und Zwangsversuchen wurde die Wirkung des Steinkleekrautes (Melilotus officinalis [L.] Pall.) und des Ölgranulates der Patschulipflanze (Pogostemon patchouly Pellet.) auf die KleidermotteTineola bisselliella Hum. untersucht. Patschuliölgranulat zeigte sowohl repellierende als auch abtötende Wirkung. Die Befallsintensität an Wollstoffproben wurde dadurch stark reduziert. Steinkleekraut zeigte nur eine kurzfristig abschreckende Wirkung auf die Falter. Längerfristig führte es dahingegen eher zu verstärktem Befall an den Wollstoffproben.
    Notes: Abstract Free-choice chamber- and compulsory-tests were carried out to investigate the effect of sweet clover leaves (Melilotus officinalis [L.] Pall.) and the granulated oil extract of the patchouli-plant (Pogostemon patchouly Pellet.) on the webbing clothes mothTineola bisselliella Hum. Granular patchouli oil was found to be both, repellent and toxic. Thus the intensity of infestation on woolen fabric samples had been reduced largely. Sweet clover leaves showed only a short-term repellent effect on the adults, whereas in long-term experiments an increase in infestation was found.

    Notes: Abstract General circulation model experiments designed to estimate the magnitude and structure of internally generated variability and to help understand the mechanisms underlying this variability are described. The experiments consist of three multi-century integrations of a rhomboidal 15, 9 level, version of the Center for Ocean-Land-Atmosphere Studies atmospheric general circulation model: a run with fixed sea surface temperatures and equinox solar radiation, a run with seasonally varying climatological sea surface temperatures and seasonally varying solar forcing, and a run with seasonally varying solar forcing in which the state of the ocean is predicted by a 3° by 3°, 16 vertical level, nearly global domain version of the Geophysical Fluid Dynamics Laboratory Modular Ocean Model. No flux correction is used in the coupled model integration. Selected surface fields of the three runs are compared to each other as well as to the observed climate. Statistical properties of variability on interannual time scales are compared between the runs. Evidence is presented that climate time scale variability in the simulations is produced by random weather time scale forcing due to the integrating effect of elements of the system with long memories. The importance of ocean variability for land climate variability is demonstrated and attributed to both the memory effect and coupled atmosphere-ocean instability.

    Notes: Abstract The problem of detection and sizing of fatigue cracks that are closed due to the reverse plastic deformation is addressed in this paper. In particular, it is suggested that the zone of plastic deformation can be identified by measuring the changes in the wave speeds in the plastically deformed material. This is supported through experimental results, first by determining the changes in wave speed with monotonic plastic deformation and then by investigating the plastic zones near fatigue cracks. The characteristic changes in the wave speeds are interpreted in terms of the theory of wave propagation in nonlinearly deformed media and elastic plastic fracture mechanics.

    Notes: Abstract One of the primary obstacles to be overcome for the development of economical fuel cells is the high cost of the membrane electrolyte. The currently favoured polymer electrolytes consist of poly(tetrafluoroethylene) backbone structures and poly(perfluorosulphonic acid) side chains. In an effort to find lower cost membranes, some radiation-grafted copolymer membranes were investigated. All the membranes contained poly(styrenesulphonic acid) side chains. Three different backbone polymer structures were studied: low-density poly(styrene), poly(tetrafluoroethylene)/poly(perfluoropropylene), and poly(tetrafluoroethylene). The results indicate that the membrane consisting of a poly(tetrafluoroethylene)/poly(styrenesulphonic acid) copolymer is a promising candidate as a fuel-cell electrolyte.

    Notes: Abstract We review progress toward developing a data-analysis system using invertebrate species lists in wetland evaluation procedures. Species lists for Mollusca, Carabidae (Coleoptera), Empidoidea and Syrphidae (Diptera), derived from samples collected at six stations along a transect established at a site on the floodplain of the River Loire (France) provide the raw data. An initial analysis using a traditional ordination method (Correspondence Analysis) is carried out. A data base was established for the 118 mollusc, carabid and syrphid species recorded, and each species was then treated not as a simple integer, but as an amalgam of digitised attributes deemed responsive to wetland conditions, in a correspondence analysis on instrumental variables. Despite the generalized nature of the attributes as defined for this example, the approach demonstrates a marked increase in interpretability of the output of analysis, in respect of the relation between the species and the floodplain environment. It also highlights the value of use of data pertaining to more than one taxonomic group and of selecting these taxonomic groups on the basis of complementarity of their ‘bioindicator’ potential.

    Notes: Abstract The development of the Rose Bengal model of cerebral infarction in the rat is monitored using rapid NMR imaging techniques. The lesion volume,T 1,T 2, and the apparent diffusion coefficient (ADC) are measured in the time interval between 24 and 168 h postinduction. VolumetricT 1-weighted 3D imaging of the lesions produced by this model is shown to have a standard error of about 5% over the first 3 days postinduction, rising to about 15% at 1 week. Both theT 1 andT 2 contrasts are found to be more durable than those in ADC and are, hence, more suitable for volumetric monitoring over this time period. An elevation in ADC is also observed in tissue outside the lesion and this is interpreted as being caused by the transport of edema away from the lesion.
