    Description: Silvicultural practices such as thinning and fertilization can affect both canopy foliage quantity and distribution, altering stand growth. The objectives of this research were to quantify the effects of tree size and silvicultural treatment on the vertical distribution of foliage of individual trees of loblolly pine (Pinustaeda L.) and to estimate foliage quantity and distribution using easily measured tree data. In three stands sampled in North and South Carolina, fertilization and (or) thinning treatments had been applied 2 years prior to sampling. A fourth stand was untreated. Nonlinear and linear regression models were developed to test the effects of silvicultural treatment on individual branch foliage biomass and whole tree foliage biomass. Vertical distributions of foliage and branches were modelled using a Weibull probability density function. Analyses indicated that individual branch foliage biomass was positively related to branch size but negatively related to distance from the top of the tree. Fertilization with nitrogen and phosphorus, or thinning, increased the foliage biomass carried by a given sized branch. Silvicultural treatment effects on individual branches translated into whole-tree foliage biomass with thinning and fertilization increasing the crown size of individual trees. Though treatment affected crown size, the distribution of foliage (and branches) remained unaffected. Because silvicultural treatments change the size of crowns for trees of given dimensions, any estimation of loblolly pine crown biomass must be site and treatment specific.

    Description: Mutations of the gene encoding p53, a 53-kilodalton cellular protein, are found frequently in human tumor cells, suggesting a crucial role for this gene in human oncogenesis. To model the stepwise mutation or loss of both p53 alleles during tumorigenesis, a human osteosarcoma cell line, Saos-2, was used that completely lacked endogenous p53. Single copies of exogenous p53 genes were then introduced by infecting cells with recombinant retroviruses containing either point-mutated or wild-type versions of the p53 cDNA sequence. Expression of wild-type p53 suppressed the neoplastic phenotype of Saos-2 cells, whereas expression of mutated p53 conferred a limited growth advantage to cells in the absence of wild-type p53. Wild-type p53 was phenotypically dominant to mutated p53 in a two-allele configuration.     Description: Introduction of a normal retinoblastoma gene (RB) into retinoblastoma cells was previously shown to suppress several aspects of their neoplastic phenotype, including tumorigenicity in nude mice, thereby directly demonstrating a cancer suppression function of RB. To explore the possibility of a similar activity in a common adult tumor, RB expression was examined in three human prostate carcinoma cell lines. One of these, DU145, contained an abnormally small protein translated from an RB messenger RNA transcript that lacked 105 nucleotides encoded by exon 21. To assess the functional consequences of this mutation, normal RB expression was restored in DU145 cells by retrovirus-mediated gene transfer. Cells that maintained stable exogenous RB expression lost their ability to form tumors in nude mice, although their growth rate in culture was apparently unaltered.     Description: Several cellular proteins form stable complexes with the proteins encoded by the adenovirus early region 1A (E1A) gene in extracts derived from adenovirus infected or transformed cells. Two of the cellular proteins that bind to E1A have been identified; one, a 105-kilodalton protein (pRb), is the product of the retinoblastoma gene, and the other, a 60-kilodalton protein, is a human cyclin A. Two other proteins that bind E1A have now been shown to be related to p34cdc2.     Description: Insulin-induced activation of extracellular signal-regulated kinases [ERKs, also known as mitogen-activated protein (MAP) kinases] is mediated by Ras. Insulin activates Ras primarily by increasing the rate of guanine nucleotide-releasing activity. Here, we show that insulin-induced activation of ERKs was enhanced by stable overexpression of growth factor receptor-bound protein 2 (GRB2) but not by overexpression of GRB2 proteins with point mutations in the Src homology 2 and 3 domains. Moreover, a dominant negative form of Ras (with Ser17 substituted with Asn) blocked insulin-induced activation of ERKs in cells that overexpressed GRB2. GRB2 overexpression led to increased formation of a complex between the guanine nucleotide-releasing factor Sos (the product of the mammalian homolog of son of sevenless gene) and GRB2. In response to insulin stimulation, this complex bound to tyrosine-phosphorylated IRS-1 (insulin receptor substrate-1) and Shc.     Description: Observations and modeling of 3- to 6-hertz seismic shear waves trapped within the fault zone of the 1992 Landers earthquake series allow the fine structure and continuity of the zone to be evaluated. The fault, to a depth of at least 12 kilometers, is marked by a zone 100 to 200 meters wide where shear velocity is reduced by 30 to 50 percent. This zone forms a seismic waveguide that extends along the southern 30 kilometers of the Landers rupture surface and ends at the fault bend about 18 kilometers north of the main shock epicenter. Another fault plane waveguide, disconnected from the first, exists along the northern rupture surface. These observations, in conjunction with surface slip, detailed seismicity patterns, and the progression of rupture along the fault, suggest that several simple rupture planes were involved in the Landers earthquake and that the inferred rupture front hesitated or slowed at the location where the rupture jumped from one to the next plane.     Description: The effects of ozone and simulated acid rain on the aboveground nutrient composition of loblolly pine (Pinustaeda L.) seedlings were studied in a 2-year open-top chamber experiment in the Piedmont of North Carolina. Multivariate analysis of variance tests indicated significant ozone response in the 2nd year but no significant simulated acid rain effect in either year. No interaction among treatments was detectable. Seedlings responded to increasing ozone exposures with increasing concentrations of nitrogen, phosphorus, potassium, magnesium, and calcium in the stem, branches, and foliage. The response of increased nutrient concentration was associated with decreased total biomass accumulation. The nutritional responses were most prominent in late season flushes of foliage and are probably related to ozone-induced premature abscission of early-season flushes. High nutrient concentrations in the stem, branches, and late-season flushes of trees exposed to elevated ozone levels suggest retranslocation and slower growth rates resulted in better aboveground nutrient status and lower nutrient demand from the soil.
