    Notes: Abstract The plasma-membrane H+-pump in guard cells generates the driving force for the rapid ion fluxes required for stomatal opening. Since our electrophysio-logical studies revealed a two fold higher pump-current density in guard cells than in mesophyll cells of Vicia faba L. we elucidated the biochemical properties of this proton-translocating ATPase in plasma-membrane vesicles isolated from both cell types. The capability of the H+ —ATPase to create an H+ gradient is maintained in plasma-membrane vesicles derived from purified guard cells via blender maceration, high-pressure homogenization and polymer separation. The H+-pumping activity of these vesicles coincides with the presence of two polypeptides of approx. 100 and 92 kDa which are recognized by a monoclonal antibody raised against the plasma-membrane H+-ATPase from Zea mays L. coleoptiles. Comparison of H+-pumping activities of isolated membranes revealed an approximately two fold higher activity in guard cells than in mesophyll cells with respect to the total membrane protein content. Furthermore, we demonstrated by western blotting that the difference in pump activities resulted from a higher abundance of the electroenzyme per unit membrane protein in guard-cell plasma membranes. We suggest that the high H+-pump capacity is necessary to enable guard cells to respond to sudden changes in the environment by a change in stomatal aperture.

    Notes: Abstract Hemicellulose components from industrial viscose fibre production are characterized by a lower cellulose content than commercial xylan and the presence of a carboxylic acid fraction originating from the alkaline degradation of carbohydrates during the process. This substrate, after neutralization, can be used by Trichoderma reesei RUT C-30 for the production of cellulase-poor xylanases, useful for the pulp and paper industry. The yields of xylanase ranged up to almost 400 units/ml, with a ratio of carboxymethylcellulase/xylanase of less than 0.015. This crude xylanase enzyme mixture was shown to be superior to that obtained on beech-wood xylan when used for bleaching and, particularly, upgrading of hard-wood chemical pulp by selective removal of the xylan components. Biochemical studies indicate that the low cellulase production by T. reesei grown on these waste hemicelluloses is the result of a combination of at least three factors: (a) the comparatively low content of cellulose in these hemicellulosic wastes, (b) the inhibitory action of the carboxylic acid fraction present in the hemicellulosic wastes on growth and sporulation of T. reesei, and (c) the use of a mycelial inoculum that is unable to initiate the attack on the cellulose components within the carbon source.

    Notes: Rhodopsin and cyclic guanosine monophosphat (cGMP)-dependent channel proteins are isolated from the rod outer segment disk membranes of dark-adopted bovine retinae and incorporated in liposomes, prepared by the method of detergent removal dialysis. The ion channel does not lose its transport function (release of Ca2+ ions by injection of cGMP) when incorporated in a liposome. Its activity depends on the degree of protein solubilization and the kind of detergent used. The highest activity is obtained by use of the detergent CHAPS. Shape, size, and size distribution of the liposomes are deduced from elastic and quasi-elastic light scattering, the liposome number density by viscometry, and the photopigment or Ca2+ content by optical absorbance. The liposomes are heterogeneous with respect to size and shape. Small unilamellar liposomes (Rh = 80 nm) and a narrow size distribution (UD = 0.16) are obtained by using the detergent CHAPS. With increasing rhodopsin content per liposome, the hydrodynamic radius Rh increases and at the same time the shape of a liposome converts from a sphere to a prolate ellipsoid. The amount of entrapped Ca2+ per liposome reaches its maximum value when the Rhodopsin nearest-neighbor distance approaches its minimum value. This suggests an intermembrane protein-lipid-protein lattice, which serves as barriere for Ca2+. The influence of temperature or total used Ca2+ content is less profound. Increasing temperature yields slightly smaller liposomes.
