    Description: A bioregenerative life support system designed to address the fundamental requirements of a functioning independent lunar base is presented in full. Issues to be discussed are associated with CELSS weight, volume and cost of operation. The fundamental CELSS component is a small, highly automated module containing plants which photosynthesize and provide the crew with food, water and oxygen. Hydrogen, nitrogen and carbon dioxide will be initially brought in from earth, recycled and their waste products conserved. As the insufficiency of buffers necessitates stringent cybernetic control, a stable state will be maintained by computer control. Through genetic engineering and carbon dioxide, temperature, and nutrient manipulation, plant productivity can be increased, while the area necessary for growth and illumination energy decreased. In addition, photosynthetic efficiency can be enhanced through lamp design, fiber optics and the use of appropriate wavelengths. Crop maintenance will be performed by robotics, as a means of preventing plant ailments.

    Description: Planetary protection (PP) issues for both a comet nucleus sample return (CNSR) mission and a Mars rover sample return (MRSR) mission are discussed, with special attention given to the PP requirements for such missions, the exobiology science objectives for the CNSR and MRSR missions, and a qualitative PP risk assessment for both mission types. A set of contamination control procedures for both missions is presented, which identify procedures for each of the mission phases (i.e., the prelaunch, launch, sample handling, transit vehicle, and earth return). Recommendations for further research and technology development are discussed.

    Description: Of all the other planets in the solar system, Mars remains the most promising for further elucidating concepts about chemical evolution and the origin of life. Exobiological objectives for Mars exploration include: determining the abundance and distribution of the biogenic elements and organic compounds, detecting evidence of an ancient biota on Mars, and determining whether indigenous organisms exist anywhere on the planet. Both approved and planned missions to Mars were evaluated for their potential to contribute to the understanding of these exobiology science objectives and an exploration strategy was developed for each objective.

    Description: This volume includes chapters on exobiology in space, chemical and early biochemical evolution, life without oxygen, potential for chemical evolution in the early environment of Mars, planetary protection issues and sample return missions, and the modulation of biological evolution by astrophysical phenomena. Papers are presented on the results of spaceflight missions, the action of some factors of space medium on the abiogenic synthesis of nucleotides, early peptidic enzymes, microbiology and biochemistry of the methanogenic archaeobacteria, and present-day biogeochemical activities of anaerobic bacteria and their relevance to future exobiological investigations. Consideration is also given to the development of the Alba Patera volcano on Mars, biological nitrogen fixation under primordial Martian partial pressures of dinitrogen, the planetary protection issues in advance of human exploration of Mars, and the difficulty with astronomical explanations of periodic mass extinctions.

    Description: The Workshops on Exobiology in Earth Orbit were held to explore concepts for orbital experiments of exobiological interest and make recommendations on which classes of experiments should be carried out. Various observational and experimental opportunities in Earth orbit are described including those associated with the Space Shuttle laboratories, spacecraft deployed from the Space Shuttle and expendable launch vehicles, the Space Station, and lunar bases. Specific science issues and technology needs are summarized. Finally, a list of recommended experiments in the areas of observational exobiology, cosmic dust collection, and in situ experiments is presented.

    Description: Mussels collected from hydrocarbon seeps in the Gulf of Mexico grow with methane as sole carbon and energy source due to a symbiotic association with methane-oxidizing bacteria. Transmission electron micrographs of mussel gills show cells with stacked intracytoplasmic membranes similar to type I methanotrophic bacteria. Methanotrophs are known to synthesize several types of cyclic triterpenes, hopanoids and methyl sterols, as well as unique monounsaturated fatty acid, double bond positional isomers that serve as biomarkers for this group. Lipid analysis of dissected mussels demonstrated the presence of these biomarkers predominantly in the gill tissue with much smaller amounts in mantle and remaining body tissues. Gill tissue contained 1150 micrograms/g dry wt. of hopanepolyol derivatives and diplopterol while the mantle tissue contained only 17 micrograms/g. The C16 monounsaturated fatty acids (16:1) characteristic of type I methanotrophic membranes dominated the gill tissue making up 53% of the total while only 5% 16:1 was present in the mantle tissue. The methyl sterol distribution was more dispersed. The predominant sterol in all tissues was cholesterol with lesser amounts of other desmethyl and 4-methyl sterols. The gill and mantle tissues contained 3461 micrograms (17% methyl) and 2750 micrograms (5% methyl) sterol per gm dry wt., respectively. Methyl sterol accounted for 44% of the sterol esters isolated from the gill, suggesting active demethylation of the methanotrophic sterols in this tissue. The use of lipid biomarkers could provide an effective means for identifying host-symbiont relationships.

    Description: The mechanism for synthesis of monounsaturated fatty acids under aerobic and anaerobic conditions was studied in the facultative anaerobic cyanobacterium, Oscillatoria limnetica. The hexadecenoic acid (C16:1) of aerobically grown O. limnetica was shown to contain both the delta 7 (79%) and delta 9 (21%) isomers, while the octadecenoic (C18:1) acid was entirely the delta 9 acid. Incorporation of [2-14C] acetate into the fatty acids under aerobic conditions resulted in synthesis of the delta 7 and delta 9 C16:1 and the delta 9 C18:1. Synthesis of unsaturated fatty acids in the presence of DCMU required sulfide. Anaerobic incubations in the presence of DCMU and sulfide (less than 0.003% atmospheric oxygen) resulted in a two-fold increase in monounsaturated fatty acids of both delta 7 and delta 9 C16:1 and delta 9 and delta 11 C18:1. The synthesis of these is characteristic of a bacterial-type, anaerobic pathway.

    Description: A primary scientific theme for the Space Exploration Initiative (SEI) is the search for life, extant or extinct, on Mars. Because of this, concerns have arisen about Planetary Protection (PP), the prevention of biological cross-contamination between Earth and other planets during solar system exploration missions. A recent workshop assessed the necessity for, and impact of, PP requirements on the unmanned and human missions to Mars comprising the SEI. The following ground-rules were adopted: (1) Information needed for assessing PP issues must be obtained during the unmanned precursor mission phase prior to human landings. (2) Returned Mars samples will be considered biologically hazardous until proven otherwise. (3) Deposition of microbes on Mars and exposure of the crew to martian materials are inevitable when humans land. And (4) Human landings are unlikely until it is demonstrated that there is no harmful effect of martian materials on terrestrial life forms. These ground-rules dictated the development of a conservative PP strategy for precursor missions. Key features of the proposed strategy include: to prevent forward-contamination, all orbiters will follow Mars Observer PP procedures for assembly, trajectory, and lifetime. All landers will follow Viking PP procedures for assembly, microbial load reduction, and bio-shield. And, to prevent back-contamination, all sample return missions will have PP requirements which include fail-safe sample sealing, breaking contact chain with the martian surface, and containment and quarantine analysis in Earth-based laboratory. In addition to deliberating on scientific and technical issues, the workshop made several recommendations for dealing with forward and back-contamination concerns from non-scicntific perspectives.
