    Notes: The racemic phthalazinone derivatives azelastine and flezelastine were resolved via formation of diastereomeric salts and fractional crystallization thereof. The optical purity of the enantiomers was checked by HPLC. Pharmacological investigations in vitro and in vivo related to antiallergic/antiasthmatic activity revealed some stereospecific differences. However, chiral discrimination could not be observed with regard to overall activity of azelastine and flezelastine. © 1993 Wiley-Liss, Inc.

    Notes: Synthesis and Pyrolysis of a Triafulvene PrecursorIn view of retro-Diels-Alder reactions (RDA reactions), the triafulvene precursor 3 has been prepared in a simple three-step synthesis by dibromocarbene addition at dibenzo-barrelene (11→12; 44%), halogen-Li exchange followed by methylation (12→14, 100%) and HBr elimination (14→3, 62%) (Scheme 3). Reactivity of the so far unknown bridged 1,1-dibromocyclopropane 12 has been explored, including reductions, allylic rearrangements, and ‘carbene dimerizations’ (Scheme 4). First experiments with respect to the thermal behavior of 3 show that RDA reaction, although occurring in most cases, is not the predominant pathway. When 3 is heated in a sealed tube without solvent, two dimers 26 and 27 are isolated in a total yield of 55% (Scheme 6). On the other hand, gas-phase pyrolysis of 3 at 400° mainly produces rearranged 28 (56%; Scheme 7). It is assumed that bridged trimethylenemethane 29 is an essential intermediate in thermal rearrangements of 3 (Scheme 8).

    Notes: Subcloning of polymerase chain reaction (PCR) fragments is often performed via blunt end ligation after previous Klenow fragment exonuclease treatment. Since insert-specific primers are at hand from the original amplification step, we used a simple PCR-based assay to determine the insert orientation of the recombinants. After purification of enriched plasmids, small aliquots were tested performing standard PCR reactions using a plasmid primer directed towards the cloning site and one of the insert specific primers, respectively. Only the distant insert primer yields a PCR product which can be visualized after gel electrophoresis. In this way both the insert orientation in the vector and the correct size of the cloned fragment is determined rapidly without sequencing or restriction fragment analysis.

    Notes: Road bitumens, when combined with polymers such as styrene-butadiene-styrene (SBS) block copolymers, show improved mechanical properties. The corresponding blends, however, are thermodynamically unstable, and phase separation quickly occurs at higher temperatures. A solution is proposed for improving the thermal stability of these blends: It is based on the formation of reversible, ionic interactions between the bitumen and the SBS, thereby leading to a mixed, ionomeric network. Both compounds are first chemically modified by the grafting of carboxylic acid groups, then mixed, and, finally, the available acid groups are partially or totally neutralized by adding zinc acetate dihydrate to the blends. Comparison of the thermal stability of the resulting ionomeric blends with that of the untreated blends is discussed. The use of ion-ion interactions to enhance miscibility, which, for the first time, has been extended to bitumen-polymer systems, is shown to be a means of obtaining stable blends.

    Description / Table of Contents: Dimethyltin Dithiosquarate: The Crystal Structure of the Benzene Solvate and of the Dimethylsulfoxide Adduct(CH3)2SnS2C4O2 · 1/3 C6H6 (I) crystallizes in the orthorhombic space group Pnma. a = 14.393(2), b = 21.668(3), c = 10.424(1) Å, Z = 12.(CH3)2SnS2C4O2 · (CH3)2SO (II) is monoclinic, space group P21/n, a = 9.918(5), b = 12.028(6), c = 12.223(6) Å, β = 108.82(3)°, Z = 4.In I there are two independent dimethyltin dithiosquarate molecules. But in both molecules the Sn atoms display weak coordinative bonds to two O atoms of adjacent dithiosquarate groups. The distances amount 2.873 Å (2×), resp. 2.678 and 2.831 Å. The coordination number of tin becomes 6 and the structure gets connected in three dimensions.In II dimethylsulfoxide is bound with the O atom to Sn (2.345 Å) and the result is a distorted trigonal bipyramid. There a more distant O atom (2.944 Å) leads to a connection of the molecules to a ribbon. The changes of the geometrical parameters are described, which result from the progressive approach of the one O atom.

Ein weiter entferntes O-Atom (2,944 Å) bewirkt hier die Vernetzung der Moleküle zu einem Band. Die Veränderungen der geometrischen Parameter als Folge der fortschreitenden Annäherung des einen O-Atoms werden beschrieben.

    Notes: The use of a porous glassy carbon (PGC) material as a packed-column SFC stationary phase has been previously demonstrated [1]. The material is further characterized in terms of its retention characteristics. The effects of variations in mobile phase composition, pressure, and temperature conditions are evaluated. Variation of temperature and pressure yielded expected results, specifically, decreased solute capacity factors with increased mobile phase density. The choice of supercritical fluid mobile phase allows the most notable control of solute retention; this was evaluated by adding low percentages of organic modifiers of varying molecular weights to the supercritical carbon dioxide mobile phase. PGC-SFC provides reversed phase characteristics similar to those found for PGC-HPLC. Porous glassy carbon has selectivity characteristics previously unavailable in supercritical fluid chromatography. Use of porous glassy carbon in supercritical fluid chromatography may provide distinct advantages in difficult analytical separations, allowing separations of molecules with only slight structural differences.

    Notes: While using porous glassy carbon (PGC) as a stationary phase for supercritical fluid chromatography (SFC), irreversible modifications of the PGC surface were observed when using 3% formic acid in supercritical carbon dioxide mobile phase at mild operating conditions. Scanning electron microscopic and energy dispersive X-ray spectroscopic analyses indicated the formation of a polymer coating on the PGC surface. The coating may be formed from a polymerization reaction of the formic acid modifier catalyzed by the presence of the carbon surface. Those using carbon-based stationary phases should be aware of this reaction when using formic acid as a modifier for SFC.
