    Notes: A gene for ethionine resistance isolated from the yeast Saccharomyces cerevisiae DKD-5D-H conferred on the yeast cells resistance to seleno-L-methionine and capability to produce S-adenosyl-L-methionine in the cells. An enzymatic study of the L-methionine synthetic pathway of L-methionine proto- and auxotrophs and in dried yeast cells with or without the gene suggested that the cloned gene for ethionine resistance is responsible for the activity of S-adenosyl-L-methionine synthase. To produce S-adenosyl-L-methionine by yeast cells transformed with the ethionine resistance gene, some culturing conditions were determined.

    Notes: Whole cells of Brevibacterium ammoniagenes IAM 1645 having the polyphosphate NAD-kinase were successfully immobilized in a polyacrylamide gel lattice. The immobilized cells were activated by treatment with organic solvents or detergents. The pH optimum of the immobilized cells for the production of NADP was 7.0, and divalent metal ions were required to maintain the elevated activity of polyphosphate NAD-kinase. Highly pure NADP was continuously produced in high yield by the immobilized cell column. The half-life of this column was about eight days.

    Notes: Incativators of cystein proteinases (CPs) were tested as inhibitors of bone resorption in vitro and in vivo. The following four CP inactivators were tested: Ep453, the membrane-permeant produrg of Ep475, a compound with low membrane pereability which inhibits cathepsins B, L, S, H, and calpain; Ep453, the membrane-permeant prodrug of Ep475; CA074, a compound with low membrane permeability which selectivly inactives cathepsin B; and CA07Me, the membrane-permanent prodrug of CA074. The test systems consisted of (1) monitoring the release of radioisotope from prelabelled mousecalvarial explants and (2) assessing the extene of bone resorption in and isolated osteoclast assay using confocal laser microscopy. Ep453, Ep475, and CA074Me inhebited both stimulated and basal bone resorption in vitro while CA074 WASA without effect; The inhibition was reversible and dose dependent. None of the inhibitors affected protein synthesis, DNA synthesis, the PTH-enhanced secretion of β-glucuronised, and N-acetyle-β-glucosaminidase, or the spontaneous release of lactate dehyrogenese. Ep453, Ep475, and CA074Me does-dependently inhibited the resorption activity of isolated areat osteoclassts cultured on bone slices with a maximal effect at 50 μM. The munber of resorption pits and their mean volume was reduced, whilest the mean administration subcutaneously at a dose of 60 μg/g body weight inhibited bone resorption in vivo as measured by an in vivo/in vitro assay, by about 20%. This study demonestration that cathepsins B,L, and/or S are involved in bone resorpotion in vitro and in vivo. Whilest cathepsin L and/or S act extracellularly, and possibly intractually, cathepsin B mediate its effects intracellularly perpheps through the activation of other proteinases involved in subsosteoclastic collagen degradition.

    Notes: Composite films composed of poly(diisopropyl fumarate-co-styrene) (PFS) and nematictype liquid crystal (E8) (40/60 by weight) were prepared by solvent casting in chloroform at room temperature. Scanning electron microscopy measurement showed that a liquid crystal phase is continuously embedded in a spongy-like PFS martrix. Optical responses of the composite films under the conditions of an externally imposed ac electric field (60-240 Vp-p, 10-1000 Hz), temperature (290-355 K), and a film thickness (8-16 μm), were determined using an He-Ne laser (wavelength 632.8 nm). The results obtained indicated that, under the conditions imposed, the output can be continuously controlled, and the response time is only of several milliseconds or less. In addition, the response to temperature sweep, heating and cooling, was reversible with only a delay, and it was interpreted in terms of the rigidity and thermal stability of the matrix polymer. © 1993 John Wiley & Sons, Inc.

    Notes: Positive- and negative-ion production from electrosprayed charged liquid droplets has been studied. For acidified solutions of arginine, Arg-Arg-Arg, Lys-Lys, and (CH3)4NBr in methanol + water (80/20, v/v), the relative intensities of CH3OH2+ ions were found to decrease faster than those of other sample ions as the distance (d) between the capillary tip and the interface plate was increased. The radial distribution of ion intensities in the spray cone was also examined. While the CH3OH2+ ions and multiply charged ions had maximum intensities in the central region of the spray cone, NH4+ and singly charged oligopeptide ions were found to be distributed in its peripheral region. The observed species-selectivity effects in the production of electrospray ions were interpreted in terms of the volatility of ions in the charged liquid droplets. The negative-ion intensities of the oligopeptide Glu-Glu were enhanced greatly when ammonia was added to the solution. An optimum ammonia concentration which gives the strongest negative-ion signals was ∼10-4M.

    Notes: Regular network polyamide films made from trimesic acid (Y) and aliphatic diamines as well as copolyamide films made from hexamethylene diamine (6) and dodecamethylene diamine (12) with Y were prepared. Prepolymers obtained by melt polycondensation for a short period of time were cast from a DMF solution and successively post-polymerized at 300°C to form a network. The resultant films were transparent, flexible and insoluble in any solvents. Heat distortion temperature (Th) measured by a penetration mode of thermomechanical analysis rapidly increased with increasing post-polymerization time, and then levelled out after 0.5-1 h. Th was 238, 231, 232, 216, 207, 187, 176 and 163°C for 4Y, 5Y, 6Y, 7Y, 8Y, 9Y, 10Y and 12Y post-polymerized for 4h, respectively. Degrees of reaction estimated from the infrared absorbance of ester and methylene groups were 88 and 83% for 6Y and 12Y post-polymerized for 4h, respectively. Two diffraction peaks appeared in the wide angle X-ray scattering (WAXS) pattern, suggesting some ordered structure owing to the regular networks. Density increased with increasing post-polymerization time and was 1.266, 1.236, 1.214, 1.175, 1.162, 1.145, 1.130 and 1.107 g/cm3 for 4Y, 5Y, 6Y, 7Y, 8Y, 9Y, 10Y and 12Y post-polymerized for 4h, respectively. WAXS patterns of copolymer films of 12/6-Y suggested the continuous change of network structure with copolymer composition. It was found that comonomer disturbs the formation of ordered network structure, and decreases the packing of networks, thus decreasing density, Th and tensile properties, and increasing dye absorption of copolymer films. From the dye absorption behavior, it was suggested that these regular network films could be utilized as membranes for the separation of molecules by varying the chain length between crosslinked sites.

    Notes: An electrospray ion source designed for a magnetic sector instrument usually has two skimmers, which are located behind a capillary exit with separate pumping stages to reduce the pressure from that of ambient air to a valve suitable for the sector instrument. This work describes an evaluation of these two skimmer voltages with respect to the charge-state distributions and the focusing efficiency of ions. The skimmer voltages were found to be closely related to the m/z values of the detected ions. The optimal difference voltage between skimmers 1 and 2 increased in proportion to the m/z values of the ions, irrespective of the molecular masses of the associated substances. This phenomenon is ascribable to the ion-focusing effect in this region rather than to the effects of energetic collisions in the region of the skimmers, and suggests that the skimmer voltages should be adjusted in accordance with the m/z values of the detected ions for efficient acquisition of all distributed ions. The work demonstrates the functional role of the skimmers in electrospray ionization mass spectrometry with a magnetic sector instrument by examining several types of peptides and proteins, as well as an analysis of a complex mixture of peptides.
