    Notes: The electron ionization mass spectra obtained, on two types of GC/MS instrument, of an impurity peak in a sample of fluorotrichloroethene (1) were those of two different compounds, viz., the epoxide of 1 and trichloroacetyl fluoride. Confirmation of the latter structure was provided by evidence from infrared spectroscopy. A study, on both instruments, of the variation of injector, source and interface temperatures has shown that the sample impurity is actually the epoxide which, at sufficiently high interface-region temperatures, can be converted to the acyl fluoride species. Ab initio molecular orbital calculations have confirmed that the acyl fluoride is more stable than the epoxide.

    Notes: The introduction of more rigorous regulations aimed at controlling the use of sulphonated dyestuffs and dyestuff intermediates has pointed to the need for screening methods for the presence of these involatile compounds. Although much work has been caried out recently on the mass spectrometric behaviour of these compounds, it has been concerned principally with the production of molecular weight information using novel ionization methods rather than with any attempt to obtain structurally diagnostic fragment ions. In this mass spectrometric investigation of a range of naphthalene sulphonic acid salts, positive-ion and negative-ion fast-atom bombardment (FAB) mass spectra have been obtained and FAB-tandem mass spectrometry (MS/MS) has been used to acquire spectra (daugher-ion, parent-ion and neutral-loss) in an attempt to develop a useful screening method. On known compounds, best results were found when using a negative-ion neutral-loss scan to detect the loss of 64 mass units (corresponding to loss of SO2). This was confirmed by successful use of this scan to screen a sample of sulphonate residues.

    Notes: The design and use of an RF-only collision-cell ramp generator is described. The generatio controls the collision energy voltage applied to the RF-only collision cell in a hybrid mass spectrometer of BEqQ geometry. The generator enables the collision-cell voltage to be increased or decreased in a linear ramp, over the full 500 V available. The start/stop functions of the ramp are synchronized under data system control to the scan of the quadrupole mass analyser. The collision-induced dissociation spectra so produced show an increase in the product ion abundance.

    Notes: The results of thin layer chromatography/mass spectrometry (TLC/MS) and thin layer chromatography/tandem mass spectrometry (TLC/MS/MS) experiments are described. Spectra from the analytes were obtained directly from the absorbent without the need for extraction prior to analysis by mass spectrometry. These results illustrate the added specificity that the tandem mass spectrometer can afford in the detection and characterization of a range of compounds separated by TLC.

    Notes: Fragmentation pathways of the synthetic pyrethroid cypermethrin and four structurally related insecticides were investigated using a tandem quadrupole mass spectrometer incorporating a hexapole collision cell under positive-ion electron impact ionization conditions. Conventional mass spectrometry using the first quadrupole analyser only and tandem mass spectrometry on selected precursor ions and product ions, and also constant neutral loss scan experiments, were used. Mechanisms and fragmentation pathways are proposed to explain the inherent stability of ions associated with the benzylphenoxy portion of this class of insecticide.

    Notes: The aim of the present series of experiments was to investigate the effect of the size of follicle from which the oocytes originate on their subsequent in vitro developmental ability. Ovarian follicles were isolated and grouped according to size (2-6 mm, 〉6 mm). Primary oocytes were carefully liberated and grouped according to morphology into one of five categories: denuded; expanded; with two or three layers of cumulus; with four or five layers; and with many (six or more) layers. Following in vitro maturation (IVM), fertilization (IVF), and culture (IVC), more oocytes with many layers of cumulus (P 〈 0.01, 70.2%, 73/104 vs. 46.8%, 87/186, respectively) and a higher proportion of blastocysts were obtained from follicles 〉 6 mm compared to 2-6 mm follicles (P 〈 0.01, 65.9%, 60/91 from 〉6 mm follicles vs. 34.3%, 34/99 from 2-6 mm follicles, respectively). Use of follicular fluid (BFF) from follicles of different sizes in the IVM medium did not significantly increase the cleavage rate or blastocyst yield compared to controls. Administration of procine folliclestimulating hormone (pFSH) to donors prior to slaughter was investigated as a possible means of increasing the number of larger sized follicles in the ovaries and, thereby, the quality of the recovered oocytes. It was found that administration of six injections of pFSH beginning 3 days prior to slaughter resulted in a significant increase (P 〈 0.001) in the proportion of follicles 〉6 mm in diameter (31.6%) compared to that in nontreated controls (6.6%) and to animals that received only four injection groups (9.4%). The blastocyst yield from oocytes originating from 〉6 mm follicles, whether from unstimulated or from pFSH-treated animals, was approximately double that of oocytes from 2-6 mm follicles (P 〈 0.01; 42.9%, 24/56 for 〉6 mm follicles vs. 22.8%, 21/92 for 2-6 mm follicles, respectively, for the 6 pFSH group; P 〉 0.05; 62.5%, 5/8 for 〉6 mm follicles vs. 32.8%, 22/67 for 2-6 mm follicles, respectively, for the control). © 1994 Wiley-Liss, Inc.

    Notes: The mass spectra of biological molecules, whose molecular mass exceeds 10 kDa, invariably contain multiply charged ions. For example, a survey scan of a small protein will produce singly, doubly and triply protonated molecules, the intensity of the doubly charged species often being greater than that of the singly charged entity. Although the spectra resulting from doubly charged peptides have not previously been studied, collisional activation of such doubly charged species may result in significant additional information pertaining to molecular structure. The techniques employed to study ions originating from multiply charged species were linked scanning of constant B/E and tandem mass spectrometry, namely low collision energy spectra acquired on a BEQQ hybrid instrument. The methodology was applied to model compounds whose tandem mass spectrometry characteristics are well known, e.g. gramicidin S and angiotensin I. The results for the product ions of the [M + 2H]2+ species of the models were obtained which highlight the methodology required for high-mass materials.
