    Notes: Abstract The inclusion complexes of 2-naphthyloxyacetic acid (NOA) and 1-naphthylacetic acid (NAA) with β-cyclodextrin have been investigated in aqueous solution. It has been demonstrated that the naphthalene derivatives form 1:1 complexes when included in the cyclodextrin. A possible structure is proposed, having an axial inclusion of the naphthalene derivatives. In the case of the β-CD: NOA complex, the naphthyl moiety is included in the cyclodextrin and the acetic acid group protrudes from the cavity, while NAA is only partially included because of the steric effect of the group in position 1. Association constants of 560±100 M−1 and 100±50 M−1 have been calculated for the β-CD: NOA and β-CD: NAA complexes, making use of the increment in the fluorescene emission produced in the inclusion process.

    Notes: Abstract A polymorphic microsatellite (Y-27H39) based on a (GATA) n repeat was recently discovered on the short arm of the human Y chromosome. We have used a simple technique based on polymerase chain reaction amplification and native polyacrylamide gel electrophoresis followed by highly sensitive silver staining to study the inheritance, the genetic stability and the allele frequency distribution of this polymorphism in the Brazilian population. We have analyzed 100 randomly chosen Caucasian Brazilian father-son pairs with established paternity. Five alleles, four base-pairs apart, were easily distinguishable. Their frequencies were: A (186 bp), 0.19; B (190 bp), 0.49; C (194 bp), 0.24; D (198 bp), 0.07; E (202 bp), 0.01. In all father-son pairs, there was complete allelic concordance. From these data, the probability of discrimination for forensic cases and the average probability of exclusion for paternity cases were both calculated to be 0.66.

    Notes: Abstract Respiration and fermentation were lower in active dry yeast (ADY) rehydrated at 0°C than in ADY rehydrated at 40°C. In agreement with other reports, it was found that membrane permeability increased during rehydration. In addition, ADY rehydrated at 0° did not reseal, even after hours of incubation at 40°C. Using 32P-nuclear magnetic resonance it was found that the cellular concentration of sugar phosphates, phosphate, pyrophosphate, NADH and ATP were lower in ADY rehydrated at 0°C. In addition, the phospholipid peak had a higher height to broadness ratio at 0°C than at 40°C, suggesting that membranes in the 0° sample were more disordered. The lower fermentation rate in ADY rehydrated at 0° could not be due solely to membrane permeation since addition of cofactors that leaked from these cells did not reactivate fermentation. In cell free extracts or in toluenized cells it was observed that some activities were modified after rehydration at 0°C. In the 40°C sample a lower activity of pyruvate decarboxylase and higher fructose-1,6-bisphosphatase and ATPase activities were detected. As a result, higher levels of ADP and pyruvate were found in the cell. Higher ADP levels could contribute to the higher fermentation rate of the cells rehydrated at 40°C. Enzyme modification might explain the low viability of ADY observed by a plating method, even in cells that were impermeable to a vital dye.

    Notes: Abstract The photo-oxidation reaction of phenothiazine has been studied in the presence ofβ-cyclodextrin (β-CD) and 2-hydroxypropylβ-cyclodextrin (HP β-CD). The influence of these organized media on the formation of the oxidation photoproduct upon UV irradiation has been investigated. Phenothiazine forms an inclusion complex with the cyclodextrins. The stoichiometry and formation constant of the complex formed with 2-hydroxypropyl β-CD have been calculated using the changes of the fluorescence emission signal and of the absorbance of the drug upon inclusion. An increase of the fluorescence intensity of the photogenerated product is attained when it becomes included inside the cyclodextrin cavity.

    Notes: Abstract The spectral characteristics of the surfactant, Triton X-100, in the absence and presence ofβ-cyclodextrin have been examined. The fluorescence of Triton X-100 is concentration dependent and is markedly enhanced in the presence ofβ-cyclodextrin. Analysis of the variations in the excitation-emission profiles of the surfactant with concentration suggests excimer emission at concentrations above the critical micelle concentration (CMC). Nuclear magnetic resonance (NMR) spectroscopy suggests that the phenyl group is included inside the CD cavity while a portion of the ethylene oxide chain extends outside the cavity. Benesi-Hildebrand type equations were derived to determine the stoichiometry and to estimate the formation constant of the CD: Sf complex.

    Notes: Abstract A model ointment consisting of white petrolatum, mineral oil and microcrystalline wax was studied using rheology, microscopy and thermal techniques. Rheograms studied over a temperature range of 25-40°C indicated that the overall rheogram shape changed little as the temperature increased. However, two inflections gradually disappeared as the temperature increased. Thermal optical analysis showed that the temperature range over which these inflections disappeared correlated with the melting transition of the components forming the secondary structure. Another series of rheograms obtained from ointments with different combinations of the raw materials showed the rheology of the ointment is primarily controlled by the white petrolatum and mineral oil and that the microcrystalline wax acts to build-up the structure by incorporating itself into the existing white petrolatum structure. Thermal optical analysis of comelts of the raw materials proved that the ointment network structure is essentially a recombination of the naturally occurring components in differing ratios. The knowledge obtained from these studies is applied to a discussion of the thermal and mechanical stresses encountered in the filling operation.

    Notes: Abstract Coffee plants were regenerated from protoplasts isolated from embryogenic cell suspension cultures derived from somatic embryos of Coffea arabica L. cv. caturra. Yields of viable protoplasts ranged from 1×105 to 6×105 protoplast/g fresh weight. Protoplast preparations usually contained no contaminating cells, and when present, the number of cells never exceeded 0.1% of the total. Plating efficiencies of protoplast ranged from 1 to 10%. Embryogenic protocolonies obtained after several subcultures in a medium supplemented with 0.5 mg/l each of benzylaminopurine, 2,4-dichlorophenoxyacetic acid and naphtaleneacetic acid, were transferred to a medium lacking plant growth regulators. Well differentiated embryos were formed in selected protocolonies that contained many embryos-like structures. Approximately 70% of the somatic embryos developed into green rooted plantlets which were succesfully transferred to vessels containing sterilized scoria. Plants grown for two months in scoria were finally transferred to greenhouse.
