    Notes: Abstract A description is given of a simple Monte Carlo processor for the simulation of generalized cellular automata systems in statistical physics and certain simpleimage processing tasks. Xilinx SRAM based FPGAs are used to implement the main functional blocks of the processor, with the consequence that it can be reconfigured at the gate level to tackle a wider variety of simulations than a non-FPGA based special purpose architecture. For a wide class of simulations the processor can increase the processing speed of its PC-AT host by more than two orders of magnitude.

    Notes: Summary We have developed a liquid fermentation medium for the submerged culture of the fungus,Zalerion arboricola, which supports the rapid production of an echinocandin-type antibiotic, pneumocandin A0 (formerly L-671, 329), in yields increased at least 4-fold over those reported previously. The improvements were achieved through medium simplification, substitution of high levels of mannitol for glycerol as the major source of carbon, and restriction of available magnesium. Antibiotic formation in batch cultures with this mannitol-based medium is not confined to the idiophase; rather production appears to be biphasic, with synthesis beginning during growth (i.e., at day 3) and increasing in rate at day 11, well after rapid growth has ended. Accumulation of antibiotic continues beyond 14 days, and by 21 days titers greater than 500 μg/ml are attained. For the synthesis of a related compound, pneumocandin B0, by a mutant strain ofZ. arboricola, the medium gives similar production kinetics and a titer of 800 μg/ml. Although supplementation of the medium with magnesium ions stimulates growth, it decreases titer by preferentially affecting the second phase of antibiotic synthesis. This decline in synthesis in the magnesium-supplemented medium is explained by the depletion of mannitol before the second phase of synthesis can begin. In contrast, mannitol in the magnesium-limited medium is used more slowly with approximately half still available at day 11 to support continued antibiotic formation.

    Notes: Summary The regiospecific glucosylation of FK 506 and immunomycin (FR 900520) at the 24-hydroxy position was performed using resting cells ofBacillus subtilis ATCC 55060. 24-Glucopyranosyl FK 506 and 24-glucopyranosyl immunomycin were isolated by methylene chloride extraction and purification using reverse phase HPLC. The metabolite structures were established using spectroscopic techniques including MS and NMR. The glucose conjugate was further confirmed by chemical degradation. Enzymatic glucosylation was demonstrated using cell-free extracts derived fromBacillus subtilis ATCC 55060. The 24-glucosyltransferase, which appears UDP-glucose dependent, was solubilized from cell membranes by treatment with 0.1% Nonidet P-40 detergent. The optimal conditions for assay of the enzyme have been determined.

    Notes: Abstract The effects of urinary chloride and nitrogen concentration and osmotic pressure on the nitrification of ammonium in a calcareous soil treated with cow urine were examined. Urinary chloride concentrations of up to 7.4 g L−1 had no effect on the rate of nitrification, as determined by the accumulation of soil nitrate. Osmotic stress, generated using a mixed salt solution, had an inhibitory effect on nitrification at soil osmotic pressures lower than or equal to −1.0 PMa. Nitrification was completely inhibited at a soil osmotic pressure of −2.6 MPa. Accumulation of nitrate after a lag phase of 18 days was noted in the −2.0 MPa soil osmotic pressure treatment, indicating some degree of adaptation or osmo-regulation within the nitrifying population at this stress level. High urine-N concentrations resulted in considerable nitrite accumulations and reduced nitrification activity through the effect of free ammonia. It is concluded that in most temperate grassland soils at near-neutral pH, urinary chloride and nitrogen are unlikely to reduce nitrification rates, except where urine-N concentrations exceed 16 g N L−1. Inhibition due to osmotic stress will be directly related to soil moisture status and may be particularly severe in dry, light-textured soils.

    Notes: Abstract According to the traditional account Mendel's paper on pea hybrids reported a study of inheritance and its laws. Hence, Mendel came to be known as “The Father of Genetics”. This paper demonstrates that, in fact, Mendel's objective in his research was finding the empirical laws which describe the formation of hybrids and the development of their offspring over several generations. Having found these laws (and not the laws of inheritance that he is generally credited with) he proposed a theoretical scheme involving the formation of germinal and pollen cells in hybrids and their combination in fertilization competent to explain why his laws took the form that they did. Mendel's research shows a pattern of development closely paralleling the stages of empirical investigation beginning at the level of qualitative description in common language rising through four levels of increasing abstraction and culminating in a fifth level, his simple mechanical theory. This mechanism met all the tests that a theory of this type must pass. In that respect Mendel was highly successful and this success might be the reason why he did not push his theory to a higher level, a sixth level involving the use of particulate determiners; despite this fact, Mendel was credited with having taken exactly that step.

    Notes: Abstract Sandeels, especiallyAmmodytes marinus are a major component of the diet of many predatory fish, seabirds and seals. The industrial fishery for sandeels is now the largest of the North Sea fisheries. A sandeel fishery in the Shetland area began in 1974 but has recently declined. This change was accompanied by dramatic declines in the breeding success of certain seabirds, particularly Arctic terns and kittiwakes. Current information on seabirds and sandeels in the Shetland area is reviewed and areas where further research is needed, highlighted. The Shetland problem illustrates the difficulties of reconciling conservation and exploitation when fundamental ecological and behavioural knowledge is lacking, and also the need to obtain further information on the ecological impact of industrial fisheries.

    Notes: Abstract A 15N labelling technique was used to measure N2O and N2 emissions from an undisturbed grassland soil treated with cow urine and held at 30 cm water tension and 20°C in a laboratory. Large emissions of dinitrogen were detected immediately following urine application to pasture. These coincided with a rapid and large increase in soil water-soluble carbon levels, some of this increase being attributed to solubilization of soil organic matter by high pH and ammonia concentrations. Emissions of nitrous oxide generally increased with time in contrast to dinitrogen fluxes which decreased as time progressed. Estimated losses of N2O and N2 over a 30 day period were between 1 to 5% and 30 to 65% of the urine N applied plus N mineralized from soil organic matter, respectively. Most of the N2 and N2O originated from denitrification with nitrification-denitrification being of minor significance as a source of N2O. Comparisons of the 15N enrichments in the soil mineral N pools and the evolved N2O suggested that much of the N2O was produced in the 5–8 cm zone of the soil. It is concluded that established grassland soils contain large amounts of readily-oxidizable organic carbon which may be used by soil denitrifying organisms when nitrate is non-limiting and soil redox potential is lowered due to high rates of biological activity and high soil moisture contents. ei]{gnR}{fnMerckx}
