    Notes: Abstract  We analyzed a functional homologue of the swallow gene from Drosophila pseudoobscura. The swallow gene of D. melanogaster plays an essential role in localizing bicoid mRNA in oocytes, and swallow mutant embryos show anterior pattern defects that result from the lack of localization of the bicoid morphogen. The pseudoobscura homologue rescues the function of swallow mutants when introduced into the genome of D. melanogaster, and its expression is similar to that of the melanogaster gene. The predicted pseudoobscura and melanogaster proteins are 49% identical and 69% conserved. The coiled-coil domain previously identified in the melanogaster swallow protein is strongly conserved in the pseudoobscura homologue, but the weak similarity of the melanogaster swallow protein to the RNP class of RNA-binding proteins is not conserved in the pseudoobscura homologue. These and other observations suggest a structural role for swallow in localizing bicoid mRNA, perhaps as part of the egg cytoskeleton.

    Notes: Abstract  Sinkhole collapse is one of the main limitations on the development of karst areas, especially where bedrock is covered by unconsolidated material. Studies of sinkhole formation have shown that sinkholes are likely to develop in cutter (enlarged joint) zones as a result of subterranean erosion by flowing groundwater. Because of the irregular distribution of pinnacles and cutters on the bedrock surface, uncertainties arise when "hit-or-miss" borehole drilling is used to locate potential collapse sites. A high-resolution geophysical technique capable of depicting the details of the bedrock surface is essential for guiding the drilling program. Dipole-dipole electrical resistivity tomography (ERT) was used to map the bedrock surface at a site in southern Indiana where limestone is covered by about 9 m of clayey soils. Forty-nine transects were conducted over an area of approximately 42,037 m2. The electrode spacing was 3 m. The length of the transects varied from 81 to 249 m. The tomographs were interpreted with the aid of soil borings. The repeatability of ERT was evaluated by comparing the rock surface elevations interpreted from pairs of transects where they crossed each other. The average difference was 2.4 m, with a maximum of 10 m. The discrepancy between interpreted bedrock-surface elevations for a transect intersection may be caused by variations in the subsurface geology normal to the transect. Averaging the elevation data interpreted from different transects improved the ERT results. A bedrock surface map was generated using only the averaged elevation data at the transect junctions. The accuracy of the map was further evaluated using data from four exploratory boreholes. The average difference between interpreted and actual bedrock surface-elevations was less than 0.4 m. The map shows two large troughs in the limestone surface: one coinciding with an existing sinkhole basin, while the other is in alignment with a small topographic valley. Because sinkholes were observed at the same elevation interval in similar valleys in the vicinity, the delineated trough may have implications for future land use at the site.

    Notes: Summary In a survey of 528 unrelated haemophilia A patients, six partial deletions of the factor VIII (FVIII) gene were detected by Southern blotting. These deletions were further mapped by a combination of Southern blotting and polymerase chain reaction amplification and found to vary in length between 4.7kb and 57kb. The frequency of detectable FVIII gene deletions (about 1%) is thus considerably lower than previously reported. Statistical analysis of currently available data did not provide any evidence for a deletion “hotspot”. Four of the six deletion patients reported here possessed inhibitors. Taken together with previous data, deletion of the FVIII gene was found to be associated with an approximately fivefold higher risk of developing inhibitors compared with other severe haemophiliacs without gene deletions.

    Notes: Abstract Two derivatives of plasmid pAT153 which arose spontaneously in phosphate-limited continuous cultures of Escherichia coli HB101[pAT153] have been characterised. The smaller plasmid, pLAB446, resulted from a deletion of 446 base pairs of DNA covering the translation start and codon sequence of the tetracycline resistance gene. The other plasmid, pLAB135, differed from pAT153 only by an A:T to G:C transition in the transcribed but untranslated leader region of the tet gene which decreased tet transcription by 84%, possibly due to the formation of a more stable stem-loop structure in the mRNA.

    Notes: Increasing restrictions on sewage sludge disposal routes and the tightening of treatment requirements have led to an escalation of sludge-disposal costs which are out of proportion to other sewage-treatment costs: consequently reducing sludge volume is becoming increasingly cost-effective. Consolidation of anaerobically digested sludge has traditionally been difficult and takes many weeks in secondary digesters or deep storage tanks. Pre-aeration of the sludge using the Aercon process can enable consolidation to take place in days instead of weeks.This paper reports the results of a pilot-scale plant designed to investigate further the mechanism by which aeration speeds up consolidation. It was concluded that the rate of consolidation depended on aeration intensity and cooling of the sludge. Inhibition of anaerobiosis was more important than gas stripping in enhancing consolidation.

    Notes: Biological damage to plants is commonly found from the Devonian but occurs most commonly from the Cretaceous. Damage inflicted on plants whilst they were alive may trigger a pathological response involving the growth of abnormal tissues. Much of the damage is caused by arthropods, particularly insects. Whilst some damage is non-taxon specific, such as simple feeding traces, other damage, such as leaf mines, galls or bark boring, may reveal the co-evolution of host specific taxa and the timing of such interactions. Damaged plants, particularly from the Cretaceous and Tertiary are described and illustrated. The geological history of the evolution of insect-related plant damage is briefly reviewed. Increased variety in the pathological response of plants is seen from the Cretaceous with the evolution of the angiosperms and diversification of numerous insect groups.

    Notes: The ratios of stable isotopes 18O/16O and 13C/12C, in sagittal otolith carbonate from two tropical demersal teleosts, red emperor Lutjanus sebae and Rankin cod Epinephelis multinotatus, from several locations in northern Western Australia, differed between sites. On a broad scale, fish from the four locations, Shark Bay, Ningaloo, Pilbara, and Broome had stable isotope values that were sufficiently different to indicate separate stocks, and it is appropriate to manage these populations of the two species independently in these areas. On a smaller scale, there may be limited mixing of these species between the Pilbara trawl fishery and the trap and line fisheries operating out of Onslow and Broome. Values of stable oxygen isotopes were strongly related to sea surface temperature, although there were some sites in shallow water where low values of stable oxygen isotopes indicated that fish were living in warm water. The use of stable oxygen and carbon isotope values is a valuable, cost effective method of determining the degree of mixing of fish stocks.

    Notes: The angular variation of the median destructive field (MDF) of magnetite particles has been determined by subjecting anisotropic samples carrying an isothermal remanent magnetization (IRM) to static AF demagnetization, with the AF at an angle to the IRM. The alignment was produced by dispersing the magnetite particles in resin in the presence of a magnetic field thus making the sample anisotropic. After tumble demagnetization, an IRM in 80 mT was given to each sample along the alignment axis. 11 different particle sizes were used covering the range 0.7–82 μm. The results for all the samples are remarkably similar in that the MDF is smallest when the AF is applied parallel to the alignment axis and greatest when the AF is perpendicular to the axis. The results are also very similar to those found experimentally for single-domain ‘tape’ particles and indicate that the reversal mechanism even in the smallest particles is not by coherent reversal as in the Stoner-Wohlfarth model. The angular variation of the MDF for all the particle sizes is described quite well by a function derived by Aharoni for single-domain particles. The MDF at θ= 0 is found to be inversely proportional to the square root of the particle size.
