    Notizen: Abstract Hemolysis is a major symptom of diseased eels infected by Edwardsiella tarda. The hemolysin gene of E. tarda strain ET16 was cloned into plasmid pSK and expressed in Escherichia coli. The mol. mass of the functional β-hemolysin was estimated to be approximately 34 kDa by gel filtration and by SDS-PAGE followed by in situ hemolysin activity analysis. The cloned fragment containing the β-hemolysin locus from E. tarda strain ET16 expressed in E. coli was estimated to be 5.3 kb in length; the deduced gene product was identical in mol. mass and properties to the extracellular products of E. tarda strain ET16. The presence of EcoRI and XbaI sites within the β-hemolysin gene of E. tarda was determined from the loss of hemolytic activity in subclones. Analysis of the DNA sequence of a 2,436-bp HaeIII-HindIII fragment that included EcoRI and XbaI sites revealed three ORFs organized as an operon that encoded three predicted polypeptides of 15,874, 7,055, and 34,804 Da. A 34-kDa polypeptide expressing hemolytic activity in cell lysates of the clone DH5α(pETH3E) is very likely the β-hemolysin encoded by the third ORF. The observation that hemolytic activity appeared in the culture medium of E. tarda, but not in that of E. coli strain DH5α(pETH3E) indicates the existence of a mechanism for transporting the hemolysin across the cell envelope in E. tarda that is different from that of E. coli.

    Notizen: Abstract The effect of load on the tribological behaviour of three different two-dimensional carbon-carbon composites, including polyacrylonitrile (PAN) fibre-pitch matrix, PAN fibre-chemical vapour infiltrated (CVI) matrix, and pitch fibre-resin/CVI hybrid matrix composites has been compared. Results indicated that friction and wear rate variations with slide distance depended on load and the type of composite. The worn surface morphology was categorized into three types (I, II and III). The friction coefficient, wear rate and temperature all increased sharply when the transition from type I to type II occurred. When the powdery type II debris was compacted to form the lubricative type III debris, the friction coefficient and wear rate declined, although never approached their initial type I levels. For all composites, a higher load can accelerate the transition from type I to type II, but impedes the transition from type II to type III. Under 2.4 MPa, the type II morphology was never observed to transform into type III morphology in this study.

    Notizen: Abstract Dysrhythmia in gastric myoelectrical activity has been frequently observed in patients with gastric motor disorders and gastrointestinal symptoms. The assessment of the regularity of gastric myoelectrical activity is of great clinical significance. The aim of this study was to develop an automated assessment method for the regularity of gastric myoelectrical activity from the surface electrogastrogram (EGG). The method proposed in this paper was based on the filter bank and neural network. First, the EGG signal was divided into frequency subbands using filter bank analysis. Second, a parameter called the subband energy ratio (SER) was computed for each subband signal. A multilayer perceptron neural network was then used to automatically classify the EGG signal into four categories: bradygastria, normal, tachygastria, and arrhythmia, using the SER as the input. The EGG recording was made using the standard method of electrogastrography by placing electrodes on the abdominal surface. The study was performed in 40 patients with various gastric motor disorders, ten healthy adults, and ten healthy children. The neural network was trained and tested using the EGG data obtained from the patients. The regularity of gastric myoelectrical activity was assessed based on the classification of the minute-by-minute EGG segments. Using the running spectral analysis method as a gold standard, the proposed automated method had an accuracy of 100% for the training set and 97% for the test set. It was concluded that the proposed method provides an accurate and automatic assessment of the regularity of gastric myoelectrical activity from the EGG. © 1999 Biomedical Engineering Society.     Notizen: Abstract The iron contents in the hair and blood samples of 37 juvenile athletes who were supplemented with 0, 8 and 16 mg Fe/day, respectively, in the food of ferrous gluconatecontaining chocolate for 3 months were determined before and after the supplementation by INAA, SRXRF and blood analysis. The experimental results showed that after supplementation of the iron-fortified food, the normal ferritin level in the blood of the male athletes was attained and the iron content in the hair was increased with supplementation, but both are not in the positive proportion. Most of the female athletes had similar results. It is suggested that supplementation of 8 mg iron/day to juvenile athletes may be desirable.

    Notizen: Abstract Spent ion-exchange resins are produced in the purification of coolant and moderator systems during the normal operation of CANDU (Canada deuterium uranium) nuclear reactors. Carbon-14 is a radionuclide of concern in disposal of ion-exchange resins because of its relatively long half-life, its potential high mobility and its ability to be easily incorporated into organisms. Only limited data are presently available on the14C concentrations of spent from CANDU reactors. To establish a more comprehensive datahase for this radionuclide, concentrations of14C were determined for two moderator resins from Bruce Nuclear Generating Station A. Mixed bed resins were separated into anion and cation fractions using a sugar solution, and the14C concentrations were determined for each fraction. Carbon-14-was located predominantly on the anion beads. Samples of anion resin were found to undergo an 81% loss in the14C concentration over a period of 160 d following the sugar separation procedure. Some evidence is given to suggest this loss in14C may result from microbial activity. Concentrations and distributions of other predominant radionuclides, such as60Co and153Gd, are discussed as well.

    Notizen: Abstract The electrogastrogram (EGG) is a surface measurement of gastric myoelectrical activity. The normal frequency of gastric myoelectrical activity in humans is 3 cycles/min. Abnormal frequencies in gastric myoelectrical activity have been found to be associated with functional disorders of the stomach. The aim of this article was, therefore, to develop new time-frequency analysis methods for the detection of gastric dysrhythmia from the EGG. A concept of overcomplete signal representation was used. Two algorithms were proposed for the optimization of the overcomplete signal representation. One was a fast algorithm of matching pursuit and the other was based on an evolutionary program. Computer simulations were performed to compare the performance of the proposed methods in comparison with existing time-frequency analysis methods. It was found that the proposed algorithms provide higher frequency resolution than the short time Fourier transform and Wigner-Ville distribution methods. The practical application of the developed methods to the EGG is also presented. It was concluded that these methods are well suited for the time-frequency analysis of the EGG and may also be applicable to the time-frequency analysis of other biomedical signals. © 1998 Biomedical Engineering Society.     Notizen: Abstract A study has been carried out to determine the levels of radioisotopes in five Canadian grains from pre-(1985) and post-Chernobyl (1986) crop years. In addition, the radioisotope levels were determined in grains from the 1988 crop year consisting of wheats, oats, barley, corn and oil seeds which included flax, sunflower and canola. The grain and oil-seed samples were ashed to reduce their volume, and the gammaemitters137Cs and40K were determined using a high-purity germanium detector and a high-purity germanium well detector. The90Sr activity measurements required further ashing of the samples followed by chemical separations and β-counting of the separated daughter90Y. The results for the 1985, 1986 and 1988 crop years are discussed and compared with results from grain crops obtained during years of atmospheric bomb tests in the 1950's.
