    Notes: Poly(L-methionine) (PMet) is one of the two sulfur containing polyamino acids. Raman, FTIR spectra, and heat capacity measurements of PMet have been well interpreted through the normal mode analysis and the density of states derived therefrom. Earlier interpretation of heat capacity data is limited because it is based on the Tarasov model, wherein the concept of group frequency and skeletal similarity are used. A special feature of some dispersion curves is their tendency to bunch in the neighborhood of the helix angle. This has been attributed to the presence of strong intramolecular interactions.     Notes: Abstract Objective: To evaluate the effect of grapefruit juice on the pharmacokinetics of artemether in plasma and saliva after a single oral dose and to detect concentration-dependent electrocardiographic changes (bradycardia and QTc prolongation). Methods: Six healthy male subjects were given a standard breakfast followed by two tablets of 50-mg artemether administered with water; 1 week later, the tablets were administered with 350 ml double-strength fresh frozen grapefruit juice. For 8 h, 17 blood- and saliva samples were collected, and 17 electrocardiograms were recorded. Drug and metabolite concentrations were measured by means of high-performance liquid chromatography with electrochemical detection. The pharmacokinetic parameters were determined using a one-compartment model. Results: Grapefruit juice significantly (P = 0.001) increased the mean peak concentration (Cmax) of artemether more then twofold from 42 (SD 17) ng/ml to 107 (28) ng/ml. The time to reach Cmax (tmax) with grapefruit juice was 2.1 (0.6) h compared with 3.6 (17) h with water (P = 0.02). The area under the concentration–time curve (AUC) almost doubled with grapefruit juice from 177 ng · h/ml to 336 ng.h/ml (P = 0.003). The elimination half-life remained unchanged (1.0 h vs 1.3 h). No major changes in the kinetics of the metabolite dihydroartemisinin were detected. Low artemether levels and zero dihydroartemisinin levels were found in saliva. No influences of artemether were observed on 17 electrocardiograms during the 8 h after drug intake – in particular there were no signs of bradycardia or QTc prolongation. Conclusion: Grapefruit juice significantly increases the oral bioavailability of artemether without an effect on the elimination half-life. It suggests a role for intestinal CYP3A4 in the presystemic metabolism of artemether.

    Notes: Cdr1p, a multidrug transporter from a pathogenic yeast Candida albicans, confers resistance to several unrelated drugs including anti-Candida drugs. We demonstrate that Cdr1p can specifically transport human steroid hormones namely β-estradiol and corticosterone. Saccharomyces cerevisiae transformant S-12, harbouring the CDR1 gene, accumulated about 3-fold less [3H]β-estradiol and about 2-fold less [3H]corticosterone than the non-transformed strain. When CDR1 was expressed in AD strain (AD-CDR1) which had seven ATP binding cassette (ABC) superfamily of putative transporter genes disrupted, the net accumulation of these hormones as compared to S-12 was significantly lower. Efflux of β-estradiol and corticosterone was inhibited by a 100-fold higher (200 nM) concentration of β-estradiol, corticosterone, ergosterol or dexamethasone, but progesterone which could not be transported by Cdr1p did not affect the efflux and thus accumulation. Interestingly, some of the drugs viz. cycloheximide, chloramphenicol, fluconazole and o-phenanthroline, to which CDR1 confers resistance, could also prevent efflux and enhance accumulation to some extent. In conclusion, we show that human steroid hormones could be the substrates for Cdr1p and the energy dependent transport mediated by it is specific for estradiol and corticosterone.

    Notes: Reactions to eight isolates of Karnal bunt, Neovossia indica, collected from seven different locations in northern India were studied on 13 host lines, including cultivars and breeding lines of Triticum aestivum, Triticum durum and Triticosecale in all possible combinations. The incidence of Karnal bunt varied from zero in PBW 34 and PBW 248 with isolates Ni8 and Ni2, respectively, to as high as 66.8% in a highly susceptible cultivar WL 711 with isolate Ni5. The differences in disease incidence among cultivars and isolates were highly significant. All the isolates could be distinguished on the basis of differential reactions on one or more of the host lines. Even the most resistant lines of durum (PDW 215), triticale (TL 1210) and wheat (HD 29) could be distinguished by the differential disease reaction with one or more of the eight isolates. The cultivar-isolate interaction for disease score was highly significant, indicating the probable existence of a gene-for-gene relationship in this host-pathogen system.

    Notes: A system, based on the principle of magnetic induction coupling, to track a particle nonintrusively in three dimensions is presented. It consists of small transmitters mounted inside the particle being tracked, and a set of receiving antennae surrounding the experimental apparatus. In Part I of the sequence of two papers, the focus is on the theoretical aspects, in particular, on developing a computational technique to solve the inverse problem of finding the three-dimensional position as well as orientation of the particle from the voltages induced in the antennae. In Part II the focus is on the actual system development, including all hardware and data acquisition aspects. The results presented here are comprehensive, as they include details of hardware/electronics, comparison of induced voltage model and actual measurements, and a set of actual tracking results. Through test experiments that include a variety of real-time trajectories, this system is tested and its success is demonstrated. Results of real trajectories of a single ball rising in a mass of other spheres in a vibrated bed are also shown. Although this system is intended for use in experimental studies of dry granular flows, it has wide applicability due to its nonintrusive nature, and is particularly useful when optical tracking techniques are not feasible. © 1998 American Institute of Physics.
