    Notes: Abstract. Volatile organic compounds (VOCs) are an important source of contamination of groundwater supplies in Massachusetts and many parts of the United States. One local response is to require sewering in wellhead protection areas as an easily enforceable policy designed to reduce the probability of VOC contamination of groundwater. Data were collected for 238 wellhead protection areas in Massachusetts on VOC contamination levels and the sewered and unsewered land uses in those aquifer recharge areas. Logistic regression procedures were used to see whether sewering had any statistical effect on likelihood of contamination of well water. The results provided limited, but not overpowering, support for the idea that requiring commercial and industrial land uses to use sewers would reduce the chance of VOC contamination.

    Notes: Abstract We have compared the sequences of a major class of kinetoplast DNA (kDNA) minicircle (pLURkE3) of Leishmania strain UR6 with other minicircle sequences from different Leishmaniaspecies. Alignment of these sequences allowed the selection of a pair of oligonucleotides suitable as primers in polymerase chain reaction (PCR) which is specific for Leishmaniaparasites. PCR with this genus-specific primer set is capable of detecting 1 femtogram of kDNA. These primers have been tested with kDNAs from both old world and new world Leishmaniaspecies. The results indicate that the primers may be suitable for detection of any kind of leishmaniasis.

    Notes: The residence times of multiple particles during the flow of a non-Newtonian suspension through a conventional holding tube assembly were compared with that in a helical holding tube assembly of identical length and tube diameter. The residence times were measured for different combinations of process parameters (viscosity, flow rate of fluid, size and concentration of particles). The effects of these parameters on Residence Time Distribution (RTD) of particles was determined and dimensionless equations were developed to predict the mean, minimum and standard deviation of residence times. The RTDs in the helical holding tube were found to be narrower than in the conventional holding tube. The RTD of particles at all combinations of process parameters were normally distributed. The ratio of mean to minimum residence time was within 1.05 to 1.11 in the helical holding tube and 1.06 to 1.16 in the conventional holding tube for the range of parameters studied.

    Notes: Residence times of nearly neutrally buoyant polystyrene particles flowing with an aqueous solution of carboxymethylcellulose were measured in each of 4 straight sections of holding tubes that were connected by 180° bends. Equations were developed to predict mean, minimum, and standard deviations of residence times in individual tubes and in the entire assembly as a function of viscosity, particle size, particle concentration, and flow rate. The residence time distribution in the 4th tube was always narrower than that in the 1st tube, and the ratio of the mean to minimum residence time was always between 1.07 and 1.20. These results can help in the design of appropriate holding tubes for food processing.

    Notes: Corn starch was extruded with 4% stearic acid in Brabender single screw and twin screw laboratory extruders. The screws were cooled and removed and samples were collected from locations along the screws. In the single screw extruder, starch gelatinization as determined by DSC and polarized light microscopy started in the compression zone and was completed in the metering zone. Starch-stearic acid complex formation as determined by iodine binding capacity and X-ray diffraction started at the same point as gelatinization and increased to a maximum near the tip of the screw. In the twin screw extruder, starch gelatinization and starch-stearic acid complex formation started at a distance of seven screw diameters and continued from there to the die end.

    Notes: It is demonstrated that matrix-like spectroscopy may be carried out in the gas phase using molecular beams of clusters and the conditions under which bulk-matrix-like behavior is achieved are illustrated. At the same time, we obtain information on the structural evolution of noble gas clusters as a function of their size. Infrared spectra for SF6 attached to noble gas clusters of argon, krypton, and xenon were recorded using a free jet cluster source and a laser photofragmentation detection technique. When a dilute mixture of the chromophore in Ar and Kr is expanded at relatively low pressures, the clusters spectra show a feature characteristic of the SF6 solvated in a defective, unannealed matrix. This feature disappears at higher source pressures (larger sizes) at which the chromophore prefers to reside on the surface of the cluster. This can be demonstrated by producing neat clusters and depositing the chromophore on them. However, on producing still larger clusters, a different absorption appears which is accurately located at the same position as the main absorption in a well-annealed matrix of Ar or Kr. This behavior is related to the transition of clusters from a Mackay icosahedral structure, shown to be the most stable for smaller clusters, to the face-centered-cubic (fcc) structure which is observed in the bulk phase. This structural transition occurs at a nozzle stagnation pressure which corresponds to an average cluster size of about 2000 atoms for both Ar and Kr. Scattering studies performed on argon clusters suggest that the fcc-type clusters correspond to the largest sizes in the cluster size distribution present in the beams. A similar structural transition for xenon cluster was not established as the SF6 appears to solvate only slightly in Xe in the size range studied here. © 1995 American Institute of Physics.

    Notes: Abstract Synthetic oligonucleotide analogs have greatly aided our understanding of several biochemical processes. Efficient solid-phase and enzyme-assisted synthetic methods and the availability of modified base analogs have added to the utility of such oligonucleotides. In this review, we discuss the applications of synthetic oligonucleotides that contain backbone, base, and sugar modifications to investigate the mechanism and stereochemical aspects of biochemical reactions. We also discuss interference mapping of nucleic acid-protein interactions; spectroscopic analysis of biochemical reactions and nucleic acid structures; and nucleic acid cross-linking studies. The automation of oligonucleotide synthesis, the development of versatile phosphoramidite reagents, and efficient scale-up have expanded the application of modified oligonucleotides to diverse areas of fundamental and applied biological research. Numerous reports have covered oligonucleotides for which modifications have been made of the phosphodiester backbone, of the purine and pyrimidine heterocyclic bases, and of the sugar moiety; these modifications serve as structural and mechanistic probes. In this chapter, we review the range, scope, and practical utility of such chemically modified oligonucleotides. Because of space limitations, we discuss only those oligonucleotides that contain phosphate and phosphate analogs as internucleotidic linkages.
