    Notes: Abstract  The National Institute for Environmental Studies (NIES) recently prepared two candidate certified reference materials (CRMs) for arsenicals to meet the growing demand for the quality assurance of arsenic speciation analysis. The NIES candidate CRM No. 14 Brown Alga was prepared from Hijiki seaweed for the certification of inorganic arsenic content, and No. 15 Scallop was prepared from adductor muscle of scallop for the certification of arsenobetaine content. The preparation of the candidate CRMs is briefly described. Cooperative analyses for total arsenic content of the candidate CRMs have been underway. The preliminary speciation analysis at NIES revealed difficulty in establishing suitable conditions for extracting arsenic species from the materials. Chromatograms of arsenic species by a high performance liquid chromatography-inductively coupled plasma mass spectrometric detection system are presented to provide information about arsenic species present in these candidate CRMs.

    Notes: Abstract Element analysis of the annual increments in a longitudinal section from the tusk of a female dugong Dugong dugon (Müller) from Exmouth, Western Australia, was carried out by X-ray fluorescence-imaging, inductively coupled plasma-atomic emission spectrometry (ICP-AES) and inductively coupled plasma-mass spectrometry (ICP-MS). Nine elements (Ba, Ca, Fe, Li, Mg, Na, P, Sr and Zn) were present in concentrations amenable to determination by these techniques. Most elements revealed both long-term trends and shorter, year-to-year, variations. For example, sodium concentrations increased from 0.55 to 0.72% with the age of the dugong. Strontium concentrations (mean 0.17%) were closely correlated (r = 0.86) with those of barium (mean 4.5 mg kg−1). Zinc concentrations increased from ∼70 to 170 mg kg−1 with dugong age, but also showed shorter-term fluctuations of ∼30 mg kg−1 that were correlated (r = 0.41) with mean annual Fremantle sea level (a measure of the Southern Oscillation Index and strength of the Leeuwin Current). The concentrations of the elements and correlations with year and between pairs of elements are discussed.

    Notes: A compact storage ring has been developed for industrial research such as x-ray lithography and material analysis. This machine is of a racetrack type with two superconducting bending magnets and only two normal conducting quadrupole magnets. The circumference is as short as 9.2 m. One quadrupole magnet per cell contributes to making the smaller machine. The injector is a synchrotron, and a full energy injection of 600 MeV is performed. The bending magnets excite a field of 3.5 T, and are operated in a persistent current mode. A decrease in a coil current is ΔI/I〈3×10−3/year. The helium consumption is as low as 2 l/h for two magnets. An iron shield of the magnet decreases a leakage flux to a terrestrial level at a point 3 m from the magnet. A beam current of 380 mA has been stored with no beam instability in spite of there being no correction of the chromaticities. Beam emittances were obtained from measured beam sizes and were in good agreement with calculated values. The coupling coefficient εy/εx is calculated as around 0.04. © 1995 American Institute of Physics.

    Notes: The 18O/16O and 13C/12C ratios in the otolith carbonate of pink snapper Pagrus auratus and tailor (bluefish) Pomatomus saltatrix, from several locations along the Western Australian coast, indicated that pink snapper stocks are location specific but that tailor stocks are less so. The hypersaline Shark Bay, on the coast of Western Australia, generated strongly characteristic isotopic signatures which serve as natural tags. Otolith carbonate from pink snapper from normal oceanic waters just north of Shark Bay showed no evidence that the fish had been in hypersaline water. Similarly, pink snapper from the hypersaline bay showed no evidence of having spent time at normal oceanic salinity. By contrast, some tailor from oceanic waters showed evidence of having spent considerable time in the bay, and some fish from the bay had oceanic signatures. This suggested that tailor were more migratory than snapper. The similarity in the distribution of the isotopic signatures (from oceanic to hypersaline) in otolith carbonate from tailor from oceanic waters north of Shark Bay (Koks Island), and from those within Shark Bay, indicated a single stock in this region (in contrast to pink snapper). Moreover, tailor from coastal south-western Australia and from the Shark Bay area could be considered seperately for some management purposes. For pink snapper stocks from oceanic waters, oxygen isotope signatures were clearly related to water temperature although the temperature relationship was obscured for fish within Shark Bay because of the strength of the signal generated by the hypersaline water. For tailor the temperature relationship was not obvious, probably because migrations of tailor smeared the temperature effect, and the hypersaline Shark Bay waters dominated, and, possibly, at the southern extemity of the range, the freshwater in some estuaries influenced the isotopic signatures of the otolith carbonate. Strontium/calcium ratios in pink snapper otoliths also indicated a separation of stocks, but for tailor overlap of signatures again suggested migratory behaviour.

    Notes: Abstract From a plasmid carrying the tnpA gene and one inverted repeat sequence (IR) of transposon Tn3, plasmids containing a structure characteristic of transposons, i.e., two IRs flanking a tnpA gene, were generated spontaneously in vivo. They appear to have arisen either through the formation of a “staggered” head-to-head dimer or by so-called one-ended transposition. These putative transposons could indeed transpose to, or form cointegrates with, a recipient plasmid. Based on these findings it is proposed that a primeval transposase gene and its target site evolved first, and subsequently gave rise to a “fully-fledged” transposon by head-to-head dimerization or one-ended transposition.

    Notes: Abstract The present research aimed to establish conditions for synchronized plantlet regeneration from rice callus based on a quantitative analysis of the relationship between intracellular 2,4-dichlorophenoxyacetic acid (2,4-D) concentration and shoot regeneration rate. To prepare the rice calli with different intracellular 2,4-D concentrations prior to regeneration, callus precultures were carried out in medium containing 4 mg/l 2,4-D and in 2,4-D-free medium for predetermined periods. As the critical intracellular 2,4-D concentration of the calli precultured in 2,4-D-free medium was too low to analyze precisely by conventional analytical methods, it was estimated using a kinetic model which described the behavior of 2,4-D by taking its uptake, metabolism and/or inactivation rates during the callus preculture into consideration. An experimental relationship between intracellular 2,4-D concentration and regeneration rate of rice calli revealed that the intracellular 2,4-D concentration should be controlled as low as 2.6×10–2μg/g fresh weight to reach the same synchronization in shoot regeneration as seen with rice seed germination. This condition was realized by feeding sugar into the 2,4-D-free medium after 4 days preculture when the carbon source was exhausted.

    Notes: Abstract Fluorine containing-cyclohexadienes (FCHDs), 3,3,6,6,-tetrafluoro-1,4-cyclohexadiene (1a), 1-chloro-3,3,6,6-tetrafluoro-1,4-cyclohexadiene (3a) and 1-bromo-3,3,6,6-tetrafluoro-1,4-cyclohexadiene (4a), were isolated in pure states with high yields (ca. 80%) by the preparative scale electrochemical fluorination of 1,4-difluorobenzene (1), 1-chloro-2,5-difluorobenzene (3) and 1-bromo-2,5-difluorobenzene (4), respectively, in Et4NF · mHF (Et = C2H5: m = 4.0, 4.45 and 4.7) at a platinum anode, followed by extraction and fractional distillation. The electrochemical fluorination of 1,2,4-trifluorobenzene (2) yielded both 1,3,3,6,6-pentafluoro-1,4-cyclohexadiene (2a) and 2,5,5,6,6-pentafluoro-1,3-cyclohexadiene (2b) with high yield (ca. 90%). Some of (2a) was also isolated in a pure state by fractional distillation. Each substrate compound (1–4) was fluorinated to the corresponding FCHDs with an almost quantitative yield when 2.0–2.1 faraday (per mol of substrate) of electricity was passed.

    Notes: Abstract Tension tests were conducted in air at room temperature on PMMA sheet specimens which had been previously soaked in a 40 vol % acetonitrile aqueous solution at 20 °C for 24 h and then dried in air at room temperature for 480 h. In contrast with an untreated specimen which fractured at a stress of 84 MPa and a strain of 9 %, shear yielding clearly took place at 42 MPa and the elongational fracture strain increased to about 148 %. No crazes were observed on the specimen surface and as a result the transparency of the PMMA was thoroughly maintained until fracture. Thus this soaking treatment may change PMMA to a completely ductile polymer without a crazing mechanism. The results of the dynamic viscoelastic measurements at 1 Hz show that the glass transition temperature was lowered to about 80 °C (as compared to about 110 °C), and the β relaxation became much sharper with a higher peak value of 20 °C (as compared to a broad curve with a peak at 50 °C). This clear β relaxation at room temperature may contribute to shear yielding and large plastic elongation of the treated PMMA.

    Notes: Abstract Charge and discharge behaviour of LiNiO2 positive electrode has been examined in organic electrolytes with different compositions. The discharge capacity of the oxide electrode at a moderate cycling rate varied with the electrolyte composition. Redox responses in potential sweep voltammetry also depended on the kind of lithium salt and solvent of the electrolyte. Electrolyte solutions of relatively low ionic conductivity generally gave lower voltammetric responses and rate capability. A.c. impedance analysis of the LiNiO2 electrode before and after the charge and discharge demonstrated that the surface chemistry of the oxide, depending on the electrolyte composition, has an important influence on the rate capability of the oxide electrode in organic electrolyte solutions.
