    Notes: Abstract We have investigated the effects of the glucocorticoid, dexamethasone, and five structural analogs on Drosophila development in an effort to identify steroid ligands that may play a role in the embryogenesis of this organism. Embryos were exposed to glucocorticoids either by direct culture in supplemented medium, or by examining embryos from adult flies raised on supplemented fly food. After exposure, embryos were examined for developmental defects. At a morphological level, exposure to dexamethasone disrupts the dorsolateral folding of the amnioserosa during germ band extension. In addition, germ band retraction and dorsal closure is also disrupted. The phenocritical period of these effects is within the first 4 h of embryogenesis. This response is dosage sensitive, with embryos responding to concentrations of dexamethasone ranging from 10−6 to 10−3M. Furthermore, glucocorticoids which are closely related structural analogs of dexamethasone also disrupt germ band retraction and dorsal closure, while other tested steroids had no effect on embryonic development. At a molecular level, expression of the gene, Krüppel, is absent from the amnioserosa of dexamethasone-treated embryos. The cuticular phenocopy resulting from exposure to dexamethasone and related glucocorticoids is morphologically similar to the mutant phenotype associated with four genes required for germ band retraction, namely hindsight, serpent, tail-up and u-shaped. The results of this study represent the first association of a glucocorticoid with dose, stage and tissue specific effects on Drosophila development at both morphological and molecular levels.

    Notes: Abstract The possibility of observing pure nuclear resonant scattering of synchrotron radiation by a multilayer structure containing the 169Tm isotope is analyzed theoretically. The main problem is the need to suppress the enormous background of radiation scattered by electrons. Two methods for the destructive interference of a synchrotron radiation beam in reflection at grazing incidence by a layered system containing Tm nuclei in one of the layers are considered, and their efficiency as applied to the conditions of third-generation synchrotron radiation sources, such as in the European Synchrotron Radiation Facility (ESRF), is calculated. An electron scattering suppression efficiency parameter is formulated as the ratio of the integrated nuclear scattering intensity (with a time delay) to the total prompt electron scattering intensity in assigned ranges of angles and energies. In the first method thin films of a special type on a substrate, viz., GIAR films, are used. In the second method a new effect, which is termed the Bragg antipeak effect and involves the destructive interference of a wave that is Bragg-diffracted in a multilayer superlattice and a wave reflected on the upper boundary of the sample, is employed. The physical properties of the Bragg antipeak effect are considered, and it is found that its efficiency is sufficient for practical use.

    Notes: Pseudomonas aeruginosa, an opportunistic pathogen, can cause life threatening infections in patients compromised by underlying respiratory disease like bronchiectasis, cystic fibrosis and diffuse panbronchiolitis. Most strains of P. aeruginosa produce some kind of protease with broad substrate specificities during the infectious state in the host. P. aeruginosa elastase, one of the strongest exotoxins, has a tissue-damaging proteolytic activity and is capable of degrading such plasma proteins as immunoglobulins, complement factor and cytokines. The present study focused on the effect of P. aeruginosa elastase and was designed to evaluate the neutrophil accumulation at the inflammation site mediated by P. aeruginosa elastase in the inflammatory response in the host. An air pouch model in rats, considered as a useful model of inflammation, was used to analyze the number of leukocytes, the volume of exudate and the concentration of interleukin-8 after the injection of P. aeruginosa elastase into the pouch cavity. The number of neutrophils and the volume of exudate in the pouch cavity increased significantly at 4 h, peaked at 8 h in a dose-dependent manner and then decreased at 24 h. The concentration of interleukine-8 in pouch fluid peaked 4 h earlier than the peak of the neutrophil number. The enzymatic activity of P. aeruginosa elastase seemed to reinforce the inflammation process. The influence of lipopolysaccharide contamination was negligible. Although these observations were made in the subcutaneous cavity, they indicate that P. aeruginosa elastase plays a role as an immunoprovocative factor in the inflammatory response in cases of infection with P. aeruginosa.

    Notes: Abstract A non-decay type fast-setting calcium phosphate cement (nd-FSCPC) has been described, which did not decay but set within approximately 5–6 min even when the paste was immersed in serum immediately after mixing, and which forms hydroxyapatite as its end product. nd-FSCPC was produced by adding sodium alginate to the liquid phase of the base cement FSCPC. Sodium alginate forms a water-insoluble gel, and reduces the process of fluid penetration into the paste which is the cause of decay. The aim of this investigation was to confirm the mechanism of the non-decaying behaviour of nd-FSCPC proposed in a previous paper, using another chemical with properties similar to those of sodium alginate. Also, it was intended to further improve both the mechanical properties and tissue response of nd-FSCPC. Chitosan, which also forms a water-insoluble gel in the presence of calcium ions and has been reported to have pharmacologically beneficial effects on osteoconductivity, was added to the liquid phase of the base cement FSCPC. The cement thus prepared showed behaviour similar to that of nd-FSCPC using sodium alginate. The cement paste did not decay but set within approximately 5–6 min even when immersed in serum immediately after mixing. DTS value of the set mass was approximately 3–4 MPa, slightly lower than that of nd-FSCPC using sodium alginate, and no inhibitory effect was observed for the transformation of cement component to apatite within the range used in this investigation (up to 1.5%). Therefore, it was concluded that the mechanism of non-decaying behaviour was, at least in part, reduction of fluid penetration into the cement paste. nd-FSCPC using chitosan showed slightly poorer mechanical properties than that using sodium alginate. However, pharmacological effects such as osteoconductivity could be expected in nd-FSCPC using chitosan. Thus, this cement may be useful as a more sophisticated bioactive cement than nd-FSCPC using sodium alginate.

    Notes: Abstract To compare the surface properties of calcium-ion (Ca2+)-implanted titanium with those of titanium and to investigate the mechanism of bone conductivity of Ca2+-implanted titanium, amounts of hydroxyl radical of Ca2+-implanted titanium and titanium were estimated. Also, the point of zero charge (p.z.c.) of oxide constituting surface oxides of Ca2+-implanted titanium and titanium was determined. Results showed that the amount of active hydroxyl radical on Ca2+-implanted titanium was found to be significantly larger than that on titanium, indicating that the number of electric-charging sites of Ca2+-implanted titanium in electrolyte is more than that of titanium. The p.z.c. values of rutile (TiO2), anatase (TiO2), and perovskite (CaTiO3), were estimated to be 4.6, 5.9, and 8.1, respectively. Thus, Ca2+-implanted titanium surface is charged more positively in bioliquid than titanium, accelerating the adsorption of phosphate ions. © 1998 Chapman & Hall

    Notes: Abstract Cellular foams were made from the aqueous solution of agar by freeze-drying. A narrow range (5–20°C min-1) of freezing rate was required to avoid damage to the microstructure of the agar foams. The size of cells in the foam decreased with increasing freezing rate. Agar foams of more than 4 wt% agar content absorbed more energy than a polystyrene foam in compression tests. Foams with a higher agar content absorbed more energy. The behaviour of agar foams in compression tests could be explained by the modified beam theory for cellular foams. Agar foams were thermally stable up to 200°C, and were also stable in a humid environment.

    Notes: Abstract Plasma and liver contents of malonic aldehyde are studied one day after administration of bromobenzene to mice pretreated with a polymeric form of zinc-metallothionein from rat liver. It is found that zinc-metallothionein injected in a dose of 1–4 mg/kg 5–10 min prior to injection of bromobenzene (2 g/kg, about 56% of LD50) markedly lowers the malonic dialdehyde level and active toxicity of this xenobiotic. Administration of a mixture modeling Zn-metallothionein (albumin, cysteine, and zinc) in a dose of 4 mg/kg has no appreciable effect on the malonic dialdehyde level raised after bromobenzene injection, and does not change its LD50. It is concluded that the protective effect of exogenous zinc-metallothionein is due to its antioxidant activity, which allows for normalization of lipid peroxidation.
