    Notes: Abstract Since the molecular identification of the first aquaporin in 1992, the number of proteins known to belong to this family has been rapidly increasing. These members may be separated into two subgroups based on gene structure, sequence homology, and function. Regulation of the water permeability of the collecting ducts of the kidney is essential for urinary concentration. Aquaporin-2 and -3, which are representative of these subgroups, are colocalized in the collecting ducts. Understanding these subgroups will elucidate the differences between aquaporin-2 and -3. Aquaporin-2 is a vasopressin-regulated water channel located in the apical membrane, and aquaporin-3 is a constitutive water channel located in the basolateral membrane. In contrast to aquaporin-3, which appears to be less well regulated, many studies have now identified multiple regulational mechanisms at the gene, protein, and cell levels for aquaporin-2, thus reflecting its physiological importance. Evidence of the participation of aquaporin-2 in the pathophysiology of water-balance disorders is accumulating.

    Notes: Nb/Al–AlOx/Nb tunnel junctions designed for x-ray detection have been manufactured and characterized at temperatures down to 0.35 K. At a bias voltage of 0.5 mV, a leakage current of the order of 0.1 pA/μm2, and a dynamic resistance exceeding 106 times the normal state resistance, were achieved in junctions with dimensions up to 200×200 μm2 and critical current densities of order 200 A/cm2. The combination of high critical current density, low leakage current, and large area represents an important step towards the development of superconducting tunnel junction detectors with improved spectroscopic performance. © 1996 American Institute of Physics.

    Notes: Abstract The third-order nonlinear optical properties of the sol-gel derived Pb(Fe1/2Nb1/2)O3-PbTiO3 (PFN-PT) thin films have been investigated by the THG method. The χ(3) values of PFN-PT thin films increased from 3.2×10−12 esu (PT) to 8.5×10−12 esu (PFN) with increasing content of Fe3+ ions which possessed very high second-hyperpolarizability, ψ(Fe2/3O). It was experimentally confirmed that complex oxides such as ABO3-type perovskites consisting of large non-transition metal A-site cations and small transition metal B-site cations exhibited high χ(3) as expected from Lines' model. It was also found that the χ(3) values of complex oxides could be estimated from the second-hyperpolarizabilities of the constituent single oxides reported so far. Because of the large remanent polarization, PT thin films may exhibit the highest χ(3) among the ferroelectric PFN-PT thin films studied in the present work. The maximum dielectric constant, ε, of 1990 at room temperature was obtained for 0.5PFN·0.5PT thin films, whose composition corresponds to so-called morphotropic phase boundary (MPB).

    Notes: Abstract Cell volume reduction is one of the most distinct morphological changes during spermiogenesis and may be largely attributable to water efflux from the cell. A strong candidate for a water efflux route, aquaporin 7 (AQP7), which is a water channel, was studied immunohistochemically in the rat testis. Immunoreactivity was restricted within the elongated spermatids, testicular spermatozoa, and residual bodies remaining in the seminiferous epithelium. Weak but distinct immunoreactivity was first observed in the cytoplasmic mass of the spermatid at step 8 of spermiogenesis. The Golgi-like apparatus became steadily immunoreactive at step 10. The plasma membrane covering the cytoplasmic mass showed strong immunoreactivity after step 16. At this step, the middle piece of the tail also showed immunoreactivity at the portion protruding into the lumen. The whole head and distal tail, where the elongated spermatid had only a limited amount of cytoplasm, showed no immunoreactivity throughout spermiogenesis. After spermiation, the immunoreactivity of AQP7 remained at the middle piece and in the cytoplasmic droplet in the testicular spermatozoon. The present observations suggest that AQP7 contributes to the volume reduction of spermatids, since this water channel protein is localized on the plasma membrane covering the condensing cytoplasmic mass of the elongated spermatid, and since the seminiferous tubule fluid is hypertonic.

    Notes: Summary We attempted to amplify a specific region of mitochondrial DNA (mtDNA) using the polymerase chain reaction (PCR) from fewer than ten mitochondria isolated individually by microdissection or use of an optical tweezer. We selected preliminarily isolated mitochondria fromPhysarum polycephalum as the model materials and tried to amplify the mtDNA region corresponding to the specific mitochondrial plasmid of this true slime mould. For separation of a few mitochondria from the mitochondrial population, we initially used a destruction method in which excluded mitochondria were disrupted by a UV laser. However, mtDNA was still amplified, although weakly, from mitochondria that had been destroyed by the UV laser. Therefore, we used an optical tweezer to trap individual mitochondria and separate them from the others. The required number of mitochondria were separated from the mitochondrial suspension through a narrow canal of isolation buffer and used directly for PCR amplification. The results showed that the mtDNA could be amplified from at least 9 mitochondria trapped by the optical tweezer.

    Notes: All dental root implants come in contact with the oral epithelium, and many complex factors are found to arise in this region. In order to perform a successful dental root implantation, it is necessary to clarify the interaction of the dental root implant material with the host defense mechanisms involved in the specific and nonspecific immune responses to many antigens in oral bacteria and their components. Recently, focusing on developing the dental root implant, the Nikon Corporation improved the surface characteristics of pure titanium even further by developing a hydroxyapatite (HA) layer formed on an anodic titanium oxide film containing Ca and P via hydrothermal treatment (SA treatment). However, since little is known about the effect of SA-treated pure titanium (HA/Ti) on the defense mechanisms of the oral membrane epithelium, we investigated (1) the in vitro proliferation of murine splenic B lymphocytes on the surface of HA/Ti in the presence of three lipopolysaccharide (LPS) concentrations and (2) interleukin-1α (IL-1α) production by the reaction of human peripheral blood mononuclear cells (PBM cells) on the surface of HA/Ti under the same concentrations. After culture, murine splenic lymphocytes were measured by uptake of 3H-thymidine, and cytokine release (IL-1α) from PBM cells was measured by ELISA. Results showed that HA/Ti had hardly any effect on the LPS-induced proliferation of B lymphocytes and IL-1α production. In vitro investigations of the effects of HA/Ti on the LPS-induced proliferation of murine splenic B lymphocytes and IL-1α from PBM cells might be a useful way of elucidating the defense mechanism between implants and the oral epithelium. © 1998 John Wiley & Sons, Inc.
