    Notes: Abstract The classical disruption method for yeast genes is by using in vitro deletion of the gene of interest, or of a part of it, with restriction enzymes. We are now routinely using a strategy that takes advantage of polymerase chain reactions (PCRs) which amplify large pieces of DNA. Since this approach results in a complete, precise deletion of the open reading frame, which is replaced by a unique restriction site, the ligated PCR can be used for the insertion of different markers of for two-step gene disruptions without an inserted marker. As we have now used this strategy for the deletion of more than ten genes we have in this report included some hints based on our experience.

    Notes: Summary. Each invariant set of a given dynamical system is part of the global attractor. Therefore the global attractor contains all the potentially interesting dynamics, and, in particular, it contains every (global) unstable manifold. For this reason it is of interest to have an algorithm which allows to approximate the global attractor numerically. In this article we develop such an algorithm using a subdivision technique. We prove convergence of this method in a very general setting, and, moreover, we describe the qualitative convergence behavior in the presence of a hyperbolic structure. The algorithm can successfully be applied to dynamical systems of moderate dimension, and we illustrate this fact by several numerical examples.

    Notes: We report full quantum close-coupling and coupled-states calculations of cross sections and rate constants for the rotational relaxation of NH(c 1Π) in collisions with Ar, based on a new ab initio description of the potential energy surface for this system. Single-collision, state-to-state thermal rate constants were extracted from measured fluorescence intensities of relaxing NH(c 1Π,v=0,J) distributions in the presence of Ar starting with singly populated rotational states. Overall, the calculated and experimental rate constants agree well, although the dependence on the initial J is somewhat different. At high J the relaxation is dominated by J→J−1e/f changing transitions. This process is facilitated by approach on the more attractive ArNH(c)A' PES followed by a curve crossing to the more repulsive A‘ PES which correlates asymptotically to the next lower rotational level. For approach in a "helicopterlike'' orientation, this process results in Jf→J−1,e transitions, while for approach in a "pinwheellike'' orientation, this process results in Je→J−1,f transitions. Thus, similar to what we found earlier for the relaxation of high rotational levels of the CH(X) radical [M. H. Alexander and P. J. Dagdigian, J. Chem. Phys. 101, 7468 (1994)], a strong v, J correlation in the reactant channel results in a strong Λ, J correlation in the product channel. A full kinetic simulation, based on the calculated cross sections, reproduces nearly quantitatively the experimental observations of the pressure dependence of the equilibration of NH(c) Λ doublet populations monitored some years ago by Quinton and Simons. © 1995 American Institute of Physics.

    Notes: Synthesis of trehalose in the yeast Saccharomyces cerevisiae is catalysed by the trehalose-6-phosphate (Tre6P) synthase/phosphatase complex, which is composed of at least three different subunits encoded by the genes TPS1, TPS2, and TSL1. Previous studies indicated that Tps1 and Tps2 carry the catalytic activities of trehalose synthesis, namely Tre6P synthase (Tps1) and Tre6P phosphatase (Tps2), while Tsl1 was suggested to have regulatory functions. In this study two different approaches have been used to clarify the molecular composition of the trehalose synthase complex as well as the functional role of its potential subunits. Two-hybrid analyses of the in vivo interactions of Tps1, Tps2, Tsl1, and Tps3, a protein with high homology to Tsl1, revealed that both Tsl1 and Tps3 can interact with Tps1 and Tps2; the latter two proteins also interact with each other. In addition, trehalose metabolism upon heat shock was analysed in a set of 16 isogenic yeast strains carrying deletions of TPS1, TPS2, TSL1, and TPS3 in all possible combinations. These results not only confirm the previously suggested roles for Tps1 and Tps2, but also provide, for the first time, evidence that Tsl1 and Tps3 may share a common function with respect to regulation and/or structural stabilization of the Tre6P synthase/phosphatase complex in exponentially growing, heat-shocked cells.

    Notes: We present a comprehensive set of state-to-state rate constants for the relaxation of isolated rotational levels of NH and ND in their (c 1Π,v=0, J) states in the presence of colliding He and Ar. The rotational states were populated in pulsed photolysis experiments by excitation on single (c,v′=0, J′←a,v″=1, J″) transitions. Fluorescence spectra of the (c,v′=0→a,v″=0) transitions were recorded to determine the relative populations caused by inelastic collisions. The master equation of the relaxation processes was solved iteratively to yield single collision rate constants. Additionally, time profiles were recorded. Rate data on the 8 and 12 lowest rotational levels are furnished for NH(c) and ND(c), respectively. These data are compared with previous results on BH(A 1Π) and NH and PH in their (A 3Π) states. The rate constants for rotational energy transfer can be represented by the exponential gap law. © 1996 American Institute of Physics.

    Notes: The relative stability of clusters of fullerenes has been investigated. By heating the clusters before ionization we have obtained mass spectra where only the monomer and (C60)13 are present in significant amounts. An approximately 20% increase of the activation energy for evaporation of a monomer from (C60)13 compared to that from (C60)14 explains the experimental results. © 1996 American Institute of Physics.

    Notes: In the yeast Saccharomyces cerevisíae, trehalose-6-phosphate (tre-6-P) synthase encoded by GGS1/TPS1, is not only involved in the production of trehalose but also in restriction of sugar influx into glycolysis in an unknown fashion; it is therefore essential for growth on glucose or fructose. In this work, we have deleted the TPS2 gene encoding tre-6-P phosphatase in a strain which displays very low levels of Ggs1/Tps1, as a result of the presence of the byp1-3 allele of GGS1/TPS1. The byp1-3 tps2Δ double mutant showed elevated tre-6-P levels along with improved growth and ethanol production, although the estimated concentrations of glycolytic metabolites indicated excessive sugar influx. In the wild-type strain, the addition of glucose caused a rapid transient increase of tre-6-P. In tps2Δ mutant cells, which showed a high tre-6-P level before glucose addition, sugar influx into glycolysis appeared to be diminished. Furthermore, we have confirmed that tre-6-P inhibits the hexokinases in vitro. These data are consistent with restriction of sugar influx into glycolysis through inhibition of the hexokinases by tre-6-P during the switch to fermentative metabolism. During logarithmic growth on glucose the tre-6-P level in wild-type cells was lower than that of the byp1-3 tps2Δ. mutant. However, the latter strain arrested growth and ethanol production on glucose after about four generations. Hence, other mechanisms, which also depend on Ggs1/Tps1, appear to control sugar influx during growth on glucose. In addition, we provide evidence that the requirement for Ggs1/Tps1 for sporulation may be unrelated to its involvement in trehalose metabolism or in the system controlling glycolysis.
