    Notes: Optical and electron spin resonance spectroscopies have been used to investigate the thermal stability of lasers based on Cr3+:LiSr0.8Ca0.2AlF6, a material in which the onset of critically inhibiting nonradiative decay at temperatures below 300 K might be expected based on results from other Cr3+-activated disordered gain media. The influence of disorder on the Cr3+ environment is shown to be very small in LiSr0.8Ca0.2AlF6 so that the 4T2→4A2 fluorescence transition is not broadened relative to that in LiSrAlF6 and the onset of significant nonradiative decay is also held above room temperature. © 1998 American Institute of Physics.

    Notes: This letter discusses the optical properties of Cr3+ ions in mixed borate crystals with the formula RX3(BO3)4, where R3+=Y3+ or Gd3+ and X3+=Al3+ or Sc3+. Measurements of the optical absorption spectra have been used to estimate the crystal field strength Dq and the Racah parameters B and C. These quantities confirm that in YAl3(BO3)4(YAB) and GdAl3(BO3)4(GAB)Cr3+ ions occupy strong crystal field sites 2E)〈E(4T2)], whereas in YSc3(BO3)4(YSB) and GdSc3(BO3)4(GSB) the Cr3+ ions occupy weak field sites 4T2)〈E(2E)]. The consequence is that the fluorescence spectrum of Cr3+:YAB and GAB reveals the sharp R lines at low temperature and R lines plus broadband at 300 K. Both Cr3+:YSB and GSB emit into the broadband 4T2→4A2 transition at all temperatures. The potential of these materials as laser gain media is discussed. © 1995 American Institute of Physics.

    Notes: Abstract The aim of this study was to determine whether lipid A-associated proteins (LAP) from two periodontopathogenic species of bacteria were able to stimulate interleukin-6 (IL-6) release from human gingival fibroblasts and myelomonocytic cells. LAP and lipopolysaccharide (LPS) were extracted from Porphyromonas gingivalis and Prevotella intermedia and added to cultures of human gingival fibroblasts and mono-mac-6 monocytic cells. Release of IL-6 into the culture supermatants was determined by ELISA. LAP and LPS from Por. gingivalis, but not from Prev. intermedia, stimulated IL-6 release from both cell types in a dose-dependent manner although LPS was less potent than LAP in inducing IL-6 release from the fibroblasts. IL-6 was detectable in cultures of both cell types following stimulation with LAP from Por. gingivalis at a concentration as low as 10 ng/ml. In response to LAP from Prev. intermedia, IL-6 was produced by mono-mac-6 cells but not by fibroblasts. Our results show that bacterial cell wall components other than LPS can induce IL-6 release from cells of the periodontium in vitro. The production of such potent immunomodulatory agents in vivo may contribute to the connective tissue breakdown characteristic of chronic periodontitis.

    Notes: Abstract Saline extracts of Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis and Eikenella corrodens contain surface-associated components of these bacteria. It has been shown that these extracts are potent stimulators of bone resorption in vitro. The possibility that the components of these surface-associated materials (SAM) could contribute to the serum immune response in patients with juvenile or adult onset forms of rapidly progressive periodontitis were investigated by direct binding ELISA. Very high titres of serum IgG antibodies to SAM from A. actinomycetemcomitans were detected in patients with localized juvenile periodontitis (LJP). Patients with adult onset rapidly progressive periodontitis (RPP) had significantly raised antibody levels to SAM from P. gingivalis. Both groups of patients had significantly raised levels of antibodies to SAM from E. corrodens compared with control sera. Thus, not only does solubilized SAM have the capacity to induce bone resorption, but it also contributes to the antigenic load on the immune system in LJP and RPP.

    Notes: Abstract: There is strong evidence implicating Actinobacillus actinomycetemcomitans as the causative agent of localised juvenile periodontitis (LJP), a disease characterised by rapid destruction of the tooth-supporting tissues. This organism possesses a large number of virulence factors with a wide range of activities which enable it to colonise the oral cavity, invade periodontal tissues, evade host defences, initiate connective tissue destruction and interfere with tissue repair. Adhesion to epithelial and tooth surfaces is dependent on the presence of surface proteins and structures such as a microvesicles and fimbriae. Invasion has been demonstrated in vivo and in vitro although the mechanisms involved are poorly understood. The organism has a number of means of evading host defences which include: (i) inhibiting polymorphonuclear leukocyte (PMN) chemotaxis; (ii) killing PMNs and monocytes; (iii) producing immunosuppressive factors; (iv) secreting proteases capable of cleaving IgG; and (v) producing Fc-binding proteins. Surface components of A. actinomycetemcomitans are potent stimulators of bone resorption and can induce the release of a range of cytokines which can initiate tissue destruction. A number of surface components can also inhibit the proliferation of fibroblasts and their production of components of the extracellular matrix. Little is known, however, regarding the way in which these factors operate in vivo to produce the pathological features of the disease.

    Notes: Abstract. Molecular chaperones, also known as heat shock proteins (hsp), are intracellular proteins found in all cells that catalyze protein folding. We have discovered that one class of bacterial molecular chaperone, the chaperonins, are potent inducers of bone resorption. To address the question of whether the osteolytic activity of the chaperonins was unique to this protein class, or was a common attribute of molecular chaperones generally, we have examined a number of bacterial and mammalian molecular chaperones for activity in the murine calvarial bone resorption assay. All the Escherichia coli molecular chaperones (groEL, groES, and dnaK) were active. The osteolytic activity of groEL was inhibited by indomethacin and the natural antagonist of interleukin-1 receptor antagonist (IL-1ra) but was unaffected by neutralization of tumor necrosis factor (TNF) or inhibition of 5-lipoxygenase. Mammalian molecular chaperones of molecular mass 27, 47, 70, and 90 kDa were also tested and, with the exception of the 47 kDa protein, all showed activity in the murine calvarial assay. Molecular chaperones appear, therefore, to have the capacity to modulate the cellular processes in bone explant cultures, resulting in resorption of the calcified matrix. The possibility that these proteins could play a role in the normal or pathological remodeling of bone is discussed.

    Notes: Abstract. Oral infections due to Candida albicans are a common occurrence in patients with acquired immunodeficiency syndrome (AIDS). The purpose of this investigation was to determine whether the yeast and hyphal forms of the organism could be killed using the light-activated antimicrobial agent toluidine blue O (TBO). Three variables were investigated: TBO concentration, laser light dose and pre-irradiation time (PIT). Irradiation with light from a helium neon (HeNe) gas laser used in conjunction with the photosensitiser TBO resulted in substantial kills of both the yeast and hyphal forms. Killing was light dose-dependent with 42 J being the most effective dose. The optimum PIT for the yeast form was 5 min, whereas killing of the hyphal form was not affected by PIT. The results of this study have shown that both forms of C. albicans are susceptible to lethal photosensitisation using TBO in conjunction with HeNe laser light, suggesting the possibility that this approach could be useful for eliminating the organism from diseased lesions in vivo.

    Notes: Abstract There is currently great interest in iodine as a micro nutrient. Both high and low intakes have been associated with thyroid cancer incidence. Development of dietary iodine monitors is needed to supplement the use of dietary recall methods which have not been well validated for iodine. In this study, 30 pooled urine samples, from ethnic groups on various islands in the South Pacific, were analyzed for iodine using epithermal instrumental neutron activation analysis (EINAA).
