Notes: A convenient DNA-based identification system is described for testing the species origin of meat samples. Probes are generated by PCR with primers binding to species-specific satellite DNA and hybridized to DNA purified from meat. This method is more robust and versatile than methods based on oligonucleotide hybridization. With the exception of a slight cross-reaction of mutton and beef, each probe only recognized the species from which it was derived. Purifying the DNA with a DNA-binding resin improved the sensitivity. Admixtures of 0.1–0.5% can be detected in raw meat and 0.5–5% in autoclaved meat samples. The method can be adapted to detect any eukaryotic species for which species-specific DNA sequences are available. This method has proven its value in the routine inspection of meat samples by revealing more cases of deliberate or accidental species substitution and admixture than conventional techniques.

Notes: Summary The structural and electronic properties of fluorinated 1-N-arylamino-1-arylmethanephosphonic acid esters were studied and related to the inhibitory effects on NADH:ubiquinone oxidoreductase (complex I). Electrostatic potential surfaces, dipole moments and molecular geometries were analysed. Based on the conformational analysis and the electronic parameters, a simple model for the active site of the fluorinated 1-N-arylamino-1-arylmethanephosphonic acid esters was developed, explaining the inhibitory power. The strongest inhibition effects were found for the 1-(N-4-trifluoromethoxyphenyl)-amino-1-phenylmethanephosphonic acid diethyl ester 1bab.

Notes: The differences in complexation behaviour of two diastereomeric tetraazamacrocycles, meso (1) and racemic (2) 5,5,7,12,12,14-hexamethyl-1,4,8,11-tetraazacyclotetradecane, are explained by different conformations in solution, 1 being one stable conformer of almost rigid structure while 2 shows higher conformational flexibility to avoid steric strain. The methods used were 1H and 13C NMR and molecular modelling.

Notes: The decomposition of 15N3F at -20°C leads to cis- and trans-15N2F2. The corresponding 19F NMR spectra were analysed as [AX]2 systems. 19F, 15N and 14N NMR spectra were simulated using the novel PC program systems DSYMPCD, DCYMPCD and WIN-DAISY running under WIN-NMR. Stereospecific coupling constants were obtained for both isomers. Comparison of the 19F NMR spectra from both the [1/2A1/2X]2 and the [1A1/2X]2 systems in 15N2F2 and 14N2F2 overcomes the inherent ambiguities of the [AX]2 analysis.

Notes: Variable-temperature 31P NMR spectroscopy was used to extract Arrhenius and Eyring parameters of the hindered rotation about amide bonds in an acylated aminophosphonate. The results permit the differentiation between stepwise or concerted rotation.
