    Notes: The regulation by phytochrome of stem elongation in light-grown plants depends on gibberellins (GAs). To investigate whether this is mediated by a change in GA metabolism, the effect of the GA biosynthesis inhibitor LAB 198 999 (an acylcyclohexadione derivative) on the end-of-day far-red (FR) response in cowpea (Vigna sinensis L.) epicotyl explants has been investigated. Growth of epicotyl explants of light-grown seedlings was enhanced when treated with far-red light before incubation in the dark (end-of-day FR effect). Low doses of LAB 198 999 (0.05 and 0.5 μg explant−1) reduced the effect of FR, whereas 5 to 50 μg explant−1 stimulated elongation of both red light (R)- and FR-treated epicotyl explants while nullifying the differences between R and FR treatments. In paclobutrazol-treated epicotyl explants, FR enhanced the response to applied GA1 and GA20, whereas LAB 198 999 increased the activity of GA1 and decreased that of GA20, [3H]Gibberellin A1, injected into the basal part of the epicotyl, was transported and metabolized mainly to [3H]GA8 in the apical 20 mm of the epicotyl. The conversion of [3H]GA1 to [3H]GA8 was dramatically reduced by both end-of-day FR treatments and LAB 198 999 applications. In addition, both treatments enhanced epicotyl elongation. It is proposed that the regulation of cowpea epicotyl growth by phytocrome is mediated, at least partially, by modifying GA1 degradation.

    Notes: Frozen muscle samples from 44 striped dolphins stranded on the Spanish Mediterranean coasts from 1990 to 1993 have been studied by means of mitochondrial DNA (mtDNA) restriction site analysis. Thirty-five of these dolphins were affected by a die-off occurring during this time in the western Mediterranean Sea. The mtDNA from each dolphin was digested with 15 restriction endonucleases that recognized 61 different restriction sites. The specific location of these sites on the mitochondrial gene map allowed us to determine the distribution of variability along this molecule. From the restriction analysis, a total of 15 different composite patterns or haplotypes was obtained and their phylogenetic relationships established both by cladistic and phenetic methods.Estimates of sequence divergence among the 15 haplotypes ranged from 0.148% to 0.919%. Haplotype diversity (average heterozygosity) obtained was 0.789, a high value compared to other available data on cetaceans, especially considering that only a small area of the distribution of the species has been sampled. Neither population subdivision nor any evidence of correlation between haplotype frequencies and temporal or geographic distribution of dolphins has been detected.

    Notes: Abstract Molecular methods allow an extremely fine strain typing that can be used to establish the population structure of bacterial species. This methodology has been used to characterize a collection of 74 uropathogenic Escherichia coli obtained from three hospitals located in geographically distant towns in Spain, some representatives of the ECOR collection and other reference strains. Genomic DNA was analyzed by RAPD (Random Amplified Polymorphic DNA) that can characterize a bacterial strain to the level of defining individual clones. The 16S rDNA-23S rDNA spacers were amplified by PCR and submitted to restriction analysis. Finally, the presence or absence of G adhesins in Escherichia coli as well as the type of adhesin (three types are known) have been shown by PCR amplification followed by digestion with restriction enzymes. As expected a wide diversity was shown by RAPD and identical patterns were only found in the case of strains isolated from the same individual, an obvious case of relapse. Analysis of the spacers' restriction patterns showed the presence of two markedly differentiated clusters that we have named α and ß. Both RAPD and spacer restriction patterns originated similar clusters of strains showing a consistency in the evolution of the global genome with the sequence variation of the ribosomal spacers. Furthermore, most of the strains having G-adhesin, with only a few exceptions, corresponded to the α rRNA spacer group. The two spacer types detected were also consistent with some phenotypic markers such as sucrose and raffinose utilization. The α and β clusters could be intraspecific groups produced by partial sexual isolation or other barriers that are originating a divergent evolution.

    Notes: Abstract Correlation methods for recognition of polychromatic range images are presented. First, polychromatic range images are described. The correlation can be performed using either a single channel compressing both the range and the color images or a multichannel approach. The advantages of both methods are discussed. Translation invariant recognition along the x, y, and z axis is obtained. In the single channel approach, the range and the hue are combined using phase and amplitude coding. Then the double-channel approach is considered using sine-coding for the input image and phase-coding for the reference image. Finally, both channels are combined in order to improve the discrimination capability of the system. Optical implementation is performed.

    Notes: Abstract. Gibberellins A1 and A3 are the major physiologically active gibberellins (GAs) present in young fruit of pea (Pisum sativum L.). The relative importance of these GAs in controlling fruit growth and their biosynthetic origins were investigated in cv. Alaska. In addition, the non-13-hydroxylated active GAs, GA4 and GA7, were identified for the first time in young seeds harvested 4 d after anthesis, although they are minor components and are not expected to play major physiological roles. The GA1 content is maximal in seeds and pods at 6 d after anthesis, the time of highest growth-rate of the pod (Garcia-Martinez et al. 1991, Planta 184: 53–60), whereas gibberellic acid (GA3), which is present at high levels in seeds 4–8 d after anthesis, has very low abundance in pods. Gibberellins A19, A20 and A29 are most concentrated in seeds at, or shortly after, anthesis and their abundance declines rapidly with development, concomitant with the sharp increase in GA1 and GA3 content. Application of GA1 or GA3 to the leaf subtending an emasculated flower stimulated parthenocarpic fruit development. Measurement of the GA content of the pods at 4 d after anthesis indicated that only 0.002–0.5% of the applied GA was transported to the fruit, depending on dose. There was a linear relationship between GA1 content and pod weight up to about 2 ng · (g FW)−1, whereas no such correlation existed for GA3 content. The concentration of endogenous GA1 in pods from pollinated ovaries is just sufficient to give the maximum growth response. It is concluded that GA1, but not GA3, controls pod growth in pea; GA3 may be involved in early seed development. The distribution of GAs within the seeds at 4 d post anthesis was also investigated. Most of the GA1, GA8, GA19, GA20 and GA29 was present in the testa, whereas GA3 was distributed equally between testa and endosperm and GA4 was localised mainly in the endosperm. Of the GAs analysed, only GA3 and GA20 were detected in the embryo. Metabolism experiments with intact tissues and cell-free fractions indicated compartmentation of GA biosynthesis within the seed. Using 14C-labelled GA12, GA9, 2,3-didehydroGA9 and GA20 as substrates, the testa was shown to contain 13-hydroxylase and 20-oxidase activities, the endosperm, 3β-hydroxylase and 20-oxidase activities. Both tissues also produced 16,17-dihydrodiols. However, GA1 and GA3 were not obtained as products and it is unlikely that they are formed via the early 13-hydroxylation pathway. [14C]gibberellin A12, applied to the inside surface of pods in situ, was metabolised to GA19, GA20, GA29, GA29-catabolite, GA81 and GA97, but GA1 was not detected. Gibberellin A20 was metabolised by this tissue to GA29 and GA29-catabolite.
