    Notes: Abstract A new technique which combines skin micro injection of minute amounts of drugs (0.5 μl) and measurement of capillary permeability by intravital fluorescence videomicroscopy and densitometry is introduced. Glass micropipettes with a tip diameter of 7–9 μm are inserted by a micromanipulator into the stratum papillare containing the capillary loops and used for microinjection by microsyringe and special dispenser. Transcapillary diffusion of sodium fluorescein applied by intravenous bolus injection is visualised by fluorescence videomicroscopy and stored on videotape. Perivascular fluorescent light intensity (FLI) is measured in arbitrary units (AU) by videodensitometry around the site of microinjection during playback of the videotapes. The method was tested by microinjection of 0.5 μl histamine (1‰ solution) at the distal tibial plateau. Mean FLI values representing microvascular permeability were 2186 AU 10 min after microinjection of histamine, 420 AU after physiologic saline and 1228 AU after histamine combined with oral intake of 20 mg cetirizine. Cetirizine significantly reduced (P 〈 0.01) the increased permeability induced by histamine. However, the mean values after injection of the H1-blocker were still significantly enhanced (P 〈 0.01) when compared to the mean values observed after injection of physiologic saline solution, which provoked only minor microtrauma. The technique offers new scope for pharmacological testing in man.

    Notes: Abstract  The rumen anaerobic fungus Neocallimastix frontalis was biolistically transformed using plasmids containing the bacterial β-glucuronidase gene (GUS) fused to the promoter sequences of the enolase gene from N. frontalis. Multiple copies of the plasmids were precipitated onto tungsten particles and delivered into zoosporangia and a mycelial mat by a helium-driven biolistic device. Transformants were detected by histochemical assay for β-glucuronidase. It was found that the enolase promoter sequences tested were responsible for the transient expression of the β-glucuronidase gene. This is the first study presenting results on the transformation of an anaerobic fungus.

    Notes: Restriction fragment length polymorphisms were generated for 52 species of lignicolous European polypores belonging to 31 different genera. An enzymatically amplified portion of the nuclear-encoded ribosomal DNA was found to be about 1800–1900 bp long for most taxa. After digestion with Hpa II, 44 distinct phenotypes were detected for the species under study; application of the additional restriction endonucleases, Hin 6 I and Hinf I, resulted in 48 species-specific and two genera-specific phenotypes. The procedure described should allow for a reliable identification of unknown mycelia of lignicolous fungi.

    Notes: Abstract  To investigate structural differences between propulsory and antigravity muscles, the spatial distribution of slow (type I) and fast (type II) muscle fibres in forelimb muscles of two species of small mammals was studied, Galea musteloides and Tupaia belangeri. Serial sections through complete forelimbs were prepared. Following histochemical fibre typing, the forelimbs were reconstructed three-dimensionally using product design software. Most forelimb muscles of both species showed a homogenous distribution of type I fibres. In the supraspinatus and triceps brachii (capita longum et laterale) muscles, however, a segregation of fibre types into ”fast” superficial areas and ”slow” deep regions was observed. Slow regions contained at least 60% type I fibres and were positioned along intramuscular extensions of the tendons of insertion. The functional implications of fibre type regionalization are discussed. An analysis of intramuscular fibre type distribution during postnatal myogenesis revealed no significant differences in muscle fibre differentiation between altricial and precocial juveniles. Differences in locomotor ability probably arise from heterochronic development of connective tissue components (endo- and perimysium).

    Notes: Abstract. Detonation propagation behavior associated with sudden expansions has been investigated both experimentally and numerically. Different mechanisms, from sustained propagation to detonation failure and reinitiation including shock and flame front decoupling and recoupling have been observed with the schlieren technique. The shock-induced flame propagation has been modeled with two-step chemistry and structured two-dimensional CFD on arbitrary geometries. The results of the numerical simulations show good correspondence to the variety of phenomena observed in experiments. Thus the numerical simulation can be used to study detonation propagation in complex geometries. It provides a tool for the design of safety devices and aids experimental investigations.
