    Notes: Abstract The natural feeding of 485 Octopus mimus (164 to 3088 g) was studied in relation to the species' life cycle and environmental seasonal variations off the north of Chile from autumn 1991 to summer 1992. Analysis of digestive-tract contents revealed that O. mimus preyed upon 25 different prey items belonging to five zoological groups (Teleostei, Mollusca, Crustacea, Echinodermata and Polychaeta). Cannibalism was only occasional. The results indicate that the diet and food intake of this species are significantly affected by sex and maturation. Senescent individuals ingest a small amount of food, and their diet is mainly based on small, not very motile prey. The food intake, expressed as body weight, of non-senescent individuals is higher in females than in males. Seasonal changes in sea-water temperature seem to be followed by adjustments in food intake. Like other Octopus species, O. mimus appears to be an opportunistic predator.

    Notes: Abstract The relationship between reproduction and condition was studied in a 15 mo sample of 919 maturing, mature and post-spawning female Octopus mimus (388 to 3714 g) caught in Iquique (North Chile). O. mimus is a semelparous species, with reproduction taking place all year round. Investment in reproductive tissues was, on average, 9.9% of mature female body weight, independent of season. However, somatic growth during maturation was dependent on season and varied between 26 and 63% of the whole-body growth in weight. The condition of females did not vary markedly until spawning, although seasonal variations were apparent, winter being the most unfavourable. Condition deteriorated dramatically after spawning, during parental care of the eggs. During this period, somatic-tissue depletion, mainly from the muscles, was 〉25% of the total body weight of mature females. The fecundity of O. mimus was probably limited by the costs associated with parental care of the eggs.

    Notes: Abstract  Analyses of groundwater are routinely performed by control laboratories within the European Union to monitor the levels of major elements. The quality control of such determinations requires the use of certified reference materials which are representative of natural samples. However, the preparation and stabilisation of natural groundwater for their major element composition is difficult to achieve and materials simulating the mean composition of real samples often represent the best alternative. In order to investigate the feasibility of preparation and stabilisation of simulated groundwater samples, various tests were necessary prior to the production of a large batch of candidate reference materials. This paper presents the preparation of simulated groundwater reference materials and the different stabilisation procedures tested.

    Notes: Abstract Atomic emission spectroscopy (AES) is used to monitor volatilization during waste vitrification with thermal plasmas. Model baste specimens consist of am alumino-silicate clay spiked with 10 wt. % Fe, Ni, or Cr. Specimens are vitrified in processing atmospheres with 0, 8.5, and 17 vol. % oxygen/argon concentrations. Particulate generated from condensation of the volatilized materials is collected following each specimen run and analyzed by energy dispersive X-ray analysis to determine composition. Spectra are collected from the ultraviolet range 240–380 nm in where the presence of metal vapor is readily detected. Correlation between line emission intensities and volatilization rates allows the establishment of provisional detection limits for the volatilization of Ni, Cr, Si, and Fe, of 1 × 103 g/s, 4 × 104 g/s, 2 × 104 g/s, and 5 × 105 g/s, respectively. The results of this investigation support the concept of using AES as an in-situ process monitor for feedback to optimize plasma processing of hazardous metal-containing waste.

Membranes from in vivo labeled cells of Rhodobacter capsulatus U43[pTX35] grown photosynthetically carried 60% of the [32P]-Pi in the “heavy” fraction (HM) after sucrose gradient sedimentation. Metal-chelating chromatography of either “heavy” or “light” (LM) membrane fractions rendered similar Bchl-protein complex profiles after octyl-glucoside treatment, including most of the radioactivity in the same corresponding elution fraction (F II). Similar labeling distribution of pigment-protein complexes was obtained for membranes of dark-grown cells induced by lowering oxygen tension. Fractions derived from HM showed highly labeled LHIα, whereas the same complex from LM was essentially [32P]-Pi-free, as revealed by SDS-PAGE followed by autoradiography. Phospholipid analysis showed a similar pattern for membranes isolated from cells photosynthetically or semiaerobically grown, being the most abundant: phosphatidylglycerol, phosphatidylethanolamine, cardiolipin, and phosphatidylcholine. Part of the phospholipids from HM comigrated with LHIα during SDS-PAGE and dissociated from the complexes only after solvent extraction and hydrophobic chromatography. However, a small amount remained always attached to LHIα, indicating an unusual strong interaction. These results suggest the existence of two operationally defined membrane regions carrying LHIα complexes differing in phosphorylation status and protein-phospholipid interaction.

    Notes: Endothelial cell differentiation into capillary structures is a complex process that requires the concerted effects of several extracellular matrix proteases, including plasminogen activators. Here, the role of tissue-type plasminogen activator (tPA) and urokinase-type plasminogen activator (uPA) was evaluated in an in vitro model of endothelial morphogenesis involving organization of human umbilical vein endothelial cells into tubular structures when they are cultured on the basement membrane preparation, Matrigel. Both uPA and tPA were detected in HUVEC cultures on Matrigel, and inhibitors of plasminogen activators or of serine proteases decreased the extent of the tube network formed by the cells. The decrease resulting from serine protease inhibitors was additive to that from matrix metalloproteinase inhibitors which have previously been shown to decrease tube formation in this model, suggesting that the two classes of proteases modulate tube formation by distinct mechanisms. Plasminogen activator inhibitor (PAl)-1 decreased tube formation by 50% when added up to 4.5 h after the initiation of an 18 h assay and caused 25% inhibition when added 9.5 h after culture initiation, indicating that the effects of plasminogen activators are not limited to an early event in the differentiation process. Steady-state expression of mRNA for uPA increased during the first several hours of culture on Matrigel, further supporting a role for PA activity throughout the process of tube formation. These findings suggested that PAs may affect multiple events during tube-forming activity. A fucosylated peptide comprising the amino-terminal domain of uPA that binds to the uPA receptor (uPAR) but lacking proteolytic activity enhanced tube formation. In contrast, a defucosylated form of the same peptide had no effect. Since fucosylation of this fragment has been shown to be essential in other models of cell stimulation by uPA-uPAR interaction, these data support the hypothesis that uPA enhances endothelial morphogenesis both through proteolytic activity and via uPAR occupancy. Plasminogen activators could facilitate angiogenesis in vivo. © 1995 Wiley-Liss Inc.
