    RB1 deletion in gonadoblastoma in an XY female (1997)

    Notes: Abstract Cytogenetic studies of normal and tumor cells in a patient with gonadal dysgenesis and bilateral gonadoblastoma were performed. The karyotype was 46,XY in peripheral blood lymphocytes and skin fibroblasts. The conserved region of the SRY gene was detected by polymerase chain reaction amplification. Sequencing of this region did not reveal any alterations. A 46,XY chromosome constitution was observed in the right gonadoblastoma, but a partial deletion of chromosome 13 was present in the left tumor. This deletion included band 13q14, where the retinoblastoma gene is mapped. The study of the polymorphism of the variable number of tandem repeats region in intron 17 of the RB1 locus disclosed loss of heterozygosity in both the left tumor, which showed the deletion of chromosome 13, and in the right tumor, where no chromosome alterations of chromosome 13 were detected. In situ hybridization covering 130 kb of RB1 showed that a partial deletion of one of the RB1 alleles had occurred in the right tumor. Since the deletions affected different alleles in each tumor, independent events must have been involved in the development of the tumors. These findings point toward a significant role of RB1 in the development of gonadoblastoma.

    Notes: Abstract Insect-transmitted plant pathogens can have positive, negative or no effects on the vector insect. Effects could be direct (caused by the pathogen) or indirect (mediated by changes in the plant). Pathogen effects on non-vector insects are less well known.Peregrinus maidis (Ashmead) (Homoptera: Delphacidae), an insect that is not a vector of the corn stunt spiroplasma (CSS), weighed less 15 days after feeding on CSS-infected corn plants relative to insects feeding on healthy corn plants. Fecundity of non-vector insects that were removed from diseased plants was higher than for insects coming from healthy plants. ForDalbulus maidis (DeLong & Wolcott) (Homoptera: Cicadellidae), an insect that transmits CSS, there were no differences in weight, longevity, eggs per day, weekly or total fecundity after feeding on healthy or on CSS-infected corn plants. Significant differences in some phloem amino acids were detected between healthy and CSS-infected plants. Infected plants also showed an increased phloem acidity as disease symptoms progressed. Feeding on CSS-infected corn plants by an insect that does not vector the pathogen infecting the plant can have favorable consequences as evidenced by an increased fecundity.

    Notes: Abstract The methylation profile of ten α-satellites was investigated in normal individuals and in ICF (Immunodeficiency, Centromeric instability, Facial abnormalities) patients. Two out of three ICF patients showed modified methylation of these sequences, reproducing a placental profile. CENP-B boxes, the binding sites of centromeric protein B, were always skewed toward nonmethylation. Unexpected results were observed in normal individuals: in somatic adult tissues the methylation pattern of α-satellite DNA varied between chromosomes, and in fetal tissues these satellites were homogeneously undermethylated. Detailed methylation analysis of CENP-B boxes revealed that unmethylated α-satellite units coexist with thoroughly methylated regions. These observations showed that the two major components of constitutive heterochromatin are differently methylated in normal somatic and fetal tissues, since classical satellites are consistently methylated. The definite changes in the methylation profile of heterochromatin in somatic chromosomes and the asynchronous timing of methylation of classical and α-satellites during development may reflect specific roles of highly repeated sequences in genomic organization.

    Notes: Abstract This paper reports a case of XY gonadal dysgenesis in two sisters. Both patients presented an eunochoid female phenotype with normal external genitalia. At laparotomy, the elder sister was found to have bilateral gonadoblastoma. Cytogenetic studies, which included G and C banding and in situ hybridization, showed that the patients had an apparently normal 46, XY karyotype. PCR analyses revealed absence of the conserved portion (HMG box) of the SRY gene and of the Y chromosome pseudoautosomal boundary region sequence in both patients. The presence of the ZFY sequence was detected by Southern hybridization in the two affected sisters. The patients' father (46, XY, no mosaicism detected in peripheral blood lymphocytes) was positive for SRY and ZFY sequences. The occurrence of gonadoblastoma is discussed in terms of the genetic factors that may lead to tumor development.

    Notes: Abstract Development projects are rapidly changing the landscape in Brazilian Amazonia. Environmental impact assessments have been required since 1986, and the regulatory system is evolving as precedents are set by each new development project. The Jatapu Dam in Roraima provides an illustration of underlying impediments to assessment of environmental costs and to due consideration being given to these assessments when decisions are made. The high priority placed on the dam by the Roraima state government is unexplainable in terms of economic returns. The place of the dam in a long-term political strategy provides the best of several possible explanations, any one of which is incompatible with a “rational” weighing of economic and environmental costs and benefits. A number of lessons can be drawn from the experience of Jatapu, but some of the problems have no solution. The barriers to rational decision making illustrated by Jatapu apply to development projects in many parts of the world.

    Notes: Abstract The proposed Cotingo Dam in Brazil's far northern state of Roraima is examined with the objective of drawing lessons for Brazil's system of evaluating environmental, social, and financial consequences of development decisions. The Cotingo Dam illustrates the difficulty of translating into practice the principles of economic and environmental assessment. Examination of the financial arguments for the Cotingo Dam indicates that justifications in this sphere are insufficient to explain why the project is favored over other alternatives and points to political factors as the best explanation of the project's high priority. Strong pressure from political and entrepreneurial interest groups almost invariably dominates decision making in Amazonia. The analysis indicates the inherent tendency of the present system to produce decisions in favor of large construction projects at the expense of the environment and local peoples. The requirements intended to assure proper weight for these concerns, such as the report on environmental impacts (RIMA) and the public hearing, fail to serve this role. Cotingo also provides a test case for constitutional protections restricting construction of dams in indigenous lands.

    Notes: The formation of an apatite-like layer is achieved by immersing Ti6Al4V and TiAl2.5Fe substrata in Hank's Balanced Salt Solution (HBSS). The layer was characterized by several techniques, namely X-ray microanalysis, X-ray diffraction and X-ray photoelectron spectroscopy. The results suggest that the layer produced by immersion in HBSS is in the form of an amorphous apatite. The pH and the concentrations of calcium and phosphate were monitored as a function of time. In vitro tests with rat bone marrow were performed in order to mimic the bone/biomaterial interface. They were performed on both immersed and non-immersed samples. The in vitro bone marrow results suggest that the apatite-like layer formed may improve the bone bonding characteristics of the studied titanium alloys.

    Notes: Abstract The effect of nitrogen-ion implantation on the electrochemical behaviour of 316 L stainless steel in a simulated physiological solution (HBSS-Hank’s Balanced Salt Solution) was studied by open-circuit potential versus time and cyclic polarization techniques, with the aim of characterizing the surfaces and choosing the best nitrogen-ion fluence. Three fluences (1015, 1016 and 1017 ions/cm2) were used. The 1016 ions/cm2 N+ fluence improves the corrosion resistance of the 316 L stainless steel.

    Notes: Abstract Nitrogen ion-implanted Ti–6Al–4V, Ti–5Al–2.5Fe and 316 L stainless steel and nitrogen or carbon sputter-coated samples were inoculated with rat bone marrow. The interface between the cell layer and the substrata was studied by X-ray photo-electron spectrometry and observed by scanning electron microscopy (SEM). Ca and P were detected on all materials after in vitro cell culture. Titanium appears to be present mainly in the form of TiO2.
