    Notes: High-purity immunoglobulins (Ig), which may be useful for immunologic supplementation of food products, were isolated from Cheddar cheese whey in a one-step process using avidin-biotinylated yolk immunoglobulin (IgY) column chromatography. Yolk antibodies specific to bovine IgG (IgYIgG) were biotinylated with biotinyl amido-hexanoic acid-N-hydroxy-sulfo-succinimide ester without any notable effect on antigen-binding activity, and coupled to immobilized avidin columns. The resulting avidin-biotinylated IgYIgG columns, with binding capacity of 50–55% (w/w percent ratio of IgG to immobilized IgYIgG), were used for specific binding of IgG from cheese whey. Elution with a commercially available eluent (Actisep) or 0.1 M glycine-HCl buffer at pH 2.8 yielded IgG with purity of 99% by radial immunodiffusion.

    Notes: The use of surface-active tracers for measuring the interfacial area between no aqueous phase liquids (NAPLs) and water was evaluated in a hydraulically isolated test cell installed in a surficial aquifer located at Hill Air Force Base (AFB), Utah. Interfacial tracers were developed at the University of Florida for quantifying immiscible fluid-fluid interfaces (air-water or NAPL-water) in porous media. Sodium dodecyl benzene sulfonate (SDBS) was used as the interfacial tracer to measure the effective NAPL-water interfacial area (aNw), while 2,2-dimethyl-3-pentanol (DMP) was used as the partitioning tracer to estimate the NAPL saturation (SN). The observed retardation of SDBS and DMP when compared to a nonreactive tracer (bromide or methanol) at eight multilevel sampling locations and one extraction well, was used to quantify the aNw and SN values averaged over the interval between the injection wells and each monitoring point. The NAPL morphology index, defined here as HN= aNw/φSN (φ= porosity), varied significantly within the test cell. In locations where the magnitude of HN was large, the NAPL was assumed to be more or less uniformly spread, providing good contact with the mobile fluid. In contrast, regions with low HN values were assumed to have NAPL that was more heterogeneously distributed as isolated patches providing poor contact with the mobile fluid. The index HN, provided by the combined use of interfacial and partitioning tracers, has important implications for NAPL source region remediation employing in situ flushing technologies.

    Notes: A fast low angle shoot (FLASH) pulse sequence in combination with a multi-inversion procedure was used to rapidly acquire spin-lattice relaxation data for food particles undergoing ohmic heating. The relaxation data acquired by magnetic resonance imaging (MRI) were analyzed using a two-component exponential model to extract both short and long spin-lattice relaxation time, T1. The longer T1 showed a better linear relationship with temperature than the shorter T1 and, therefore, was used to produce more reliable temperature maps. The MRI temperature maps constructed for ohmically heated food particulates showed an entirely different heating pattern than would be found for similar but conventionally heated particulates and indicated that the temperature inside the particulate could be higher than near the surface of the particulate.

    Notes: A water-soluble fraction (WSF) was prepared by filtering 10 times diluted yolk (pH 5.0) stored at 4–6°C through Whatman No. 52 or No.1 filter paper for delipidation. However, solid-phase delipidation using an octadecyl column after filtration through a cellulose powder column was more practical for industrial application. After ultrafiltration (UF) of the WSF (pH 9.0,1.5M NaCl), results showed: Amicon 80–85% recovery, 74–99% purity; Harp 72–74% recovery, 81–84% purity; A/G 75% recovery, 89% purity; Koch 88% purity.

    Notes: A serial filtration system was developed for separating immunoglobulin from egg yolk (IgY). Egg yolk was diluted 10 times with water, adjusted to pH 5.0, frozen, then thawed. The diluted yolk was diafiltered through a hard filter paper at 4-6°C to produce a clear water-soluble fraction (WSF). The WSF was then delipidated by filtering through a hydrophobic membrane filter. Ultrafiltration of delipidated WSF at pH 9.0, at 4-6°C, in the presence of 1.5M NaCl resulted in electrophoretically pure (〉90%) IgY with a recovery of about 92%. Advantages of this method are the use of only a few mild chemicals and the potential of developing a continuous process.

    Notes: The acid phosphatase of an atypical Citrobacter sp. was purified in two isoforms, designated CPI and CPII, which had different Km values for glycerol 1-phosphate and glycerol 2-phosphate The enzyme was not inhibited by the end-product glycerol. Enzyme activity was increased in the presence of phosphate acceptor molecules having free hydroxyl groups (glycerol, methanol, ethanol). 31P-nuclear magnetic resonance spectroscopy indicated transfer of the liberated phosphate onto the alcohol, with the de novo production of (e.g.) glycerol 1-phosphate by enzyme supplemented with phosphomonoester substrate and glycerol.

    Notes: Synthesis and validation of crop models for assessment of of the impact of elevated atmospheric CO2 concentration and anticipated global warming on crop production require crop response data obtained under field-like conditions. The temperature gradient chamber (TGC) with the facility for CO2 enrichment allows the creation of various CO2 and temperature regimes for crops over the entire growth period with relatively inexpensive construction and running costs. The TGC develops a temperature gradient along its longitudinal axis using solar energy during the day and heating at night while maintaining the natural diurnal cycle. The temperature gradient and the CO2 concentration in the TGC are regulated by computer control of the air ventilation rate through the TGC and of the CO2 release rate. Longitudinal gradients of CO2 concentration and water vapour pressure deficit of air in the TGC were generally less than 5% and ±0.2 kPa, respectively.A CO2 enrichment experiment on rice in the TGC showed that a doubling of the CO2 concentration markedly enhanced crop dry matter production. Temperature had less effect on dry matter production, although panicle dry weight was greatly decreased at higher temperature as a result of high-temperature-induccd sterility of rice spikelets. Since rice spikclets are most sensitive to high temperature at the moment of flowering, and their flowering habit is highly synchronized with the diurnal courses of environmental conditions, the TGC is a useful tool in understanding rice responses to changes in atmosphere and temperature.
