    Notes: Abstract A cordycepin-resistant mutant strain of Saccharomyces cerevisiae (CD-R2) was found to be deficient in adenosine kinase. This mutant accumulated S-adenosylhomocysteine during growth in the presence of exogenous adenosine and it grew in a pseudohyphal manner in the presence of this nucleotide.

    Notes: Oroblanco and Melogold are hybrids obtained from pummelo and grapefruit. Limonoids and flavonoids in both juices were analyzed. Oroblanco and Melogold juices contained low concentrations of limonoid glucosides, an average of 99 and 59 ppm, respectively. However, they contained relatively high concentrations of bitter limonoid aglycones, limonin and nomilin, at levels above the limonin bitterness threshold. For comparison, limonoid glucosides in juices of grapefruit, another pummelo hybrid, were also analyzed. Limonin glucoside was the major limonoid glucoside in all juices analyzed. Nomilin glucoside and nomilinic acid glucosides were also present. Oroblanco and Melogold juices contained bitter flavonoids normally found in grapefruit and pummelo including naringin, neohesperidin and poncerin in total amount of 440 ppm in Oroblanco juice and 495 ppm in Melogold juice. They also contained several other nonbitter flavonoids found in grapefruit.

    Notes: In a study on improving utilization of waste from processing of Satsuma mandarin (Citrus unshiu Marcov.) the limonoid glucosides of fruit, juice, and by-products were measured using HPLC and TLC. All materials had the 17-β-D-glucopyranoside derivatives of limonoids reported in other commercial citrus fruit. Citrus molasses was a good source for industrial scale extraction of limonoid glucosides. An extraction system using polystyrene divinylbenzene resins, was developed which could be expanded to industrial scale.

    Notes: The concentrations of limonoid glucosides in orange molasses, a byproduct of citrus juice processing, were measured. The 17-β-D-gluco-pyranoside derivatives (glucosides) of limonin, nomilin, nomilinic acid and obacunone were present and quantified. Orange molasses contained an average of 568 ppm of total limonoid glucosides at 11.8°Brix. Orange molasses (3.8L) contained up to 12.5g of total limonoid glucosides. The molasses could be used as a source for commercial isolation of citrus limonoid glucosides.

    Notes: Juice and seeds from 16 cultivars of mature pummelo fruits were analyzed for limonoid content. Pummelo juice contained an average of 18 ppm limonin and 29 ppm total limonoid glucosides. Compared to other juices, ptmnnelo contained very high concentrations of limonin and very low concentrations of limonoid glucosides. Limonin, nomilin, obacunone and trace amounts of deacetyhromilin were found in pummelo seeds. The 17–β-D-glucopyranoside derivatives (glucosides) of nomilin, nomilinic acid and obacunone were also present. Total limonoid aglycone concentration in the seeds ranged from 773 ppm to 9,900 ppm and total limonoid glucosides ranged from 130 ppm to 1,912 ppm.

    Notes: Micellar-enhanced ultrafiltration (MELT) and air stripping were evaluated for surfactant-contaminant separation and surfactant recovery. Two linear alkyl diphenyloxide disulfonate (DPDS) surfactants were evaluated with the contaminants naphthalene and trichloroethylene. A separation model developed from micellar partitioning principles showed a good correlation to batch MEUF studies, whereas flux analysis highlighted concentration polarization effects in relation to hydrophobe length. MEUF effectively concentrated the surfactant-contaminant system (93 to 99 percent retention); however, this did not result in surfactant-contaminant separation. Batch and continuous flow air stripping models were developed based upon air/water ratio, surfactant concentration, and Micellar partitioning; model predictions were validated by experimental data. Sensitivity analyses illustrated the decline in contaminant-surfactant separation with increasing surfactant concentration (e.g., TCE removal efficiency declines from 83 percent to 37 percent as C-16 DPDS concentration increases from 0 to 55 mM). This effect is greater for more hydrophobic contaminants (naphthalene vs. TCE) and surfactants with greater solubilization potential (C16-DPDS vs. C-12 DPDS). The resulting design equations can account for this effect and thus properly size air strippers to achieve the desired removal efficiency in the presence of surfactant micelles. Proper selection and design of surfactant-contaminant separation and surfactant recovery systems are integral to optimizing surfactant-enhanced subsurface remediation.

    Notes: To investigate the influence of pressure stress on the cell cycle of Schizosaccharomyces pombe, we used a cold-sensitive nda3-KM311 mutant which arrests cell division at a step similar to the mitotic prophase, proposed by Hiraoka and colleagues (Cell 39 (1984) 349–358), under the restrictive temperature, 20°C. The nda3-KM311 cells were first aerobically grown at 30°C, transferred to 20°C for 4 h and shifted to a permissive temperature of 36°C for 15 min. The cells were treated with 100–200 MPa pressure and studied by electron and fluorescence microscopy. At 100 MPa, the nuclear membrane was damaged and the matrix of mitochondria had an electron-dense area. At 150 MPa, the nuclear membrane was broken over broad areas; numerous small vacuoles had fused into large pieces. Actin patches were concentrated in the central region and actin rings were seen in the 20°C-grown cells. Even at 100 MPa, specific actin distribution was lost. Although at 100 MPa, long and fine actin cables were seen all over the cells, large actin patches and the actin rings remained in the center of the cell. They changed into thick and short cables at 150 MPa and above 200 MPa they decomposed but the actin ring was visible even with faint fluorescence. Immunoelectron microscopic observation confirmed this phenomenon.

    Notes: It has been demonstrated that endogenous cytokines including γ interferon (IFN-γ), tumour necrosis factor-α (TNF-α), and interleukin-6 (IL-6) play protective roles but that IL-4 and IL-10 play detrimental roles in nonlethal Listeria monocytogenes infection in mice. In this paper, we studied the roles of endogenous cytokines in a lethal infection with L. monocytogenes in mice. TNF-α and IL-6 titres in the bloodstreams, spleens and livers paralleled bacterial numbers in the organs, and both these cytokines and the bacterial numbers peaked just before the mice died. The high titres of TNF-α notably detected in the circulation in lethal infection were different from those in nonlethal infection. The maximum production of IFN-γ was observed before the peaks of TNF-α and IL-6, and IFN-γ almost disappeared from the bloodstreams and organs just before the mice died. No notable difference of IFN-γ titres between lethal infection and nonlethal infection in the specimens obtained from mice was observed. IL-10 was also detected in the bloodstreams earlier than the peaks of TNF-α and IL-6 during lethal infection, while IL-4 was never detected in the sera. The administration of monoclonal antibodies (mAbs) against TNF-α, IFN-γ, IL-6, IL-4 or IL-10 failed to rescue mice from lethal L. monocytogenes infection, whereas anti-TNF-α mAb and anti-IFN-γ mAb prevented mice from lethality by high-dose endotoxin shock. These results suggest that lethality in L. monocytogenes infection might not be determined solely by these cytokines.

    Notes: Expression of the enzymes for anaerobic nitrate respiration of Pseudomonas aeruginosa requires two CRP/FNR-related transcriptional regulators, ANR and DNR. Activity of the consensus CRP- or FNR-dependent promoter in the anr and dnr deficient mutants was investigated. The CRP-dependent promoter was active in the mutant strains. Both regulators could activate the promoter with a consensus FNR-binding motif. DNR-dependent activation was nitrite-dependent, whereas activation by ANR was not, suggesting that only DNR is involved in sensing nitrogen oxides.

    Notes: We have investigated the shear-wave polarization anisotropy in the north-eastern Japan arc by using waveforms from many local earthquakes at various depths. We used a cross-correlation method to detect the shear-wave splitting. For intermediate-depth and deep events, the observed fast shear-wave oscillation directions (FSODs) are oriented parallel to the dip direction of the slab at the western stations, while they are oriented perpendicular to the dip direction at the eastern stations. These observations indicate that significant anisotropy exists in the mantle wedge, and that its nature is quite different between the eastern and the western parts.
