    Notes: Various aspects of the Nahm equations in three and seven dimensions are investigated. The residues of the variables at simple poles in the seven-dimensional case form an algebra. A large class of matrix representations of this algebra is constructed. The large N limit of these equations is taken by replacing the commutators by Moyal brackets, and a set of nontrivial solutions in a generalized form of Wigner distribution functions is obtained. © 1999 American Institute of Physics.

    Notes: Abstract The c-type cytochrome and protein profiles were compared for a number of cultures of Paracoccus denitrificans obtained from a range of culture collections. The cultures fell into two groups corresponding to the two original isolates of this bacterial species. One group, which included NCIMB 8944, ATCC 13543, ATCC 17741, ATCC 19367, Pd 1222 and DSM 413, were similar or identical to LMD 22.21. The second group, including DSM 65 and LMG 4218, were similar or identical to LMD 52.44. These groupings were not compatible with the recorded history of culture deposition. Mass spectrometry and amino acid sequence comparisons showed that the cytochrome c-550 of the LMD 52.44 culture group differed by 16% from that of the LMD 22.21 group, and yet was only 1% different from the cytochrome c-550 of Thiosphaera pantotropha. These results suggest that consideration should be given to creation of a new species of Paracoccus pantotropha, which would include Thiosphaera pantotropha and Paracoccus denitrificans LMD 52.44.

    Notes: Abstract A promoter probe shuttle vector suitable for the isolation of promoter elements from coryneform bacteria was constructed. This vector carried the neomycin phosphotransferase (NPTII) gene from transposon Tn 5 as a reporter gene, and was capable of replication in both Escherichia coli and Brevibacterium flavum. The vector was used in the construction of a B. flavum library of 899 independently isolated promoter clones. Promoters with a wide range of activities in B. flavum, including some very strong promoter elements, were isolated. Comparative analysis suggests that significant differences between B. flavum and E. coli may exist in the determinants of promoter strength.

    Notes: Salt extractable proteins were isolated from hand expressed juice, rag and peel of Valencia oranges. Each tissue extract was divided into dialysis supernatant (DS) and precipitate (DP) (forms during dialysis). DP, DS and DS heated for 2 min at 80°C (HDS) were added to pasteurized, reconstituted frozen concentrated orange juice at 5 U · mL−1 of pectinmethylesterase. Samples were incubated either at 25°C for 14 days or 4°C for 28 days and periodically sampled to determine the effects of tissue extracts on juice cloud stability. Tissue specific differences were observed for the rate of juice cloud precipitation and among the HDS, DS, and DP fractions of a given tissue. HDS fractions destabilized juice cloud more rapidly than DS or DP at both 25 and 4°C.

    Notes: In the finite-difference modeling of unsaturated infiltration by Richards’equation, standard interblock conductivity means like the arithmetic, geometric, and harmonic have been shown to produce flows different from true Darcian by up to orders of magnitude (Warrick 1991; Baker 1995a, 1998). This may have significant impact on models used for contaminant transport of nuclear and other hazardous waste. Previously, it has been impractical to calculate Darcian means in a running model. This paper introduces the analytic solution for Darcian means for the case of an exponential relative conductivity relation, and one possible approximation for general use, a piecewise Brooks-Corey Darcian (PBCD) mean. It compares the PBCD mean to the arithmetic mean in a modern mass-conservative numerical model (Celia et al. 1990) for vertical space steps from 0.8 to 105 cm, using soil parameters from Haverkamp et al. (1977). The results demonstrate that even in a model with mass balance good to one part in 108, non-Darcian flow errors are alive and well, costing up to 50% of the total mass infiltrated with the arithmetic mean and large model space steps. The new PBCD mean reduces that figure to 0.5% for this example soil.

    Notes: Both the U.S. Environmental Protection Agency (EPA) and the American Society for Testing and Materials (ASTM) provide guidance for selecting statistical procedures for ground water detection monitoring at Resource Conservation and Recovery Act (RCRA) solid and hazardous waste facilities. The procedures recommended for dealing with large numbers of nondetects, as may often be found in data for volatile organic compounds (VOCs), include, but are not limited to, Poisson prediction limits (in both the EPA guidance and ASTM standard) and Poisson tolerance limits (EPA guidance only). However, many of the proposed applications of the Poisson model are inappropriate. The development and application of the Poisson-based methods are explored for two types of data, counts of analytical hits and actual concentration measurements. Each of these two applications is explored along two lines of reasoning, a first-principles argument and a simple empirical fit.The application of Poisson-based methods to counts of analytical hits, including simultaneous consideration of multiple VOCs, appears to have merit from both a first principles and an empirical standpoint. On the other hand, the Poisson distribution is not appropriate for modeling concentration data, primarily because the variance of the distribution does not scale appropriately with changing units of measurement. Tolerance and prediction limits based on the Poisson distribution are not scale invariant. By changing the units of observation in example problems drawn from EPA guidance, use of the Poisson-based tolerance and prediction limits can result in significant errors. In short, neither the Poisson distribution nor associated tolerance or prediction limits should be used with concentration data. EPA guidance does present, however, other, more appropriate, methods for dealing with concentration data in which the number of nondetects is large. These include nonparametric tolerance and prediction limits and a test of proportions based on the binomial distribution.

    Notes: Discrete-depth ground water samplers are used during environmental screening investigations to collect ground water samples in lieu of installing and sampling monitoring wells. Two of the most commonly used samplers are the BAT Enviroprobe and the QED HydroPunch I, which rely on differing sample collection mechanics. Although these devices have been on the market for several years, it was unknown what, if any, effect the differences would have on analytical results for ground water samples containing low to moderate concentrations of chlorinated volatile organic compounds (VOCs). This study investigated whether the discrete-depth ground water sampler used introduces statistically significant differences in analytical results. Our goal was to provide a technical basis for allowing the two devices to be used interchangeably during screening investigations.Because this study was based on field samples, it included several sources of potential variability. It was necessary to separate differences due to sampler type from variability due to sampling location, sample handling, and laboratory analytical error. To statistically evaluate these sources of variability, the experiment was arranged in a nested design. Sixteen ground water samples were collected from eight random locations within a 15-foot by 15-foot grid. The grid was located in an area where shallow ground water was believed to be uniformly affected by VOCs. The data were evaluated using analysis of variance.This study indicates that when sampling for short-chain. chlorinated aliphatic compounds such as trichloroethylene, spatial variability from sampling location (over even a relatively small area) exceeds the differences between the Enviroprobe and HydroPuneh I. Therefore, the two sampling devices can be used interchangeably without reducing the internal consistency of the resulting data. However, when sampling for chlorinated aromatics, the choice of sampling device max introduce significant differences in the analytical results.

    Notes: Exfoliated skin was collected from bowriding dolphins with the use of a sterilized nylon scrub pad affixed to a wooden dowel. Initial tests of the effectiveness of the technique and dolphin behavioral responses were conducted on dusky dolphins off Kaikoura, New Zealand. During 14 sampling days, 128 contacts using this procedure were made with bowriding dolphins, of which 114 showed behavioral response. Responses during sampling were mild, with 11% of contacted individuals showing no visible response, and 66% of individuals which could be monitored for 30 sec after contact returning to bowride within 30 sec. Mean return time was 10 sec postcontact and did not vary significantly for groups of different sizes. Behavioral controls suggest that a proportion of responses could be explained by typical dolphin behavior in the presence of boats. Seventy-eight percent of contacts resulted in successful collection of tissue samples. Sample time was three minutes on average. Size of group and behavioral state did not appear to influence sample time. Preliminary genetic analyses revealed that tissue collected by this technique was suitable for amplification and sequencing of mitochondrial DNA (mtDNA) via PCR. Comparisons of mtDNA control region sequences with those from known L. obscurus and other delphinid samples verified that this technique was robust against contamination from elements in sea water.
