Notes: Abstract Grass puffer Fugu niphobles, captured in November 1986 at Shimoda, Japan, and then reared at different temperatures ranging from 10 to 29°C, was examined for microflora changes in the skin, gill and intestines. At 10°C, the skin and gill were colonized mainly by Pseudomonas, Flavobacterium and Moraxella, while at 29°C Vibrio and Flavobacterium appeared abundantly. The intestinal microflora, consisting of Vibrio, Pseudomonas and Flavobacterium, revealed little temperature dependence, although the intrageneric composition of Vibrio changed conspicuously depending on the change of water temperature. Vibrio group 1, tentatively identified as V. alginolyticus, was detected in all tissues examined at 20 and 29°C. This, along with the fact that V. alginolyticus produces tetrodotoxin, suggests that the tetrodotoxin contained in puffer is, at least partly, accounted for by this bacterial species inhabiting intestines.

Notes: We have isolated a mitochondrial rps7 gene from sugar beet (Beta vulgaris L. cv. TK81-O). It is present in a single genomic environment, with northern blot analysis showing transcription of this gene. Sequence analysis of RT-PCR-derived cDNA of rps7 showed that three RNA-editing events occurred on rps7 mRNAs. As far as we know, this is the first report of the complete identification of the RNA editing of a mitochondrial rps7 in the angiosperms.

Notes: Summary Localization of γ-aminobutyric acid (GABA) in the ventrolateral medulla oblongata of the rat was studied, using antisera directed against GABA molecule fixed to bovine serum albumin. Within the rostral portion of the ventrolateral medulla, GABA-like immunoreactive neurons were found in the lateral wing of the raphe magnus and in the region of the paragigantocellular reticular nucleus. In the caudal portion of the ventrolateral medulla, a lesser number of GABA-stained neurons were found in the region around the nucleus reticularis lateralis. GABA-like immunoreactive punctate structures were also found throughout the ventrolateral medulla. These results provide further evidence for the existence of GABAergic neurons in the ventrolateral medulla oblongata of the rat.

Notes: Abstract We have constructed a physical map of the mitochondrial DNA of a cytoplasmic male sterile (CMS) sugar beet line, TK81-MS, and compared it with that published for normal fertile sugar beet (cv. TK81-O) to clarify the differences between the CMS and normal mitochondrial genomes. The TK81-MS genome is present as a single circular molecule of 481.8 kb, or as two molecules of 184.9 and 296.9 kb. The CMS genome was found to be highly rearranged relative to the normal mitochondrial genome, with at least fifteen rearrangement and/or inversion events being required to align the two DNAs. Analysis of transcription patterns of known mitochondrial genes and rearranged regions revealed six genes, coxI, coxII, atpA, atp6, rps3, and orf324, whose expression is altered in the CMS line relative to the normal line. Of these six, only the coxI transcript pattern differs between male-sterile and fertility-restored genotypes, making it likely that the coxI locus is involved in mediating CMS in sugar beet.

Notes: Abstract Recessive alleles (va, va 1 , va 2 , etc) of the tobacco Va locus confer resistance to potato virus Y (PVY). To elucidate the mechanism underlying this resistance, we attempted to identify randomly amplified polymorphic (RAPD) markers that reveal polymorphism between two nearly isogenic lines (NILs) that differ in their susceptibility to PVY. Using each of 500 primers and 800 pairs of primers, we identified over 100 RAPD fragments that differed between the NILs. We applied these RAPD primers or primer combinations to an F2 population obtained from a cross between the susceptible line BY4 and the resistant va 2 -bearing NIL, F55. It was found that only 10 RAPD markers were polymorphic between resistant and susceptible plants. Unexpectedly, these markers were all linked to Va. All 10 RAPD markers were present in all 8 susceptible varieties tested. At least one RAPD marker was not detected in 8 out of 10 resistant varieties. Southern analysis revealed that the sequences of markers were not present in the genomes of resistant varieties, and the markers were found in individually distinct positions on the chromosomes of susceptible tobacco varieties. These results strongly suggest that the resistance conferred by va is due to deletions at the Va locus governing susceptibility to PVY.

Notes: Abstract The high-pressure and temperature equation of state of majorite solid solution, Mj0.8Py0.2, was determined up to 23 GPa and 773 K with energy-dispersive synchrotron X-ray diffraction at high pressure and high temperature using the single- and double-stage configurations of the multianvil apparatuses, MAX80 and 90. The X-ray diffraction data of the majorite sample were analyzed using the WPPD (whole-powder-pattern decomposition) method to obtain the lattice parameters. A least-squares fitting using the third-order Birch-Murnaghan equation of state yields the isothermal bulk modulus, K T0  = 156 GPa, its pressure derivative, K′ = 4.4(±0.3), and temperature derivative (∂K T /∂T) P = −1.9(±0.3)× 10−2 GPa/K, assuming that the thermal expansion coefficient is similar to that of pyrope-almandine solid solution.

Notes: Abstract  Single crystalline San Carlos olivine (1 mm cube) was transformed to (Mg,Fe)2SiO4β-phase at 13.5–15 GPa, 1030–1330 °C for 0–600 min using a multi-anvil high pressure apparatus. The α-β transformation occurred by incoherent surface nucleation and interface-controlled growth and recovered partially transformed samples showed sharply defined reaction rim. The growth rate of the β-phase rim significantly decreased with time and the growth eventually ceased. TEM observations revealed that many dislocations were created in both the relict olivine just near the α-β interface and the β-phase in the rim, which show evidence for deformation caused by interfacial stresses associated with the misfit elastic strain of the transformation. The observed tangled dislocation texture in β-phase suggested that the β-phase rim was hardened and relaxation of the interfacial stress was retarded. This probably caused a localized pressure drop in the relict olivine and decreased the growth rate. Time-dependent growth rates of β-phase is possibly controlled by the rheology of β-phase, which must be considered for the prediction of the olivine metastability in the subducting slabs.

Notes: Abstract: We have measured the dissociation of 8B in the Coulomb field of 208Pb at Ein=51.9 MeV/nucleon and extracted the cross section of the 7Be(p,γ)8B reaction at 0.4 ≤ Erel≤ 3 MeV, which is of importance for the 8B solar-neutrino production rate. The extracted astrophysical S17 factors are consistent with our earlier Coulomb dissociation measurement, and agree with the values deduced from the direct capture measurements by Filippone et al., Vaughn et al. and Hammache et al. The S factor at zero energy was extracted to be 18.9±1.8 eV-b with the help of theoretical energy-dependence.
