    Notes: Abstract  Fusion of mononucleated myoblasts into parallel arrays of mutinucleated myotubes is an essential step in skeletal myogenesis. The formation of such a highly ordered structure requires myoblasts to come together, orient and align in the correct location prior to fusion. We report here that fetal and neonatal myoblasts can use topographical features as strong guidance cues in vitro. Myoblasts were cultured on multiple grooved substrata of varying dimensions, and the axial orientations of individual cells were recorded. Both fetal and neonatal myoblasts aligned parallel with the direction of deep grooves (2.3–6.0 µm), which is correlated well with the location of myoblasts in similar sized grooves during secondary myogenesis. Fetal myoblasts also responded to shallower grooves (0.04–0.14 µm) by aligning parallel or perpendicular to the direction of the grooves, indicating the ability of these cells to respond to fine elements normally encountered within the developing muscle architecture. In contrast, neonatal myoblasts failed to respond to shallow grooves, adding to the suggestion that fetal and neonatal myoblasts may represent separate populations of myoblasts. Overall, the results demonstrate that myoblasts respond to large and small features of the physical topography in vitro and indicate that structural elements in the microenvironment of the muscle may play a critical role in myoblast spatial organization during myogenesis.

    Notes: Crystals have been grown of two of the domains of CD55. This is the first report of crystallization of a short consensus repeat (SCR) domain containing protein. CD55 is a widely expressed polymorphic glycoprotein, which functions as a complement regulator by inhibiting assembly and promoting destruction of C3 and C5 convertases. As a key regulator of complement, CD55 is implicated in the hyperacute rejection of xenografts from pigs into primates. It is also commonly hijacked as a receptor by viruses (e.g. medically important echoviruses and coxsackieviruses) and bacterial pathogens (e.g. certain pathogenic strains of Escherichia coli). Here, crystallization of a virus-binding fragment expressed in yeast, consisting of two of the four extracellular SCR domains of CD55, is reported. The recombinant domains have been crystallized in 30% polyethylene glycol (PEG), 0.2 M sodium acetate, 0.1 M sodium acetate trihydrate pH 4.6 using the sitting-drop vapour-diffusion method. Two crystal forms are observed (orthorhombic and monoclinic) and a native data set to 1.65 Å resolution has been collected from the monoclinic form at the Synchrotron Radiation Source, Daresbury, UK.

    Notes: This paper describes a model which was developed to simulate the effect of automated liming on the quality of a Welsh lake (Llyn Brianne) and its outflow river. The objective was to model the amount of lime required to dose the input streams under a variety of conditions. It was concluded that it would be feasible to install automatic-dosing equipment on the two input streams to deliver sufficient lime to ensure that the pH of the lake and water downstream was controlled at a pH value of 6.0 or greater, thus stabilizing water quality for fish and other aquatic biota.

    Notes: Human and environmental needs sometimes conflict: the more for the environment, the less for the people and vice versa. Environmental needs include soil water, wetlands, lakes, rivers and estuaries. The paper recognizes that all these aspects must be catered for, but focuses on the flow needs of rivers; in particular, it suggests a method to determine river-flow targets which are objective, sustainable, integrated with river-quality characteristics and economically justified. Such targets could help the Environment Agency to balance human and environmental water needs in a rational manner.
