    Description: Submyeloblative conditioning regimens for bone marrow transplantation (BMT) are desirable to reduce host toxicity in many settings. We previously demonstrated that hosts conditioned with 160 cGy or with two sequential doses of the antimetabolite 5-fluorouracil (5-FU) displayed ~50% and ~35% donor chimerism 6 months after transplantation of 20 x 106 fresh congenic bone marrow (BM) donor cells, respectively. Donor chimerism in 160 cGy- or 5-FU-conditioned mice was proportional to the reduction in host long-term repopulating ability (LTRA) caused by the conditioning regimen (~80–90% vs. ~55%, respectively). In this study, we sought to determine if donor marrow homing efficiency also correlated with long-term engraftment in these models of submyeloablative conditioning. We hypothesized that sequential doses of the cell cycle-specific agent 5-FU (5-FU x 2) may damage the marrow microenvironment less than low-dose radiation, resulting in more efficient homing in 5-FU x 2-conditioned mice. C57Bl6/J (Bl/6) hosts were conditioned with either 5-FU x 2 administered on days -5 and -1 (150 mg/kg IP) or with 160 cGy given the day of transplantation (day 0). Bl/6 (CD 45.2+) hosts were transplanted with 20 x 106 PKH-26 stained Bl/6 or with Boy J (CD 45.1+) whole BM cells via tail vein injection. The percentage of homed cells 20 hours after BMT in 5-FU x 2-conditioned mice was 3.53 % ± 3.1% (n = 13 from 3 independent experiments), while the percentage of homed cells in the 160 cGy group was 8.67 % ± 2.1% (n = 12; p = 0.0375). The fraction of donor cells which homed to the spleen or remained in the peripheral blood after 20 hours was similar between conditioning regimens. This difference in homing efficiency led us to consider other possible explanations as to why BM cells home less efficiently in 5-FU x 2-conditioned hosts than in low-dose irradiated mice. We hypothesized that 5-FU x 2 may have a more injurious effect on the marrow microenvironment than previously thought. Examination of marrow stromal cell potential using a colony-forming unit-fibroblast (CFU-F) assay revealed that marrow from 5-FU x 2-treated mice (n = 3) contained 1.1 ± 0.4 colonies/marrow femur equivalent (FE), whereas marrow from 160 cGy-treated mice (n = 3) contained 33.8 ± 13.4 colonies/FE. In comparison, marrow from untreated mice (n = 3) averaged 95.6 ± 27.5 colonies/FE. These data suggest that homing efficiency correlates with donor chimerism in these two models of submyeloablative conditioning, and establish at least two potential mechanisms (a lesser reduction of host LTRA, and decreased donor cell homing efficiency) for the lower level of long-term donor chimerism observed in 5-FU x 2- compared to 160 cGy-conditioned hosts. Furthermore, 5-FU x 2 conditioning appears to disrupt the marrow microenvironment to a greater degree than low-dose radiation. Current efforts are aimed at exploring the molecular interactions of homed donor cells with conditioned marrow.

    Notes: The synthesis of 1′-phenylthio-1-ferroceneboronic acid resulted in its trimerization with the loss of three molecules of water to form crystalline 2,4,6-tris(1′-phenylthio-1-ferrocenyl)boroxin, C48H39B3Fe3O3S3 or [Fe3(C15H12B3O3)(C11H9S)3]. The asymmetric unit contains two boroxin molecules conjugated with the attached cyclopentadienyl rings to form an aromatic ring. The cyclopentadienyl rings of five of the six ferrocenes are in an eclipsed conformation; the cyclopentadienyl rings of the other ferrocene are in the gauche form. Hydrolysis of the boroxin ring with aqueous base apparently is needed to produce the corresponding boronic acid required in a Suzuki coupling reaction.

    Notes: : The effects of molecular weights, concentrations and solvents of chitosan, soaking times, and soybeans/chitosan solution ratios on growth and quality of soybean sprouts were investigated. Among 5 chitosans, treatment with 493 kDa chitosan was the most effective in increasing total weight, vitamin C content, and hardness. Use of acetic acid rather than lactic acid as a chitosan solvent increased hypocotyl weight and vitamin C content. Soaking of soybeans in chitosan solution for 8 h increased total weight by 13% and vitamin C content by about 10%. Optimal cultivation conditions involve soaking in 0.05% chitosan with 493 kDa in 0.05% acetic acid for 8 h and a soybean/chitosan solution ratio of 1:4.

    Notes: : Physicochemical characteristics (molecular weight, nitrogen, ash, degree of deacetylation, bulk density, viscosity, and residual amino acids) of 6 commercial chitosans differed with products. Significant correlations were observed between molecular weight and viscosity and between nitrogen and degree of deacetylation. Binding capacities of water, fat, and three different dyes also varied depending on the products. Results of correlations between binding capacities and physicochemical characteristics indicated that both water and fat binding capacities were significantly negatively correlated with bulk density. In addition, fat binding capacity showed a significant correlation with viscosity. Significant negative correlations were observed between dye binding capacities of three red, yellow, and blue dyes and ash.

    Notes: Seven bacteria were isolated from spoiled tofu and identified as Bacillus sp. (S08), B. megaterium (S10), B. cereus (S17, S27, S28, S32), and Enterobacter sakazakii (S35). In a paper disc test with 6 chitosans and 6 chitosan oligomers of different molecular weights, chitosans showed higher antimicrobial activity than did chitosan oligomers at a 0.1% concentration. Results of inhibitory effects of 6 chitosans on growth of Bacillus sp. (S08) failed to detect viable cells after incubation for 24 hrs at 37 〈inlineGraphic alt="equal-or-greater" extraInfo="nonStandardEntity" href="urn:x-wiley:00221147:JFDS1511:eg" location="eg.gif"/〉C, even at 0.02% concentration. With B. megaterium (S10) and B. cereus (S27), a 3 to 4 log cycle reduction was found in the chitosan-treated group. The growth of Enterobacter sakazakii (S35) was completely suppressed in the presence of 0.04% chitosan except for 1 chitosan product. The minimum inhibitory concentration of chitosan differed with products and isolates, ranging from 0.005% to above 0.1%.
