    Notes: Abstract. Pharmacologic tools were used to identify receptors in functional studies by measuring either transepithelial current (I sc ) in strial marginal cells (SMC) or cAMP production in stria vascularis (SV). Further, receptors were identified in SV as transcripts by cloning and sequencing of reverse-transcriptase polymerase chain reaction (RT-PCR) products. Experiments were performed using tissues isolated from gerbils unless specified otherwise. I sc under control conditions was 1090 ± 21 μA/cm2 (n= 213) in gerbil SMC and 2001 ± 95 μA/cm2 (n= 6) in murine SMC. Direct stimulation of adenylate cyclase with 10-5 m forskolin but not with 10−5 m 1,9-dideoxy-forskolin resulted in an increase in the I sc by a factor of 1.14 ± 0.01 (n= 6). The vasopressin-receptor agonist 10−8 m Arg8-vasopressin had no significant effect on I sc in gerbil and murine SMC. The β-adrenergic agonists isoproterenol, norepinephrine and epinephrine stimulated I sc with an EC 50 of (6 ± 2) × 10−7 m (n= 28), (3 ± 1) × 10−6 m (n= 40) and (7 ± 2) × 10−6 m (n= 38), respectively. Isoproterenol stimulated cAMP production in SV with an EC 50 of (5 ± 2) × 10−7 m (n= 8). The β-antagonist 10−4 m propanolol completely inhibited 2 × 10−5 m isoproterenol-induced stimulation of I sc . The β-antagonists atenolol, ICI118551 and CGP20712A inhibited isoproterenol-induced stimulation of I sc with a K DB of 1 × 10−7 m (pK DB = 6.96 ± 0.15, n= 14), 1 × 10−7 m (pK DB = 7.01 ± 0.14, n= 15), 2 × 10−9 m (pK DB = 8.73 ± 0.13, n = 19), respectively. CGP20712A inhibited isoproterenol-induced cAMP production with a K DB of 1 × 10−10 m (pK DB = 9.94 ± 0.55, n= 9). RT-PCR of total RNA isolated from SV using primers specific for the β1-, β2- and β3-adrenergic receptors revealed products of the predicted sizes for the β1- and β2- but not the β3-adrenergic receptor. Sequence analysis confirmed that amplified cDNA fragments encoded gene-specific nucleotide sequences. These results demonstrate that K+ secretion in SMC is under the control of β1-adrenergic receptors but not β2-adrenergic or vasopressin-receptors and that the β1-subtype is the primary β-adrenergic receptor in SV although SV contains transcripts for both β1- and β2-adrenergic receptors.

    Notes: Abstract The dtaAX gene encoding a pyridoxal 5′-phosphate (pyridoxal-P)-dependent low-specificity d-threonine aldolase was cloned from the chromosomal DNA of Alcaligenes xylosoxidans IFO 12669. It contains an open reading frame consisting of 1,134 nucleotides corresponding to 377 amino acid residues. The predicted amino acid sequence displayed 54% identity with that of d-threonine aldolase from gram-positive bacteria Arthrobacter sp. DK-38, but showed no significant similarity with those of other known pyridoxal-P enzymes. This gram-negative bacterial enzyme was highly overproduced in recombinant Escherichia coli cells, and the specific activity of the enzyme in the cell extract was as high as 18 U/mg (purified enzyme 38.6 U/mg), which was 6,000 times higher than that from the wild-type Alcaligenes cell extract. The recombinant enzyme was thus feasibly purified to homogeneity by ammonium sulfate fractionation and DEAE-Toyopearl chromatography steps. The recombinant low-specificity d-threonine aldolase was shown to be an efficient biocatalyst for resolution of l-β-3,4-methylenedioxyphenylserine, an intermediate for production of a therapeutic drug for Parkinson's disease.

    Notes: Abstract. We have synthesized the ytterbium fulleride compounds YbxC60 (x=1–6) at a range of lower temperature (530 °C–570 °C). Except for the nominal composition Yb3C60 compound, which has been reported in the references, we also obtained a new phase Yb6C60, whose structure is orthorhombic with space group P21212 and unit cell parameters a=21.28 Å, b=20.13 Å and c=15.89 Å. The YbxC60 compounds are sensitive to oxygen, the samples exposed in air convert to an amorphous phase and Yb2O3 compound.

    Notes: Identifies the intermetallics formed in Sn-0.5Cu-3.5Ag and Sn-3.5Ag-0.5Cu-0.5B (wt%) lead-free solders, and the influence of boron on these precipitates. SEM, TEM and SIMS were employed to reveal the difference of microstructure in both solder alloys. It was found that the intermetallics formed were Ag3Sn and Cu6Sn5. Both solders were found to have a dispersion structure of Ag3Sn particles with network-shaped subgrains. The microstructure of Ag3Sn was also found to become finer and more uniform in Sn-3.5Ag-0.5Cu-0.5B solder, due to the addition of boron, while there was little effect from the boron on the Cu6Sn5 phase.

    Notes: An electron crystallographic image-processing technique based on the combination of high-resolution electron microscopy and electron diffraction has been developed to investigate the commensurate structural modulation in the high-Tc superconductor (Pb0.5Sr0.3Cu0.2)Sr2(Ca0.6Sr0.4)Cu2Oy. After symmetry averaging, a structure image was obtained by image deconvolution at the resolution limited by that of the electron microscope. Then phase extension was employed to enhance the image resolution up to about 1.25 Å by means of the electron diffraction data corrected with an empirical method. In the final projected potential map, the occupational and/or positional modulation is clearly observed for all atoms, including oxygen. The key points of determining superstructures by the technique are studied and discussed.

    Notes: : The characteristics of polysaccharides isolated from milk and soymilk kefir grains, and the composition of flavor and volatile compounds from soymilk kefir were investigated. Soymilk kefir grains revealed lower polysaccharide content than milk kefir grains, with a polysaccharide profile consisting primarily of glucose and galactose, with the former predominating. The apparent molecular weight of the polysaccharide was estimated at 1.7 × 106 Da. In comparison to nonfermented soymilk, the concentrations of the key volatile compounds for soymilk kefir (such as acetaldehyde, acetone, diacetyl, and ethanol) increased, while n-hexanal concentration decreased. The acetaldehyde level for soymilk kefir was slightly higher, but levels for the other volatile compounds were lower than for milk kefir.

    Notes: The thermal stability of NiSi films in Ni/Pt/(111)Si and Ni/Pt/(100)Si bilayered systems was investigated. In the Ni/Pt/(111)Si system, the NiSi films formed at 640 °C have an epitaxial relationship of (100)NiSi(parallel)(111)Si and [01¯0]NiSi(parallel)[011¯]Si with the substrates. On the other hand, those formed on (100)Si substrates were polycrystalline although the experimental parameters are the same as those in Ni/Pt/(111)Si samples. In both cases, NiSi and PtSi formed a solid solution Ni1−xPtxSi following Vegard's law. While the thermal stability of NiSi films is improved in both cases compared with the Ni/Si system, the thermal stability of the textured NiSi films formed on (111)Si substrates is higher than their polycrystalline counterparts formed on (100)Si substrates. This is attributed to the reduced interfacial energy and consequently increased activation energy for the nucleation of NiSi2 due to the formation of textured NiSi films on (111)Si substrates. At 900 °C, the Ni(Pt)Si film formed in Ni/Pt/(100)Si system decomposed into separate phases of NiSi and PtSi with noticeable lattice deformation. Differences between the texture evolution of NiSi films in Ni/Pt/Si and Ni(Pt) alloy/Si systems were also reported and explained from both kinetic and thermodynamic aspects. © 2001 American Institute of Physics.
