Notes: Abstract Particle image velocimetry (PIV) has been used in order to measure the three mean components and turbulence intensities of the velocity vector in a swirling decaying flow induced by a tangential inlet in an annulus. This kind of flow motion is found to be very complex, exhibiting three-dimensional and non-axisymmetric characteristics coupled with a free decay of the swirling intensity along the flow path, thereby making it difficult to study. A method allowing the measurement of the three components of the velocity flow-field with a standard PIV system with two-dimensional acquisitions, is presented. The evolution of each velocity component between the inlet and the outlet of the annulus is obtained. Furthermore, the PIV technique is extended to the measurement of turbulent characteristics such as turbulent intensities and dimensionless turbulent energy. The main characteristics of the swirling flow are discussed and the swirl number is estimated as a function of the axial distance from the tangential inlet.

Notes: R-phycoerythrin, a light-harvesting component from the red algae Gracilaria chilensis, was crystallized by vapour diffusion using ammonium sulfate as precipitant agent. Red crystals grew after one week at 293 K and diffracted to 2.70 Å resolution. Three serial macroseeding assays were necessary to grow a second larger crystal to dimensions of 0.68 × 0.16 × 0.16 mm. This crystal diffracted to 2.24 Å resolution using synchrotron radiation at beamline BM14 of the European Synchrotron Radiation Facility (ESRF) at Grenoble, France and was used for structure determination. Data were collected at 100 K to a completeness of 98.6%. The crystal was trigonal, space group R3, with unit-cell parameters a = b = 187.3, c = 59.1 Å, α = β = 90, γ = 120°. Data treatment using the CCP4 suite of programs indicated that the crystal was twinned (〈I2〉/〈I〉2 = 1.41). Molecular replacement was performed with AMoRe using the R-phycoerythrin from Polysiphonia urceolata [Chang et al. (1996), J. Mol. Biol. 249, 424–440] as a search model. In order to overcome the twinning problem, SHELX97 was used for the crystallographic refinement. The twin fraction was 0.48, indicating a nearly perfect hemihedrally twinned crystal. The final Rwork and Rfree factors are 0.16 and 0.25, respectively. All the residues and chromophores of the α- and β-chains are well defined in the electron-density maps. Some residues belonging to the γ-linker are also recognizable.

Notes: : An ultra high-pressure homogenizer (20 to 350 MPa) was used to realize fine food emulsions stabilized by soy proteins. The first aim of the work was to understand how dynamic high-pressure processing affects soybean globulin conformation. Then, the effect of homogenizing pressure on the emulsions structure and rheology was investigated. High-pressure homogenization caused denaturation of proteins due to strong mechanical forces and high temperatures encountered in the valve. Droplet sizes of emulsions were greatly reduced with high-pressure homogenization and Newtonian liquid emulsions were converted into shear-thinning emulsion gels by homogenization at pressures above 250 MPa. Hydrophobic interactions between proteins were supposed to cause the gel-like network structure of emulsions.

Notes: [Auszug] The crystal structure of the tetrameric α2β2 acetyl-coenzyme A synthase/carbon monoxide dehydrogenase from Moorella thermoacetica has been solved at 1.9 Å resolution. Surprisingly, the two α subunits display different (open and closed) conformations. Furthermore, X-ray data ...

Notes: Techniques to obtain optimal value from existing hydrogeological information are neglected in many ground water contamination studies, resulting in great costs to the public. Data collection and routine interpretations should not be good enough for hydrogeologists, who should be inherently adept at keen early perspectives and applying useful generalizations. Techniques are available to obtain and explain expanded knowledge from piecemeal and imprecise information at the early stages of site studies. Scarcity of precise data should not be a deterrent. This needed mindset should precede the prevailing mindset of initial collection of extensive new data, followed by data interpretation and mathematical modeling. The existing mindset is often guided by rigid regulations. The procedure of interlocking the past with the present to predict the future through the systematic use of generalizations and retrodictive inferences builds a prior conceptual model explanation (PCME). This early PCME is an organized, mental picture or discernment of a hydrogeological setting displaying three-dimensional hydrogeologic features, the discernment being capable of explaining in narrative form the general fluid mechanics and contaminant behavior within imprecise but reasonable bounds. This characterization is logically followed by possible cause-and-effect relations and predictions of extended consequences of natural and human actions. A site PCME has sound support from the cross-checking of interrelated factors that leads to almost interlocking proofs.

Notes: Candida albicans is the single, most frequently isolated human fungal pathogen. As with most fungal pathogens, the factors which contribute to pathogenesis in C. albicans are not known, despite more than a decade of molecular genetic analysis. Candida albicans was thought to be asexual until the discovery of the MTL loci homologous to the mating type (MAT) loci in Saccharomyces cerevisiae led to the demonstration that mating is possible. Using Candida albicans mutants in genes likely to be involved in mating, we analysed the process to determine its similarity to mating in Saccharomyces cerevisiae. We examined disruptions of three of the genes in the MAPK pathway which is involved in filamentous growth in both S. cerevisiae and C. albicans and is known to control pheromone response in the former fungus. Disruptions in HST7 and CPH1 blocked mating in both MTLa and MTLα strains, whereas disruptions in STE20 had no effect. A disruption in KEX2, a gene involved in processing the S. cerevisiae pheromone Mfα, prevented mating in MTLα but not MTLa cells, whereas a disruption in HST6, the orthologue of the STE6 gene which encodes an ABC transporter responsible for secretion of the Mfa pheromone, prevented mating in MTLa but not in MTLα cells. Disruption of two cell wall genes, ALS1 and INT1, had no effect on mating, even though ALS1 was identified by similarity to the S. cerevisiae sexual agglutinin, SAG1. The results reveal that these two diverged yeasts show a surprising similarity in their mating processes.

Notes: Cobalt nanocrystal three dimensional (3D) arrays were fabricated by deposition of a ferrofluid on a highly oriented pyrolitic graphite substrate and by applying a magnetic field parallel to the substrate during the deposition process. Depending on the strength of the magnetic field, regular stripes of cobalt nanocrystals are obtained. The influence of the deposition process parameters was investigated. The magnetic properties of these 3D arrays are discussed in terms of orientation of the easy axes and ferromagnetic domain formation. © 2002 American Institute of Physics.

Notes: We report the self-formation of nanometer-size silicon islands on a silicon-on-insulator (SOI) substrate that is associated with simple thermal treatment in the range of 500–900 °C. We study the island formation process versus the temperature of the thermal annealing, the thickness of the top silicon layer, and the presence of a native oxide on this top layer. The island size distribution is also studied. To follow the chemical evolution of the top layer, we used in situ Auger electron spectroscopy in an ultrahigh vacuum chamber. The island morphology is studied using ex situ atomic force microscopy (AFM). The formation temperature increases with the thickness of the top silicon layer and can be explained by thermal stress induced at the Si/SiO2 interface. From a technological point of view, this study shows the limitation of a SOI substrate with a thin silicon top layer under thermal treatment. On the other hand, it opens up an easy way in which to build silicon dots on an insulator. Finally, we present preliminary data that show the possibility of charging these nanocrystallites with an AFM tip. © 2002 American Institute of Physics.
