    Notizen: Through the principal use of the reduced magnetohydrodynamic version of the finite aspect ratio code [L. A. Charlton et al., J. Comput. Phys. 86, 270 (1990)], an m/n=1/1 resistive kink mode was poloidally rotated with the accompanying rotational shear. It was observed that the growth rate of this unstable mode can either decrease or increase as the applied equilibrium rotation is increased to near poloidal sonic speeds. Shear in the poloidal rotation profile is stabilizing, but only if the destabilizing effects of bulk rotation can be overcome. Therefore, the mode's stability was sensitive to the location of the rotation's peak relative to the eigenmode's spatial extent. The destabilizing effects of bulk rotation are apparently a rotationally enhanced beta, and the stabilizing effects appear to be caused by exceeding a critical rotational shear spatially averaged over the eigenmode structure. © 2001 American Institute of Physics.

    Notizen: The theory of beat wave excitation in a slightly inhomogeneous plasma is presented. While the theory is general it applies directly to the experiments performed in Alaska by the ionospheric heating facilities HIPAS [High Power Auroral Simulation, Radio Sci. 25, 1269 (1990)] and HAARP [High Frequency Active Auroral Research Project, Geophys. Res. Lett. 25, 257 (1998)]. A ray tracing (WKB) formalism appropriate for computations is developed. The computational implementation of this formalism and extensive results will be presented in a follow up to this paper. Also the beat wave excitation of upper-hybrid waves is investigated analytically. The complicated trajectories of these waves in the plasma are described. When this beat wave reaches the plasma wave resonance it can, in the HIPAS–HAARP experiments, attain nonlinear amplitude. The electrostatic upper-hybrid waves are trapped around the density maximum of the ionosphere. This trapping is investigated in detail. Beat wave pumping of the trapped modes is possible using HAARP and HIPAS or with split beams from HAARP. © 2001 American Institute of Physics.

    Notizen: Abstract The Myc/Max/Mad network comprises a group of transcription factors whose distinct interactions result in gene-specific transcriptional activation or repression. A great deal of research indicates that the functions of the network play roles in cell proliferation, differentiation, and death. In this review we focus on the Myc and Mad protein families and attempt to relate their biological functions to their transcriptional activities and gene targets. Both Myc and Mad, as well as the more recently described Mnt and Mga proteins, form heterodimers with Max, permitting binding to specific DNA sequences. These DNA-bound heterodimers recruit coactivator or corepressor complexes that generate alterations in chromatin structure, which in turn modulate transcription. Initial identification of target genes suggests that the network regulates genes involved in the cell cycle, growth, life span, and morphology. Because Myc and Mad proteins are expressed in response to diverse signaling pathways, the network can be viewed as a functional module which acts to convert environmental signals into specific gene-regulatory programs.

    Notizen: The fish community of the small (17·5 ha) intermittently open East Kleinemonde estuary was sampled between 1994 and 1997 to estimate population size, standing stock, growth and productivity. The estuarine-spawning species were numerically more abundant (n c. 750 000) but due to their small size contributed only 11·7% to the total biomass. The total annual productivity of all fishes in the estuary (n c. 890 000), with a standing stock of 28·44 g m −2, was calculated at 55·89 g m−2 year−1. The small sparid Rhabdosargus holubi with a production estimate of 41·35 g m−2 year−1 accounted for 〈74% of the total fish production in this estuary.

    Notizen: Summary The function of the Mycobacterium tuberculosis eukaryotic-like protein serine/threonine kinase PknG was investigated by gene knock-out and by expression and biochemical analysis. The pknG gene (Rv0410c), when cloned and expressed in Escherichia coli, encodes a functional kinase. An in vitro kinase assay of the recombinant protein demonstrated that PknG can autophosphorylate its kinase domain as well as its 30 kDa C-terminal portion, which contains a tetratricopeptide (TPR) structural signalling motif. Western analysis revealed that PknG is located in the cytosol as well as in mycobacterial membrane. The pknG gene was inactivated by allelic exchange in M. tuberculosis. The resulting mutant strain causes delayed mortality in SCID mice and displays decreased viability both in vitro and upon infection of BALB/c mice. The reduced growth of the mutant was more pronounced in the stationary phase of the mycobacterial growth cycle and when grown in nutrient-depleted media. The PknG-deficient mutant accumulates glutamate and glutamine. The cellular levels of these two amino acids reached approximately threefold of their parental strain levels. Higher cellular levels of the amine sugar-containing molecules, GlcN-Ins and mycothiol, which are derived from glutamate, were detected in the ΔpknG mutant. De novo glutamine synthesis was shown to be reduced by 50%. This is consistent with current knowledge suggesting that glutamine synthesis is regulated by glutamate and glutamine levels. These data support our hypothesis that PknG mediates the transfer of signals sensing nutritional stress in M. tuberculosis and translates them into metabolic adaptation.

    Notizen: The predominant filamentous bacteria which are found in bulking activated sludge in the UK have been assessed from a microscopic analysis of samples taken over a three-year period. There were major differences in the floc biology of samples received from plants treating predominantly domestic wastewaters and those of industrial origin. Most samples (84%) of domestic origin were dominated by two filament types: Microthrix parvicella was the dominant filament in 63% of the samples and Type 021N dominated in 21% of the samples. Nostocoida limicola and Sphaerotilus natans dominated in the remaining 16% of samples. The samples of industrial origin showed much greater filament diversity, with eight filament types routinely observed in high numbers. Type 021N was the most prevalent and M. parvicella was not observed in the industrial samples. Many domestic samples from sequencing batch reactors were dominated by M. parvicella. Based upon knowledge of the predominant filament type from microscopic examination. it was possible to identify the likely causes for their proliferation and suggest long-term solutions to achieve their eradication.

    Notizen: Polyamine levels and activities of enzymes of polyamine biosynthesis and catabolism were examined in the barley cultivar Delibes (Ml1al + Ml(Ab)) reacting hypersensitively to the powdery mildew fungus, Blumeria graminis f. sp. hordei (race CC220). Levels of free putrescine and spermine and of conjugated forms of putrescine, spermidine and spermine were greatly increased 1–4 d following inoculation of barley with the powdery mildew. These changes in polyamine levels were accompanied by elevated activities of the polyamine biosynthetic enzymes ornithine decarboxylase (ODC), arginine decarboxylase (ADC) and S-adenosylmethionine decarboxylase (AdoMetDC) and the polyamine catabolic enzymes diamine oxidase (DAO) and polyamine oxidase (PAO). Activities of two enzymes involved in conjugating polyamines to hydroxycinnamic acids, putrescine hydroxycinnamoyl transferase (PHT) and tyramine feruloyl-CoA transferase (TFT) were also examined and were found to increase significantly 1–4 d after inoculation. The possibility that the increased levels of free spermine, increased polyamine conjugates, and increased DAO and PAO activities are involved in development of the hypersensitive response of Delibes to powdery mildew infection is discussed.

    Notizen: Five transposon mutants of Francisella novicida were isolated that are compromised in their ability to grow in mouse macrophages in vitro. Sequence analysis of the DNA flanking the transposon insertions identified the genes that were interrupted in these mutants. One of the inactivated loci corresponds to the Francisella tularensis gene that encodes a 23-kDa protein that is the most prominently induced protein following macrophage infection. Another insertion was localised to approximately 2 kb upstream of the gene encoding the 23-kDa protein. By analysis of the incomplete Francisella genome sequence it was surmised that these two insertions disrupt different portions of a putative operon that encodes four proteins, none of which have discernible functions. Three other interrupted loci associated with poor intramacrophage growth showed similarity at the deduced amino acid level to alanine racemase, the ClpB heat-shock protease, and the purine biosynthetic enzyme, glutamine phosphoribosylpyrophosphate amidotransferases.
