    Description: Lung disease is the major cause of morbidity and mortality in cystic fibrosis, an autosomal recessive disease caused by mutations in CFTR. In cystic fibrosis, chronic infection and dysregulated neutrophilic inflammation lead to progressive airway destruction. The severity of cystic fibrosis lung disease has considerable heritability, independent of CFTR genotype. To identify genetic modifiers, here we performed a genome-wide single nucleotide polymorphism scan in one cohort of cystic fibrosis patients, replicating top candidates in an independent cohort. This approach identified IFRD1 as a modifier of cystic fibrosis lung disease severity. IFRD1 is a histone-deacetylase-dependent transcriptional co-regulator expressed during terminal neutrophil differentiation. Neutrophils, but not macrophages, from Ifrd1-deficient mice showed blunted effector function, associated with decreased NF-kappaB p65 transactivation. In vivo, IFRD1 deficiency caused delayed bacterial clearance from the airway, but also less inflammation and disease-a phenotype primarily dependent on haematopoietic cell expression, or lack of expression, of IFRD1. In humans, IFRD1 polymorphisms were significantly associated with variation in neutrophil effector function.     Description: Schistosoma mansoni is responsible for the neglected tropical disease schistosomiasis that affects 210 million people in 76 countries. Here we present analysis of the 363 megabase nuclear genome of the blood fluke. It encodes at least 11,809 genes, with an unusual intron size distribution, and new families of micro-exon genes that undergo frequent alternative splicing. As the first sequenced flatworm, and a representative of the Lophotrochozoa, it offers insights into early events in the evolution of the animals, including the development of a body pattern with bilateral symmetry, and the development of tissues into organs. Our analysis has been informed by the need to find new drug targets. The deficits in lipid metabolism that make schistosomes dependent on the host are revealed, and the identification of membrane receptors, ion channels and more than 300 proteases provide new insights into the biology of the life cycle and new targets. Bioinformatics approaches have identified metabolic chokepoints, and a chemogenomic screen has pinpointed schistosome proteins for which existing drugs may be active.     Description: We investigated extraneural manifestations in scrapie-infected transgenic mice expressing prion protein lacking the glycophosphatydylinositol membrane anchor. In the brain, blood, and heart, both abnormal protease-resistant prion protein (PrPres) and prion infectivity were readily detected by immunoblot and by inoculation into nontransgenic recipients. The titer of infectious scrapie in blood plasma exceeded 10(7) 50% infectious doses per milliliter.     Description: The Rho guanosine triphosphatases (GTPases) Rac1 and Rac2 are critical signaling regulators in mammalian cells. The deletion of both Rac1 and Rac2 murine alleles leads to a massive egress of hematopoietic stem/progenitor cells (HSC/Ps) into the blood from the marrow, whereas Rac1-/- but not Rac2-/- HSC/Ps fail to engraft in the bone marrow of irradiated recipient mice. In contrast, Rac2, but not Rac1, regulates superoxide production and directed migration in neutrophils, and in each cell type, the two GTPases play distinct roles in actin organization, cell survival, and proliferation.     Notes: A novel method for characterizing a high actuating capability light-weight actuator isdescribed and analyzed. The actuator comprises a truss core and smart face-sheets which result inbending. The deformation of this beam actuator is obtained by changing voltage inside face-sheetsmade from smart material. Materials selections are discussed for practice, and then optimization isdesigned to ascertain minimum weight or maximum achievable displacement subject to two typesof constrains. Results show the composite trusses have higher stiffness and strength than metaltrusses. Comparisons with two corrugated core actuators, the truss-core design has better actuatingcapability at specified weight

    Notes: The direct methods of breaking the phase ambiguity intrinsic in one-wavelength anomalous scattering (OAS) data and MAD phasing are powerful methods in their own rights. In a different context, in addition to their success in phasing OAS data, direct methods can also be useful in the treatment of MAD data. The idea has been tested with the MAD data at 2.5 Å resolution from the protein human adenosine kinase [Mathews et al. (1998), Biochemistry, 37, 15607–15620]. The results showed that the incorporation of direct methods in MAD phasing led to a significant improvement of phases over those obtained from the conventional MAD phasing method alone, as indicated by improved map correlation coefficients (with the existing model), reduced phase errors by 4.5° and improved map connectivity.

    Notes: Direct methods of breaking phase ambiguities in protein crystallography have been introduced in powder diffraction analysis. This is aiming at ab initio solution of noncentrosymmetric structures using two-wavelength anomalous powder diffraction data. The known structure of the hydrogen bromide salt of leotidine (C14H20O2N2·HBr) in space group P212121 was used for simulating two-wavelength anomalous powder diffraction with the Br atom as anomalous scatterer. X-ray wavelengths are selected at \lambda_1 = 0.920 and \lambda_2 = 1.500 Å. Unique reflections from the diffraction pattern of \lambda_2 were able to locate the Br atom accurately. All overlapping diffraction peaks were uniformly partitioned to decompose into single reflections. Structure-factor amplitudes were then extracted. With these and the substructure of Br atoms, unique phases for centric reflections (hk0, h0l and 0kl) and phase doublets for noncentric reflections were obtained. The direct method was used to break the phase ambiguity leading to an interpretable electron-density map, from which five cycles of Fourier iteration yielded the complete structure.

    Notes: An epizootic in pond cultured sea bass, Lateolabrax japonicus, was caused by Nocardia sp. in Taiwan, in September and October 1997. The cumulative mortality within 1 month was 17.5% (3500 out of 20 000 fish) and diseased fish were 7 months old with total lengths from 25 to 30 cm. Multiple, yellowish white nodules, 0.1–0.2 cm in diameter, were scattered in the gill, heart, liver, spleen and kidney. Histopathologically, typical granulomatous lesions appeared in those organs. The morphology of isolated bacteria from brain heart infusion (BHI) medium or Lowenstein–Jensen medium (LJM) were bead-like filaments, as shown by Ziehl-Neelsen's (ZN) staining method. The gross lesion and histopathological changes found in experimentally infected fish were similar to those in naturally infected fish. Based on the growth characteristics, morphological and biochemical properties of the bacterium, and histopathological changes, the isolated bacteria were identified as Nocardiaseriolae. This is the first report of N. seriolae-infected sea bass in aquaculture.
