    Description: Soil moisture–atmosphere feedbacks play an important role in the regional climate over many regions worldwide, not only for the mean climate but also for extreme events. Several studies have shown that the extent and severity of droughts and heat waves can be significantly impacted by dry or wet soil moisture conditions. To date, the impact of soil moisture on heavy rainfall events has been less frequently investigated. Thus, we consider the role of soil moisture in the formation of heavy rainfall using the Oder flood event in July 1997 as an example. Here, we used the regional climate model CCLM as an uncoupled standalone model and the coupled COSTRICE system, where CCLM is coupled with an ocean and a sea ice model over the Baltic and North Sea regions. The results from climate simulations over Europe show that the coupled model can capture the second phase (18–20 July) of heavy rainfall that led to the Oder flood, while the uncoupled model does not. Sensitivity experiments demonstrate that the better performance of the coupled model can be attributed to the simulated soil moisture conditions in July 1997 in Central Europe, which were wetter for the coupled model than for the uncoupled model. This finding indicates that the soil moisture preceding the event significantly impacted the generation of heavy rainfall in this second phase. The better simulation in the coupled model also implies the added value that the atmosphere–ocean coupling has on the simulation of this specific extreme event. As none of the model versions captured the first phase (4–8 July), despite the differences in soil moisture, it can be concluded that the importance of soil moisture for the generation of heavy rainfall events strongly depends on the event and the general circulation pattern associated with it.     Description: Ovarian cancer is the leading cause of death among gynecological cancers, but is poorly amenable to preoperative diagnosis. In this study, we investigate the feasibility of “optical biopsy,” using high-optical-resolution photoacoustic microscopy (OR-PAM) to quantify the microvasculature of ovarian and fallopian tube tissue. The technique is demonstrated using excised human ovary and fallopian tube specimens imaged immediately after surgery. Quantitative parameters are derived using Amira software. The parameters include three-dimensional vascular segment count, total volume and length, which are associated with tumor angiogenesis. Qualitative results of OR-PAM demonstrate that malignant ovarian tissue has larger and more tortuous blood vessels as well as smaller vessels of different sizes, while benign and normal ovarian tissue has smaller vessels of uniform size. Quantitative analysis shows that malignant ovaries have greater tumor vessel volume, length and number of segments, as compared with benign and normal ovaries. The vascular pattern of benign fallopian tube is different than that of benign ovarian tissue. Our initial results demonstrate the potential of OR-PAM as an imaging tool for fast assessment of ovarian tissue and fallopian tube and could avoid unnecessary surgery if the risk of the examined ovary is extremely low.

P elements of two different subfamilies designated as M- and O-type are thought to have invaded host species in the Drosophila obscura group via horizontal transmission from external sources. Sequence comparisons with P elements isolated from other species suggested that the horizontal invasion by the O-type must have been a rather recent event, whereas the M-type invasion should have occurred in the more distant past. To trace the phylogenetic history of O-type elements, additional taxa were screened for the presence of O- and M-type elements using type-specific PCR primers. The phylogeny deduced from the sequence data of a 927-bp section (14 taxa) indicate that O-type elements have undergone longer periods of regular vertical transmission in the lineages of the saltans and willistoni groups of Drosophila. However, starting from a species of the D. willistoni group they were transmitted horizontally into other lineages. First the lineage of the D. affinis subgroup was infected, and finally, in a more recent wave of horizontal spread, species of three different genera were invaded by O-type elements from the D. affinis lineage: Scaptomyza, Lordiphosa, and the sibling species D. bifasciata/D. imaii of the Drosophila obscura subgroup. The O-type elements isolated from these taxa are almost identical (sequence divergence 〈1%). In contrast, no such striking similarities are observed among M-type elements. Nevertheless, the sequence phylogeny of M-type elements is also not in accordance with the phylogeny of their host species, suggesting earlier horizontal transfer events. The results imply that P elements cross species barriers more frequently than previously thought but require a particular genomic environment and thus seem to be confined to a rather narrow spectrum of host species. Consequently, different P element types acquired by successive horizontal transmission events often coexist within the same genome.

    Notes: Abstract The Jupiter gold deposit in the northeastern Eastern Goldfields Province of the Yilgarn Craton of Western Australia is hosted in greenschist facies metamorphosed tholeiitic basalt, quartz–alkali-feldspar syenite, and quartz–feldspar porphyry. Syenite intrudes basalt as irregularly shaped dykes which radiate from a larger stock, whereas at least three E–W and NE–SW striking quartz–feldspar porphyries intrude both syenite and basalt. Brittle–ductile shear zones are shallow-dipping, NW to NE striking, or are steep-dipping to the south and west. Quartz ± carbonate veins that host gold at Jupiter occur in all lithologies and are divided into: (1) veins that are restricted to the shear zones, (2) discrete veins that are subparallel to shear zone-hosted veins, and (3) stockwork veins that form a network of randomly oriented microfractures in syenite wallrock proximal to shallow-dipping shear zones. The gold-bearing veins comprise mainly quartz, calcite, ankerite, and albite, with minor sericite, pyrite, chalcopyrite, galena, sphalerite, molybdenite, telluride minerals, and gold. Proximal hydrothermal alteration zones to the mineralised veins comprise quartz, calcite, ankerite, albite, and sericite. High gold grades (〉2 g/t Au) occur mainly in syenite and in the hanging walls to shallow-dipping shear zones in syenite where there is a greater density of mineralised stockwork veins. The Jupiter deposit has structural and hydrothermal alteration styles that are similar to both granitoid-hosted, but post-magmatic Archaean lode-gold deposits in the Yilgarn Craton and intrusion-related, syn-magmatic, syenite-hosted gold deposits in the Superior Province of Canada. Based on field observations and petrologic data, the Jupiter deposit is considered to be a post-magmatic Archaean lode-gold deposit rather than a syn-intrusion deposit.

    Notes: Recent advances in the Internet have afforded professional accounting firms a form of promotion to enhance their image as technologically competent. Currently, over 2,000 accounting firms have Web sites registered with The List of CPA Firms Directory. The objective of this paper is to conduct a preliminary content analysis of the web pages of accounting firms. A total of 346 Web sites were randomly selected. The contents of the Web pages were examined and analysed for inclusion of client choice factors, basic company information, value added features, and graphics. The registration of the accounting firms' Web sites by search engines, Web yellow pages, CPA directories, and state CPA societies was also investigated. Generally, firms did a good job of providing basic client desired information about the firm, but they must register their Web sites with search engines that are commonly available and must encourage more interactivity with existing and potential clients by providing free information such as newsletters and links to relevant Web sites. They need to develop greater expertise in using the specific features and advantages of the Internet medium.

    Notes: The iron deficiency-dependent regulation of isiAB transcription in Synechocystis sp. PCC 6803 was analyzed by fusion of modified isiAB promoter fragments to gfp and in vivo quantification of Gfp fluorescence. For the putative Fur box only a slight repressing impact on promoter activity could be shown. In a heteroallelic fur mutant a corresponding incomplete repression of isiAB transcription under iron-replete conditions confirmed the role of Fur in isiAB regulation. However, a 90 bp region upstream of the putative −35 box of the isiAB promoter was essential for full promoter activity under iron-deplete conditions. This pattern indicates a dual promoter regulation by both a repressing mechanism exhibited via the Fur system and an unknown activating mechanism.

    Notes: The expression of the chlorophyll a-binding, iron stress-induced protein IsiA is part of the cyanobacterial response to iron deficiency. A new isiA gene from the filamentous heterocystous cyanobacterial strain, Fischerella muscicola PCC 73103, was identified using standard and inverse PCR. While in unicellular cyanobacterial strains isiA is organized in an operon with isiB (encoding flavodoxin), in Fischerella not an isiB gene but another chlorophyll-binding protein encoding gene was identified downstream of isiA, which shows significant similarities to Pcb-like protein encoding genes known from prochlorophytes. The expression of both genes was clearly activated under iron deficiency. Although isiA and pcbC were independently transcribed, the size of the pcbC transcript indicates a large iron-regulated operon. Beside a 10-fold increase of isiA transcript content iron-starved cells of Fischerella showed a blue-shift in the red chlorophyll a absorption peak. In addition, chlorophyll fluorescence at 77 K was dominated by an emission peak at 685 nm. These features are in accordance with the characteristics of IsiA accumulation in iron-starved unicellular cyanobacteria, suggesting identical IsiA function in heterocystous strains in spite of different genetic organization.

    Notes: A mutant of Synechocystis sp. strain PCC6803 was obtained by random cartridge mutagenesis, which could not grow at low sodium concentrations. Genetic analyses revealed that partial deletion of the sll0273 gene, encoding a putative Na+/H+ exchanger, was responsible for this defect. Physiological characterization indicated that the sll0273 mutant exhibited an increased sensitivity towards K+, even at low concentrations, which was compensated for by enhanced concentrations of Na+. This enhanced Na+ demand could also be met by Li+. Furthermore, addition of monensin, an ionophore mediating electroneutral Na+/H+ exchange, supported growth of the mutant at unfavourable Na+/K+ ratios. Measurement of internal Na+ and K+ contents of wild-type and mutant cells revealed a decreased Na+/K+ ratio in mutant cells pre-incubated at a low external Na+/K+ ratio, while it remained at the level of the wild type after pre-incubation at a high external Na+/K+ ratio. We conclude that the Sll0273 protein is required for Na+ influx, especially at low external Na+ concentrations or low Na+/K+ ratios. This system may be part of a sodium cycle and may permit re-entry of Na+ into the cells, if nutrient/Na+ symporters are not functional or operating.
