    Notes: We present a quantum chemical investigation of the vibronic structure in the electronic spectra of three α,ω-dithienylpolyenes. Equilibrium structures and vibrational force fields of ground and excited electronic states are computed at semiempirical and ab initio levels of theory. The molecular parameters are employed to calculate displacement parameters for totally symmetric modes which are then used to model the vibronic structure of emission and absorption spectra. The observed spectral features are well reproduced by the computed spectra and two characteristic frequency regions are identified for the emission spectra. The first, above 1500 cm−1, is due to the C(Double Bond)C stretching activity typical of trans polyenes. In addition, the strong activity in emission around 1430 cm−1 is rationalized by considering that the two etherocyclic rings add, to the central polyenic chain, a conjugated carbon skeleton with cis linkages. In analogy with the oligomers of thiophene, the cis linkages, along with vibronic coupling, are responsible for the remarkable activity observed in emission in this frequency region. © 2001 American Institute of Physics.

    Notes: Gymnodinium catenatum is one of several dinoflagellates that produce a suite of neurotoxins called the paralytic shellfish toxins (PST), responsible for outbreaks of paralytic shellfish poisoning in temperate and tropical waters. Previous research suggested that the bacteria associated with the surface of the sexual resting stages (cyst) were important to the production of PST by G. catenatum. This study sought to characterise the cultivable bacterial diversity of seven different strains of G. catenatum that produce both high and abnormally low amounts of PST, with the long-term aim of understanding the role the bacterial flora has in bloom development and toxicity of this alga. Sixty-one bacterial isolates were cultured and phylogenetically identified as belonging to the Proteobacteria (70%), Bacteroidetes (26%) or Actinobacteria (3%). The Alphaproteobacteria were the most numerous both in terms of the number of isolates cultured (49%) and were also the most abundant type of bacteria in each G. catenatum culture. Two phenotypic (functional) traits inferred from the phylogenetic data were shown to be a common feature of the bacteria present in each G. catenatum culture: firstly, Alphaproteobacteria capable of aerobic anoxygenic photosynthesis, and secondly, Gammaproteobacteria capable of hydrocarbon utilisation and oligotrophic growth. In relation to reports of autonomous production of PST by dinoflagellate-associated bacteria, PST production by bacterial isolates was investigated, but none were shown to produce any PST-like toxins. Overall, this study has identified a number of emergent trends in the bacterial community of G. catenatum which are mirrored in the bacterial flora of other dinoflagellates, and that are likely to be of especial relevance to the population dynamics of natural and harmful algal blooms.

    Notes: It was observed previously that the deletion of the open reading frame YNL107w (YAF9) was highly pleiotropic in yeast and caused defective growth phenotypes in the presence of several unrelated inhibitors, including caesium chloride. We have selected multicopy extragenic suppressor genes, revealing that this phenotype can be suppressed by overdosing the transcription factors BDF1 and GAT1 in the yaf9Δ strain. We focused our analysis on suppression by BDF1 and performed a genome-wide transcript analysis on a yaf9Δ strain, compared with the wild-type and BDF1-suppressed strains. YAF9 deletion has a clear effect on transcription and leads to modulation of the level of expression of several genes. Transcription of a considerable portion of the underexpressed genes is restored to wild-type levels in the BDF1-suppressed strain. We show by chromatin immunoprecipitation that both Yaf9p and Bdf1p bind to promoters of some of these genes and that the level of H3 and H4 acetylation at one of these promoters is significantly lowered in the yaf9 deleted strain, compared with the wild-type and the BDF1-suppressed strains.

    Description / Table of Contents: Zusammenfassung Ein Metadolerit aus den Ophiolithen des Nördlichen Apennins (Vignola im Panaro-Tal bei Modena, Italien) entspricht in den Gehalten an Ti, P, Zr, Cr, Y und Nb den Tholeiiten der Ozeanböden. Er weicht hingegen von typischen ozeanischen Tholeiiten dadurch ab, daß er 1. primäre Alkaliamphibole enthält, daß 2. seine Klinopyroxene im Diagramm TiO2−MnO−Na2O nicht in die Felder der Tholeiite der Ozeanböden fallen und daß 3, der berechnete U-Gehalt für die mit seinen magmatischen Mineralen im Gleichgewicht stehenden liquiden Phase ähnlich jener ist, die für Alkalibasalte angegeben wird. Diese Ergebnisse schränken die generelle Bedeutung, die den Ophiolithen des Nördlichen Apennins bisher allgemein zugeschrieben wurde, ein: Nicht alle entstanden an sich ausdehnenden Rücken ähnlich jenen der heutigen Ozeanböden. Die metamorphen Parageneses zeigen, daß das Gestein bei niedrigem Druck, wahrscheinlich in ozeanischer Umgebung, Umwandlungen erlitt; dann folgten Rekristallisationen unter höherem Druck, die wohl zeitlich mit einer Orogenese zusammenfielen.
    Notes: Summary A metadolerite of the ophiolites of the Northern Apennines (Vignola, Panaro Valley, near Modena, Italy) behaves as ocean-floor tholeiites when Ti, P, Zr, Cr, Y and Nb are considered. However, it deviates from typical oceanic tholeiites in that(1) it contains primary alkali amphiboles, (2) its clinopyroxenes plot in the diagram TiO2−MnO−Na2O not in the fields of the ocean-floor basalts, and (3) the calculated uranium content of the liquid in equilibrium with its magmatic minerals is similar to the one reported for alkali basalts. These results put constraints on the generalized significance attributed to the ophiolites of the Northern Apennines, i. e. not all of them were generated at spreading ridges similar to the “normal ridge segments” of the present-day ocean-floors, as has been generally accepted by now. The metamorphic parageneses indicate that the rock underwent low-pressure transformations, probably in oceanic environment, followed by recrystallizations at higher pressure, reasonably in concomitance with an orogenesis.

    Notes: Abstract  Transgenic white poplar plants (Populus alba L.) expressing the nptII gene and the bar gene from Streptomyces hygroscopicus have been produced using Agrobacterium tumefaciens-mediated gene transfer. Eleven kanamycin-resistant plant lines were obtained with a transformation frequency of 7%. Successful genetic transformation was confirmed by Southern and northern analyses. The level of resistance to the commercial preparation of phosphinothricin (Basta; Roussel-Hoechst Agrovet) was evaluated by in vitro and in vivo assays. Using in vitro selective conditions for phosphinothricin, only plantlets from four kanamycin-resistant independent lines remained green and continued to grow and root. After transfer to the growth chamber, all selected transgenic lines were shown to be completely resistant to the herbicide Basta with doses equivalent to 6 l ha–1 (normal field dosage) and were tolerant at concentration of 12 l ha–1. This is the first report describing the genetic transformation of a P. alba clonal cultivar of commercial interest with a gene of agronomic value.

    Notes: Abstract A landrace (‘Moscatella’) and a commercial cultivar (‘Starkspur Red’) of Malus pumila Mill. were maintained and proliferated in vitro for ≥ four years. A factorial experiment, planned to evaluate the response of the two cultivars in conservation in determined slow growth conditions, was carried out for eighteen months. Single node cuttings were stored in 4 different media, at 4 °C, in dark conditions, in microvessels to assess the feasibility of reducing space in in vitro gemplasm banks. Culture viability after storage was evaluated after 6, 8,12 and 18 months of storage. Both varieties showed high survival percentages for up to a year of conservation, but the landrace's capability to resume growth dropped dramatically afterwards. On the contrary, ‘Starkspur Red’ maintained substantially unchanged capacity of resuming vigorous growth after 18 months of conservation. Overall, microvessels appeared to be suitable for storing single node cuttings under slow growth conditions up to a year at least. The described techniques could be useful for in vitro germplasm collections where frequent subculturing enhances the risk of genetic changes and personnel, energy and materials costs limit the amount of genotypes that can be managed.

    Notes: Abstract Thirteen landraces of cowpea (Vigna unguiculata (L.) Walp. subsp. unguiculata cv. gr. Unguiculata) have been morphologically and agronomically characterised as part of a plan for safeguarding and protecting plant biodiversity typical of Lake Trasimeno (Umbria, central Italy). Sixty plants per accession were scored for plant vigour, growth habit, flower colour, days to flowering, days to first mature pod, pod length, total number of pods produced, seed colour pattern, 100 seed weight, total seed yield and virus susceptibility. With the exception of vigour and growth habit, all landraces appeared to be well differentiated from one another for all the characters considered. Strategies and problems related to their in situ conservation are discussed.
