    Notizen: Weather damage reduces the value of commercial mungbean. but hard-seededness can reduce the level of damage. However. attempts lo breed large- and hard-seeded mungbean varieties have been unsuccessful. To understand the relationship between seed weight and hard-seededness. these trails were investigated using a quantitative trail loci (QTL) mapping approach with a recombinant inbred population derived from a cross between a completely soft-seeded variety and a completely hard-seeded genotype. The two parental genotypes also had a sixfold difference in seed weight. QTL analyses revealed four loci for hard-seededness and 11 loci for seed weight. Two of the hard-seeded ness loci co-localized with seed weight QTL. When seed weight was used as a covariate in the analysis of hard-seededness from the field data, two of the four hard-seeded QTL remained significant with the effect al one of these remaining unchanged. These results explain why retaining hard-seededness in large seeded mungbean lines has been unsuccessful. The existence of a persistent locus, however. indicated that breeding large and persistently hard-seeded varieties of mungbean may be possible.

    Notizen: The brown planthopper (BPH) is one of the most destructive insect pests of rice. Resistant varieties have proved to be one of the most economic and effective measures for BPH management. In this study, an indica rice ‘DV85’ showed resistance to biotype 2 of BPH by bulked seedling test, and a recombinant inbred line (RIL) population derived from a cross between a susceptible rice ‘Kinmaze’ and ‘DV85’ was phenotyped to map genetic factors conferring BPH resistance in ‘DV85′. Composite interval mapping revealed that one quantitative trait locus (QTL) with a LOD score of 10.1 was detected between XNpb202 and C1172 on chromosome 11. This QTL was designated as Qbph11. Qbph11 explained 68.4% of the phenotypic variance of BPH resistance in this population. The allele from the resistant parent ‘DV85’ at Qbph11 reduced the damage caused by BPH feeding and would be very useful in breeding resistant rice varieties via marker-assisted selection.

    Notizen: A precision scale landscape model designed for agricultural applications is described in this paper. The Precision Agricultural Landscape Modeling System (PALMS) is a combination of two process-based models: a diffusive wave runoff model with ponding (described in detail) and a biosphere model with a crops module (briefly reviewed). Several innovations, including numerical formulations for the hydrologic properties of the soil surface with crusting, slope/tillage angle interactions, and change of roughness and detention storage with cumulative precipitation have been included. The model is compared to observations on one 1.8 ha field planted with maize and soybeans during four growing seasons, and one 24 ha field planted with maize during one growing season. Daily average soil moisture is simulated well (within 5 percent volumetric), except in extended runoff/ponding episodes. Physical processes not simulated in the model suggest possible explanations for model errors. Planned improvements for PALMS are also presented.

    Notizen: Erianthus arundinaceus has a number of important agronomic traits including good ratooning ability, tolerance to both drought and water logging, disease resistance and vigour and is of interest as a potential source of parental germplasm to sugarcane breeders. However, to date, attempts to produce fertile hybrids between sugarcane (Saccharum spp.) and E. arundinaceus have been unsuccessful. Microsatellite markers that generated genus-specific markers and were highly polymorphic within sugarcane were identified. The microsatellite markers and 5S rDNA PCR were used to screen intergeneric (F1) clones from Saccharum officinarum×E. arundinaceus crosses, and two Saccharum backcross progeny (BC1) populations derived from crosses between selected F1 clones and sugarcane (Saccharum spp.), to identify genuine Saccharum spp. BC1 progeny. The 5S rDNA PCR marker and highly polymorphic microsatellites with Erianthus-specific bands confirmed the F1 parent of the two putative BC1 populations was a S. officinarum×E. arundinaceus hybrid and allowed the identification of the genuine BC1 progeny from selfs of the F1 parent. This is the first verification of BC1 progeny from an F1 intergeneric hybrid x sugarcane (Saccharum spp.) clone with molecular markers and confirms the introgression of E. arundinaceus germplasm into sugarcane. It should now be possible to exploit genes of value from E. arundinaceus in sugarcane breeding programmes.

    Notizen: Yield and its components were investigated by using a population of 241 recombinant inbred lines (F9 RILs) derived from an elite hybrid rice cross of ‘Zhenshan 97’בMinghui 63′. Quantitative trait loci (QTLs) for causal analysing of yield traits were detected at different yield component (YC) influences by conditional and unconditional QTL mapping methods. The number of QTLs significantly affecting yield was different at component-special influence. Some QTLs controlling yield identified in one component influence were undetectable at the others. More QTLs for yield could be detected at different YC influences. It is possible to reveal that causal gene expression for yield could be different at different YC influences. Mapping QTLs for component effects of yield could help us in understanding the nature of cause-effect traits for the formation of grain yield.

    Notizen: Among the most important crops in developing countries are banana and plantain. However, the production is threatened by increasingly virulent forms of Fusarium wilt, and therefore, intensive breeding programmes are being carried out worldwide. As conventional field studies of banana resistance to this disease are time-consuming and destructive, an easy-to-do procedure was previously developed to differentiate field-grown resistant and susceptible banana cultivars at leaf level. Such a procedure involved the in vitro treatment of fungal culture filtrates on to field-grown adult leaves and the measurement of lesion areas 48 h later. The present report includes measurements of other indicators such as biochemical compounds. The cultivar ‘Gross Michel’ (susceptible) and cv. ‘FHIA-01’ (resistant) leaves were treated with Fusarium oxysporum f. sp. cubense race 1 culture filtrates. Evaluations were performed 48 h after leaf treatment. Compared with culture medium-treated leaves (control treatment), fungal metabolites produced leaf lesions, decreased freephenolic contents and increased protein levels in both cultivars. In ‘FHIA-01’, the culture filtrate increased contents of cell wall-linked phenolics and the pool of aldehydes (except malondialdehyde). Fungal metabolites did not cause variations in peroxidase activity, chlorophyll pigment contents or malondialdehyde level in any cultivar. The use of Fisher's linear discriminant analysis to differentiate resistant and susceptible banana cultivars in breeding programmes is also a novel aspect of this report. Such an estimation was performed from a data matrix that included the effects of the fungal metabolites (leaf lesion area and levels of free and cell wall-linked phenolics, aldehydes, except malondialdehyde, and proteins) on banana leaves of seven cultivars (four susceptible and three resistant).

    Notizen: Eight dolerite outcrops in Preseli, south Wales were measured in situ using field-portable XRF analysis, in order to compare two different analysis (“sampling”) strategies, and to investigate geochemical variability within and between outcrops. A sampling strategy of two (neighbouring but independent) measurements at each of a maximal number of locations dispersed over an outcrop was the more effective in indicating the overall chemical variance of that outcrop. Analysis of variance indicated that much of the observed variance within individual outcrops stems from real geochemical variability rather than from sampling and analytical factors. Standard ANOVA F tests showed that several of the studied outcrops are heterogeneous at the 5% significance level for one or more elements. Geochemical distinctions between some outcrops were demonstrated using discriminant analysis. PXRF analysis offers an alternative approach to conventional characterisation of outcrops, which is often based on laboratory analysis of small numbers of samples. However, PXRF data are affected by rock weathering and may require correction for this if they are to be compared with analyses of fresh rock.

    Notizen: : Gelation capacity of a native amaranth protein isolate was studied. Structural properties of gels prepared at different protein concentration and heating conditions were analyzed. Proteins present in amaranth isolates obtained by water extraction at pH 9.0 and subsequent isoelectric precipitation are able to form gels of yellowish appearance. Gel color intensity increased while luminosity decreased with increasing protein concentrations. High protein concentration allowed the formation of matrices with high water-holding capacity. In addition, increasing the heating temperature resulted in gels of high luminosity and low water-holding capacity. The increase of protein concentration (10% to 20% w/v) as well as the increase of heating temperature (70°C to 95°C) and heating time (10 to 30 min) resulted in the formation of a more ordered matrix with smaller pores, mainly stabilized by disulfide bonds and, at a lower extent, by noncovalent interactions (specially hydrogen bonds and hydrophobic interactions). Both amaranth globulin (11S globulin and P globulin) participated in gel structure via high-molecular-weight aggregates (〉100 kD). Gel structure was stabilized via noncovalent bonds by monomer species of 42 kD and those of molecular mass lower than 20 kD localized in the interstitial spaces of gel matrix.

    Notizen: Atypical psychrophilic Aeromonas salmonicida isolates were obtained from farmed and wild fish in Northeastern North America. These bacteria were isolated between 1992 and 2001 and carried tetracycline resistance (Tcr) plasmids of approximately 58 kb. The nine isolates had plasmids which could be divided into four groups based on the specific tetracycline resistance (tet) gene carried [tet(A) or tet(B)], incompatibility of the plasmid [IncU or other], whether the plasmid carried the IS6100 sequences, the sul1 gene, coding for sulfonamide resistance, the dfrA16 gene, coding for trimethoprim resistance, and/or carried a complete Tn1721, and their ability to transfer their Tcr plasmids to an Escherichia coli recipient at 15 °C. Five of the isolates, with genetically related Tcr plasmids, were able to transfer their plasmids to an E. coli recipient at frequencies ranging from 5.7 × 10−4 to 2.8 × 10−6 per recipient. The 1992 isolate carried a genetically distinct plasmid, which transferred at a slightly higher rate. The three remaining isolates carried one of two genetically different plasmids, which were unable to transfer to an E. coli recipient. Conjugal transfer at 15 °C is the lowest temperature that has been documented in bacteria.

    Notizen: Sodalis glossinidius is known exclusively in endosymbiosis with tsetse flies (Genus: Glossina) and is one of the few insect bacterial symbionts that have been successfully cultured in vitro. This study details improved isolation and solid culture protocols that allow for a standardised and rapid preparation/maintenance of clonal material from individual flies. The isolation and culture of S. glossinidius was confirmed by partial sequencing of the 16S rDNA gene and specific PCR. In addition, the growth dynamics and changes in cell viability during liquid culture are described. The potential for culture of other endosymbiont taxa is discussed.
