    Description: There is an increasing demand to efficiently process emerging types of queries, such as progressive queries (PQs), from contemporary database applications including telematics, e-commerce and social media. Unlike conventional queries, a PQ consists of a set of step-queries (SQ). A user formulates a new SQ on the fly based on the result(s) from the previous SQ(s). Existing database management systems were not designed to efficiently process such queries. In this paper, we present a novel technique to efficiently process a special type of PQ, called monotonic linear PQs, based on dynamically materialized views. The key idea is to create a superior relationship graph for SQs from historical PQs that can be used to estimate the benefit of keeping the current SQ result as a materialized view. The materialized views are used to improve the performance of future SQs. A new storage structure for the materialized views set is designed to facilitate efficient search for a usable view to answer a given SQ. Algorithms/strategies to efficiently construct a superior relationship graph, dynamically select materialized views, effectively manage the materialized views set and efficiently search for usable views are discussed. Experiment results demonstrate that our proposed technique is quite promising.

    Description: Anthropogenic nitrogen (N) emissions to atmosphere have increased dramatically in China since 1980s, and this increase has aroused great concerns on its ecological impacts on terrestrial ecosystems. Previous studies have showed that terrestrial ecosystems in China are acting as a large carbon (C) sink, but its potential in the future remains largely uncertain. So far little work on the impacts of the N deposition on C sequestration in China’s terrestrial ecosystems has been assessed at a national scale. Aiming to assess and predict how ecological processes especially the C cycling respond to the increasing N deposition in China’s forests, recently researchers from Peking University and their partners have established a manipulation experimental network on the ecological effects of the N deposition: Nutrient Enrichment Experiments in China’s Forests Project (NEECF). The NEECF comprises 10 experiments at 7 sites located from north to south China, covering major zonal forest vegetation in eastern China from boreal forest in Greater Khingan Mountains to tropical forests in Hainan Island. This paper introduces the framework of the NEECF project and its potential policy implications.

    Description: IgG was traditionally thought to neutralize virions by blocking their attachment to or penetration into mucosal epithelial cells, a common site of exposure to viruses. However, we describe an intracellular neutralizing action for an influenza hemagglutinin-specific monoclonal antibody, Y8-10C2 (Y8), which has neutralizing activity only at an acidic pH. When Y8 was applied to the basolateral surface of Madin–Darby canine kidney cells expressing the rat neonatal Fc receptor for IgG (FcRn), it significantly reduced viral replication following apical exposure of the cell monolayer to influenza virus. Virus neutralization by Y8 mAb was dependent on FcRn expression and its transport of IgG. As both FcRn and Y8 mAb bind their partners only at acidic pH, the Y8 mAb is proposed to carry out its antiviral activity intracellularly. Furthermore, the virus, Y8 mAb, and FcRn colocalized within endosomes, possibly inhibiting the fusion of viral envelopes with endosomal membranes during primary uncoating, and preventing the accumulation of the neutralized viral nucleoprotein antigen in the nucleus. Prophylactic administration of Y8 mAb before viral challenge in WT mice, but not FcRn-KO mice, conferred protection from lethality, prevented weight loss, resulted in a significant reduction in pulmonary virus titers, and largely reduced virus-induced lung pathology. Thus, this study reveals an intracellular mechanism for viral neutralization in polarized epithelial cells that is dependent on FcRn-mediated transport of neutralizing IgG.

    Description: Initiation of DNA replication in eukaryotes is exquisitely regulated to ensure that DNA replication occurs exactly once in each cell division. A conserved and essential step for the initiation of eukaryotic DNA replication is the loading of the mini-chromosome maintenance 2–7 (MCM2–7) helicase onto chromatin at replication origins by Cdt1. To elucidate the molecular mechanism of this event, we determined the structure of the human Cdt1-Mcm6 binding domains, the Cdt1(410–440)/MCM6(708–821) complex by NMR. Our structural and site-directed mutagenesis studies showed that charge complementarity is a key determinant for the specific interaction between Cdt1 and Mcm2–7. When this interaction was interrupted by alanine substitutions of the conserved interacting residues, the corresponding yeast Cdt1 and Mcm6 mutants were defective in DNA replication and the chromatin loading of Mcm2, resulting in cell death. Having shown that Cdt1 and Mcm6 interact through their C-termini, and knowing that Cdt1 is tethered to Orc6 during the loading of MCM2–7, our results suggest that the MCM2–7 hexamer is loaded with its C terminal end facing the ORC complex. These results provide a structural basis for the Cdt1-mediated MCM2–7 chromatin loading.

    Description: Liquid-phase hydrogenation using a Pd/Al 2 O 3 catalyst provides a potential technique for the reduction of cumene hydroperoxide (CHP) to α-cumyl alcohol (CA). In this paper, CHP hydrogenation was carried out in a cocurrent downflow trickle-bed reactor over a wide range of reaction conditions to study the reaction and deactivation kinetics. The proposed intrinsic rate expression for CHP hydrogenation is based on an Eley-Rideal mechanism that accounts for an irreversible surface reaction between the absorbed CHP with nonabsorbed hydrogen molecules. During CHP hydrogenation, an exponential decay in activity of the Pd/Al 2 O 3 catalyst and the presence of residual activity were observed. A kinetic deactivation model with residual activity was developed. Based on reaction and deactivation kinetics, catalyst deactivation was attributed to oxidation of the catalyst surface by CHP. The presence of residual activity was due to the partial reduction of oxidized catalyst surface by hydrogen. A comprehensive investigation of the catalytic hydrogenation of cumene hydroperoxide in cumene over a Pd/Al 2 O 3 catalyst is presented, including the effects of residence time, reaction temperature and H 2 pressure on conversion and catalyst deactivation. Attempts are made to simulate the performance of operating conditions with kinetic models.

    Description: IspG is a 4Fe4S protein involved in isoprenoid biosynthesis. Most bacterial IspGs contain two domains: a TIM barrel (A) and a 4Fe4S domain (B), but in plants and malaria parasites, there is a large insert domain (A*) whose structure and function are unknown. We show that bacterial IspGs function in solution as (AB)2 dimers and that mutations in either both A or both B domains block activity. Chimeras harboring an A-mutation in one chain and a B-mutation in the other have 50% of the activity seen in wild-type protein, because there is still one catalytically active AB domain. However, a plant IspG functions as an AA*B monomer. We propose, using computational modeling and electron microscopy, that the A* insert domain has a TIM barrel structure that interacts with the A domain. This structural arrangement enables the A and B domains to interact in a “cup and ball” manner during catalysis, just as in the bacterial systems. EPR/HYSCORE spectra of reaction intermediate, product, and inhibitor ligands bound to both two and three domain proteins are identical, indicating the same local electronic structure, and computational docking indicates these ligands bridge both A and B domains. Overall, the results are of broad general interest because they indicate the insert domain in three-domain IspGs is a second TIM barrel that plays a structural role and that the pattern of inhibition of both two and three domain proteins are the same, results that can be expected to be of use in drug design.
