    Description: From November 2004 to December 2007, size-segregated aerosol samples were collected all-year-round at Dome C (East Antarctica) by using PM10 and PM2.5 samplers, and multi-stage impactors. The data set obtained from the chemical analysis provided the longest and the most time-resolved record of sea spray aerosol (sea salt Na+) in inner Antarctica. Sea spray showed a sharp seasonal pattern. The highest values measured in winter (Apr-Nov) were about ten times larger than in summer (Dec-Mar). For the first time, a size-distribution seasonal pattern was also shown: in winter, sea spray particles are mainly submicrometric, while their summer size-mode is around 1-2 µm. Meteorological analysis on a synoptic scale allowed the definition of atmospheric conditions leading sea spray to Dome C. An extreme-value approach along with specific environmental based criteria was taken to yield stronger fingerprints linking atmospheric circulation (means and anomalies) to extreme sea spray events. Air mass back-trajectory analyses for some high sea spray events allowed the identification of two major air mass pathways, reflecting different size distributions: micrometric fractions for transport from the closer Indian-Pacific sector, and sub-micrometric particles for longer trajectories over the Antarctic Plateau. The seasonal pattern of the SO4**2- /Na+ ratio enabled the identification of few events depleted in sulphate, with respect to the seawater composition. By using methanesulphonic acid (MSA) profile to evaluate the biogenic SO4**2- contribution, a more reliable sea salt sulphate was calculated. In this way, few events (mainly in April and in September) were identified originating probably from the "frost flower" source. A comparison with daily-collected superficial snow samples revealed that there is a temporal shift between aerosol and snow sea spray trends. This feature could imply a more complex deposition processes of sea spray, involving significant contribution of wet and diamond dust deposition, but further work has to be carried out to rule out the effect of wind re-distribution and to have more statistic significance.

    Notes: Examination of 55 clinical isolates of uropathogenic Escherichia coli producing the CNF1 toxin demonstrated that the cnf1 gene is systematically associated with a hly operon via a highly conserved hlyD-cnf1 intergenic region (igs, 943 bp) as shown in the J96 UPEC strain. We examined if this association could reflect a co-regulation of the production of these toxins. Translation of cnf1 from an immediately upstream promoter has been shown to be controlled by means of an anti-Shine–Dalgarno sequence present in the cnf1 coding sequence [fold-back inhibition (cnf1 fbi)]. The cnf1 fbi was not regulated by elements present in the igs. An RNA covering the full hlyD sequence, the igs and extending on the cnf1 gene, was then detected in the J96 strain. This RNA could be part of a HlyCABD mRNA. Transcription of the haemolysin operon requires RfaH antitermination activity. Inactivation of rfaH in J96 resulted in a 100-fold reduction of the CNF1 content of bacteria. The production of CNF1 from a plasmidic igscnf1 DNA was not sensitive to RfaH, indicating that this factor acted on cnf1 transcription via the hly promoter. This way the cnf1 fbi mechanism might be overcome by transcription of cnf1 from the haemolysin promoter and antitermination by RfaH. This constitutes a novel system of bacterial virulence factors co-regulation.

    Notes: Iota-toxin from Clostridium perfringens type E is a binary toxin consisting of two independent proteins, an enzymatic Ia and binding Ib component. Ia catalyses ADP-ribosylation of actin monomers, thus disrupting the actin cytoskeleton. In this report, we show that Ia plus Ib applied apically or basolaterally induce a rapid decrease in the transepithelial resistance (TER) of CaCo-2 cell monolayers and disorganization of actin filaments as well as the tight and adherens junctions. Ib alone, on the apical or basolateral side, slowly decreased the TER without affecting the actin cytoskeleton, possibly via pore formation. Interestingly, the two iota-toxin components inoculated separately on each cell surface induced cytopathic effects and a TER decrease. Anti-Ib sera, raised against the whole molecule or the Ia docking domain and applied to the opposite cell side versus Ib, neutralized the TER decrease. In addition, radioactive Ib incubated in the basolateral compartment was detected on the apical side by selective cell surface biotinylation. This argues for a transcytotic routing of Ib to mediate internalization of Ia from the opposite cell surface. Bafilomycin A1 also prevented the cytopathic effects of Ia and Ib applied separately to each cell side, possibly by blocking translocation of Ia into the cytosol and/or the intracellular transport of Ib. Ib is either routed into the cell independently of Ia, trans-cytosed and permanently exposed on the opposite cell surface or continuously recycled between an endosomal compartment and the cell surface.

    Notes: Abstract A series of pseudo amorphous cellulose samples were reacted with catalytic amounts of 2,2,6,6-tetramethyl-1-piperidine oxoammonium salt (TEMPO), sodium hypochlorite and sodium bromide in water. In all samples the primary alcohol groups were selectively oxidised into carboxyl groups, and several water-soluble polyglucuronic acid sodium salts were obtained with different molecular weights. With this reaction system, the degradation of the amorphous cellulose samples may be minimized, provided the oxidation is performed at 4 °C and at constant pH 10, with controlled amounts of TEMPO and sodium hypochlorite.

Experts and consultants in water management usually work with a limited set of analysis and project planning methods. This implies that expertsand consultants have a restricted spectrum for bidding on watermanagement projects and that the engagement of a specific expertor consultant introduces a biased approach to water management.To alleviate these limitations, experts and consultants shouldhave a readily accessible collection of analysis and project management methods at their disposal, coupled with guidelinesfor combining these methods to best approach a water managementproject. In this paper we propose a taxonomy for classifying methods used in water management. Using this taxonomy, we derivea Project Management Specification Model (PMSM) to help expertsselect, combine, and operationalize methods to match best therequirements posed by a project. The taxonomy and PMSM were derived and validated by analyzing more than 90 methods togetherwith experts in the field of water management, and by using themwithin two new project proposals made by an engineering company.We could conclude that PMSM's broad view on water managementhelps experts and consultants choose flexible analysis and project management approaches, and that it increases the involvement of, and acceptance by, all parties involved in water management.

    Notes: Abstract The long-lived radionuclide 129I (T 1/2 = 15.7 My) occurs in the nature in very low concentrations. Since the middle of our century the environmental levels of 129I have been dramatically changed as a consequence of civil and military use of nuclear fission. Its investigation in environmental materials is of interest for environmental surveillance, retrospective dosimetry and for the use as a natural and man-made fracers of environmental processes. We are comparing two analytical methods which presently are capable of determining 129I in environmental materials, namely radiochemical neutron activation analysis (RNAA) and accelerator mass spectrometry (AMS). Emphasis is laid upon the quality control and detection capabilities for the analysis of 129I in environmental materials. Some applications are discussed.

    Notes: Abstract Based on Bayesian statistics, characteristic limits such as decision threshold, detection limit and confidence limits can be calculated taking into account all sources of experimental uncertainties. This approach separates the complete evaluation of a measurement according to the ISO Guide to the Expression of Uncertainty in Measurement from the determination of the characteristic limits. Using the principle of maximum entropy the characteristic limits are determined from the complete standard uncertainty of the measurand.
